The first complete mitochondrial genome of genus Phytomia (Diptera: Syrphidae).

ABSTRACT: The complete mitochondrial genome (mitogenome) of Phytomia zonata was sequenced in this study, it is the first complete mitogenome in the genus Phytomia. The wholly sequence is a circular DNA, and with 15,716 bp in length including 37 typical genes. No gene arrangement was observed in this sequence. The common start and stop codons were ATN and TAA, respectively. A phylogenetic tree was generated based on dataset of 19 complete mitogenomes from hoverflies and two Lauxaniidae species which as outgoup, and shows Phytomia is closer to Eristalis, supports the monophyly of Syrphinae.

Project description:The complete mitochondrial genome of Syrphus ribesii was determined in this study. The double-stranded circular DNA molecule was 16,530 bp in length, containing 37 typical genes: 13 protein-coding genes (PCGs), 2 rRNA genes, 22 tRNA genes, and an A + T-rich region. Thirteen PCGs were 11,196 bp in size, encoding 3720 amino acids in total. All the PCGs started with ATN, except the COI used TTG as its initiation codon. Most PCGs terminated with standard codon TAA, while the COI ended with T and the ND5 ended with TA. The lrRNA and srRNA genes were 1341 bp and 793 bp in length, respectively. The A + T-rich region harbored some typical structures characteristic of the dipterans. The phylogenetic tree showed that Syrphus ribesii was closely related to Eupeodes corollae, and the Syrphidae and Pipunculidae constituted a monophyletic group within the Syrphoidea.

The complete mitochondrial genome of Helophilus virgatus (Diptera: Syrphidae: Eristalinae) with a phylogenetic analysis of Syrphidae.

Project description:The complete mitochondrial genome (mitogenome) of Helophilus virgatus (Coquilletti, 1898) was sequenced. Its whole mitogenome was 15,742 bp in length, and all 37 genes were in the ancestral gene arrangement. ATN was used as start codon in most of PCGs except for ND1 and COX1, which used TTG. Besides, ATP8, ND3, ND5, and ND6 ended with an incomplete T, the others used TAA as termination codons. A phylogenetic tree based on 13 PCGs from 14 species (11 Syrphidae and 3 outgroup species) shows that Syrphinae and Eristalinae form a sister group, and supports the monophyly of Syrphidae.

The complete mitochondrial genome of Platycheirus albimanus (Diptera: Syrphidae: Syrphinae) and phylogenetic analysis of the Syrphidae.

Project description:The complete mitochondrial genome of Platycheirus albimanus (Fabricius, 1781) was sequenced. The length of the mitogenome is 16,648 bp and consists of 37 genes including 22 transfer RNA (tRNAs), 13 protein-coding (PCGs) and 2 ribosomal RNA (rRNAs). The 13 PCGs initiate with the start codon ATN, except for COX1 and ATP6 which use TTG. All of the PCGs ended with TAA, apart from ND4 and ND4L terminated by incomplete T--. The ML tree based on complete mitogenomes from 25 species (22 Syrphidae and 3 outgroup taxa) suggests that the tribe Melanostomini is more closely related to the Syrphini. The phylogenetic analysis supports the monophyly of Syrphinae, and the paraphyly of the Eristalinae. This mitogenome information for P. albimanus could facilitate future studies of evolutionarily related insects.

The complete mitochondrial genome of Melanostoma mellinum (Linnaeus, 1758) (Diptera: Syrphidae) and phylogenetic analysis.

Project description:In this study, the complete mitochondrial genome (mitogenome) of Melanostoma mellinum (Linnaeus, 1758) was sequenced using the-next generation sequencing technology. The assembled mitogenome of M. mellinum has a total length of 16,055bp and contains 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), and 2 ribosomal RNA genes (rRNAs). The results of phylogenetic reconstruction based on the combined mitochondrial gene dataset indicated that M. mellinum belongs to Melanostoma genus with a close relationship to Melanostoma orientale, but the monophyly of the tribe Bacchini is not well supported.

The complete mitochondrial genome of Syritta pipiens (Linnaeus, 1758) (Diptera: Syrphidae) and phylogenetic analysis.

Project description:The complete mitochondrial genome (mitogenome) of Syritta pipiens (Linnaeus, 1758) was sequenced with 15,745 bp in length including 37 genes and a non-coding region. The overall nucleotide composition showed a strong AT bias. Most protein-coding genes (PCGs) used ATN as the start codon while ATP6 and ND1 used TTG, and stopped by TAA or TAG but ND5 ended with an incomplete T. Phylogenetic trees were reconstructed based on the 24 complete mitochondrial sequences from Syrphidae using the methods of maximum-likelihood (ML) and Bayesian inference (BI), resulted in S. pipiens clustered into the clade of Eristalinae, which conformed to the traditional classification, but the trees did not support the monophyly of Eristalinae. More molecular data is needed for further study.

Characterization and phylogenetic analysis of the complete mitochondrial genome of Eristalia cerealis (Diptera: Syrphidae).

Project description:The complete mitochondrial genome of Eristalia cerealis was sequenced and reported here. The circle genome of the syrphid fly is 15,348 bp in length. There are 38 sequence elements including 13 protein coding genes, 22 tRNA genes, 2 rRNA genes, and a control region. The order of all elements was the same with that of E. tenax. With 2 species from Muscidae and Drosophilidae as outgroups, phylogenetic relationships of 10 Syrphidae species based on mitogenomes were in complete agreement with their taxonomic relationships based on morphological characteristics. Our result will provide more fundamental data to the development of the molecular systematics of Syrphidae.

First complete mitochondrial genome of Pselliophora (Diptera, Tipulidae): genome description and phylogenetic implications.

Project description:Pselliophora is widely distributed in Eurasia and China. To explore the characteristics of the mitogenome of Pselliophora and reveal phylogenetic relationships, the mitogenome of Pselliophora bifascipennis Brunetti, 1911 was sequenced and annotated. This is the first complete mitochondrial genome in this genus. Its mitogenome is 15821 bp in length, containing 13 protein-coding genes, 22 tRNA and 2 rRNA genes. Nucleotide compositions of its whole mitogenome are 39.09% for A, 38.49% for T, 13.42% for C, and 9.01% for G. Consistent with previous observations of Tipulidae species, the mitogenome of Pselliophora bifascipennis is highly conserved in gene size, organization and codon usage, and secondary structures of tRNAs. Most tRNAs have the typical clover-leaf structure. The control region is 1006 bp long with an A + T content of 92.7%. Phylogenetic tree analysis using the sequences of the mitochondrial genomes of Pselliophora bifascipennis and other Tipulidae species showed that Pselliophora bifascipennis is closely related to Tanyptera hebeiensis. These two species are grouped on the same branch, which is in accordance with the traditional morphological classification. The results of this study lay a foundation for screening molecular markers of mitochondrion for molecular identification and genetic structure research in Tipulidae species.

Characterization and Phylogenetic Implications of the Complete Mitochondrial Genome of Syrphidae.

Project description:In this study, the complete mitochondrial genomes (mitogenomes) of two hoverfly species of Korinchia angustiabdomena (Huo, Ren, and Zheng) and Volucella nigricans Coquillett (Diptera: Syrphidae) were determined and analyzed. The circular mitogenomes were 16,473 bp in K. angustiabdomena (GenBank No. MK870078) and 15,724 bp in V. nigricans (GenBank No. MK870079). Two newly sequenced mitogenomes both contained 37 genes, and the gene order was similar with other syrphine species. All the protein-coding genes (PCGs) were started with the standard ATN codons; and most of PCGs were terminated with a TAA stop codon, while ND1 in K. angustiabdomena ended with a TAG codon, and ND5 terminated with truncated T stop codons in both species. The phylogenetic relationship between K. angustiabdomena and V. nigricans with related lineages was reconstructed using Bayesian inference and Maximum-likelihood analyses. The monophyly of each family considered within Muscomorpha was confirmed by the clades in the phylogenetic tree, and superfamily of the Oestroidea (Calliphoridae, Sarcophagidae, and Oestridae) was unexpectedly found to be a paraphyletic group based on our selected data. This mitogenome information for K. angustiabdomena and V. nigricans could facilitate future studies of evolutionarily related insects.

Phylogenetic insights based on the first complete mitochondrial genome of Isomyia nebulosa (Diptera: Calliphoridae).

Project description:To investigate the phylogenetic position of Isomyia Walker, 1860, a genus that suffered from frequent revisions of the taxonomic status following the subfamily Rhiniinae (Diptera, Calliphoridae), we sequenced, assembled, annotated, and analyzed the first complete mitochondrial genome (mitogenome) of Isomyia nebulosa (Townsend, 1917) in this study. This mitogenome is 16,438 bp in length, with a standard set of 13 protein-coding genes (PCGs), 22 tRNAs, two rRNAs, and an A + T riched non-coding region without genetic rearrangement as most dipteran mitogenomes, but long intergenic nucleotides (IGNs) between trnQ and trnM are found. The phylogeny yielded by both Bayesian inference and maximum-likelihood analysis for all mitochondrial PCGs and rRNAs of 23 mitogenomes supports the monophyly of the family Calliphoridae and the subfamilies Calliphorinae, Chrysomyinae, and Luciliinae. In addition, I. nebulosa diverged anterior to the above-mentioned three calliphorid subfamilies with high genetic distances.

First records of Pseudodoros nigricollis Becker (Diptera: Syrphidae) from Cyprus.

Project description:BackgroundThe hoverfly Pseudodoros nigricollis Becker, 1903 is a rarely collected species, of which information on its distribution and ecology is poorly understood.New informationIn this paper the first records of the hoverfly Pseudodoros nigricollis from Cyprus are provided and discussed. The discovery indicates that this Afrotropical species is approaching the European continent. Short notes on the habitat in which it has been collected are provided. The relationship with the mealy plum aphid Hyalopterus pruni is discussed. Clues on further research are given.
