The complete plastid genome of Delonix regia (Hook.) Raf. (Leguminosae).

ABSTRACT: Delonix regia, a plant species of the legume family native to Madagascar, has been widely cultivated in the tropical and subtropical regions as an ornamental tree due to its remarkable showy orange-red flowers over summer. Here we report for the first time the complete plastid genome of this species, which has a typical circular structure with a total length of 162,756 bp and contains two inverted repeat regions (IRs, 25,544 bp), a large single copy region (LSC, 92,490 bp), and a small single copy region (SSC, 19,178 bp). The phylogenetic analysis based on the complete plastome sequences of this species and those of the related species from GenBank strongly suggested that D. regia is nested in the subfamily Caesalpinioideae and is sister to a clade consisting of Erythrophlium fordii and the old-sense Mimosoideae.

Endophytism of Fusarium Solani in Meristems of Delonix Regia (Bojer ex Hook.) Raf.

Project description:A bright yellow pigment-producing endophytic fungus strain GMRS1 was isolated from the meristems of Delonix regia (Bojer ex Hook.) Raf. The fungus was identified as Fusarium solani on grounds of morphological and molecular analysis. The chemical composition of the crude pigments was partially analyzed by Quadrupole Time-of-flight Liquid chromatography/Mass spectrometry/Mass spectrometry and the major compounds were reported. The crude pigments were proved toxic and unsafe by acute oral toxicity study on Wistar rats. The total cholesterol and glucose levels of the rats ingested with crude pigments were significantly elevated than the control rats. The treatment rats were further observed with damaged liver and kidney tissues. A comprehensive multi-directional exploration of phytochemicals and bioactivities of flower extracts from Delonix regia (Bojer ex Hook.) Raf., Cassia fistula L. and Lagerstroemia speciosa L.

Project description:Delonix regia (Bojer ex Hook.) Raf., Cassia fistula L. and Lagerstroemia speciosa L. are three ornamental plants that produce colorful flowers. The present study aimed to evaluate the phytochemicals and bioactivities of methanolic extracts of flowers from Delonix regia (DrFME), Cassia fistula (CfFME), and Lagerstroemia speciosa (LsFME). The presence of ten different chemical classes in varying degrees was confirmed while qualitatively screened. During quantitative determination, LsFME possesses the highest amount of total phenolic (418.0 mg/g), flavonoid (50.8 mg/g), and tannin (256.3 mg/g) contents. The extracts showed excellent antioxidant capacity in a concentration-dependent manner with the lowest IC50 value (41.51 ?g/mL) displayed by LsFME. LsFME paralyzed the experimental worms at 2.95 min and killed at 3.96 min. DrFME was found to be more effective in thrombolytic (35.5% clot lysis) and anticoagulant activities. Negligible hemolytic activity (IC50 > 200 ?g/mL) found for all extracts which suggest their less potential toxicity. The in vivo experiments revealed that the CfFME has the highest analgesic (64.34% pain inhibition) activity while LsFME has the highest antidiarrheal (70.27% inhibition) and antihyperglycemic (46.94% inhibition) activities at 400 mg/kg of body weight doses. This study has shown the presence of phytochemicals and potential bioactivities which indicates the possibility of these flowers to be used as a source of phytochemicals as well as safe and effective natural medicine.

The complete plastid genome of Zenia insignis Chun (Leguminosae).

Project description:Zenia insignis Chun is a rare and threatened species of the monotypic genus Zenia Chun in the subfamily Dialioideae of Leguminosae. The complete plastid genome of Zenia insignis was sequenced for the first time which also represents the first complete plastid genome of the subfamily Dialioideae. The total length of this genome is 159,358 bp with a large single copy (LSC) region (88,507 bp), a small single copy (SSC) region (18,689 bp), and two inverted repeat regions (IRs, 26,081 bp). We recovered 128 functional genes, including 83 protein-coding genes, 37 tRNA genes and 8 rRNA genes. Phylogenetic analysis strongly supported a sister relationship between Z. insignis and the clade consisting of Caesalpinioideae and Papilionoideae.

The complete plastid genome sequence of Phyllodium pulchellum (L.) Desv. (Leguminosae).

Project description:This study investigated the complete plastid genome of Phyllodium pulchellum, which represents the first report of the complete plastome for the genus Phyllodium in the tribe Desmodieae of the subfamily Papilionoideae. De novo assembly and annotation showed that the plastid genome is a typical quadripartite structure consisting of two inverted repeats (IR = 24,286 bp), one small single copy (SSC = 18,334 bp), and one large single copy (LSC = 82,715 bp). We found 111 unique genes, including 77 protein-coding genes, 30 tRNA genes, and 4 rRNA genes. Phylogenetic analysis based on the newly sequenced plastid genome of Ph. pulchellum and 11 plastomes obtained from GenBank recovered a strongly supported phylogenetic backbone of the tribe Desmodieae and a sister-relationship between Phyllodium and Ohwia.

Phylogenetic analysis and morphology of Ganoderma multipileum, a Ganoderma species associated with dieback of the metropolitan woody plant Delonix regia (Boj. ex Hook.) Raf. in Vietnam.

Project description:Ganoderma multipileum, a wood decay mushroom, was initially discovered and classified in Taiwan through the analysis of its morphology and the internal transcribed spacer (ITS) region of nuclear ribosomal DNA. In this study, we identified a mushroom associated with the dieback of Delonix regia (Boj. ex Hook.) Raf., a woody ornamental street tree in Vietnam, as Ganoderma multipileum. This classification was based on phylogenetic analysis of ITS, RPB2, and TEF1 sequences, as well as morphology assessment and scanning electron microscope observation of basidiospores. The phylogenetic analysis revealed that the specimens collected in Vietnam formed a monophyletic group of Ganoderma multipileum with a high bootstrap value and posterior probability (100%/1.00). Furthermore, the morphological features consistent with laccate Ganoderma, including a thin pileipellis composed of enlarged and bulbous hyphae, and the basidiomes exhibited two different phenotypes. Notably, scanning electron microscopy of the basidiospores revealed ovoid spores with numerous echinules, providing the first documented evidence of this characteristic for Ganoderma multipileum. This research represents the first recorded instance of Ganoderma multipileum in Vietnam associated with the dieback of Delonix regia.

Project description:Bauhinia × blakeana Dunn, or the Hong Kong Orchid Tree, is a popular ornamental plant in the tropical and subtropical regions. In this study, we report and characterise the complete plastid genome of B. × blakeana in an effort to provide genomic resources for genetic utilization. The complete plastome is 156,100 bp in length and contains the typical quadripartite structure, including two inverted repeat (IR) regions of 25,847 bp, a large single-copy (LSC) region of 86,484 bp and a small single-copy (SSC) region of 17,922 bp. 126 genes are annotated, including 81 protein-coding genes, 37 transfer RNA genes, and eight ribosomal RNA genes. The phylogenetic analysis based on the plastomes from B. × blakeana and 11 previously reported species of Cercidoideae suggested a sister-relationship between B. × blakeana and B. acuminata L. with strong bootstrap support.

Delonix regia Leaf Extract (DRLE): A Potential Therapeutic Agent for Cardioprotection.

Project description:Delonix regia (Boj. Ex. Hook) is a flowering plant in the pea family found in tropical areas and its leaves are used informally to treat diseases in folk medicine. However, the cardioprotective effects in this plant are still unclear. In this study, we found that the Delonix regia leaf extract (DRLE) (400 mg/kg/d) can reduce the mortality rate in an isoproterenol (ISO)-induced heart injury and hypertrophy mouse model. Decreased serum levels of creatine phosphokinase, LDH, GOT, TNF-alpha and increased nitric oxide levels were found in DRLE-treated ISO-injured mice. In the in vitro study, the porcine coronary artery exhibited vasodilation effect induced by DRLE in a dose-dependent manner. In the DRLE toxic test, overdose of DRLE showed the high safety in normal mice and may have the ability to remove the metabolic wastes in blood. In conclusion, we demonstrated for the first time that DRLE has the cardioprotective effects by activating the vasodilation through NO pathway and preventing the myocyte injury via inhibition of TNF-alpha pathway. We suggest that DRLE may act as a promising novel herbal medicine for cardioprotection.

Project description:We sequenced and assembled the whole chloroplast genome of the Australian-endemic shrub Platylobium obtusangulum. The total size of the genome is 150,090 base pairs (bp), including two inverted repeat regions of 25,511 bp each, one large single copy region of 80,567 bp and a small single copy region of 18,501 bp. The genome has a GC content of 36.7% and includes 127 annotated genes (83 protein coding, 36 tRNA genes and eight rRNA genes). Phylogenetic analysis of chloroplast genomes placed the Platylobium obtusangulum genome in the expected position of the Mirbelioid clade in the legume family (Leguminosae: Papilionoideae).
