Partial mitochondrial genome of the Sanchiang Tree Toad Hyla sanchiangensis (Anura: Hylidae).

ABSTRACT: We reported the partial mitochondrial genome (mitogenome) for Hyla sanchiangensis (Anura: Hylidae), a arboreal frog and endemic in China. The length of partial mitogenome of H. sanchiangensis was 15,664 bp, and contained PCGs (COX1-3, ND1-6, ND4L, ATP6, ATP8 and CYTB), 2 ribosomal RNA genes, 22 transfer RNA genes, and 2 non-coding regions of a L-strand replication origin and a partial loop region. The overall base composition of the sequence is 29.91% A, 29.86% T, 14.58% G, and 25.65% C, with a total A + T content of 59.77%. The result of phylogenetic analysis showed that H. sanchiangensis formed a clade with other species belonging to the genus of Hyla. This mitogenome data could help in evolutionary biology and population genetics of the Hylid species.

Project description:The complete mitochondrial genome of a treefrog (Hyla sp.) was determined. The circular mitochondrial genome is 18,288 bp long and encodes 13 proteins, 22 transfer RNAs, and 2 ribosomal RNAs. Phylogenetic analysis of its full genome sequences showed that H. sp. was closely related to H. ussuriensis and H. japonica rather than Dryophytes suweonensis, consistent with the results from each protein-coding gene and a cluster between 12S rRNA and 16S rRNA. The present study will provide essential genomic information for biogeographical distribution and evolutionary history of an endemic treefrog, H.sp.

Sequencing and analysis of the complete mitochondrial genome of Hyla suweonensis (Anura: Hylidae).

Project description:The present study reports the complete mitochondrial genome of the Suwon tree frog Hyla suweonensis from South Korea. This endangered species is endemic to Korea. The circular mitogenome of H. suweonensis includes 16,895 bp length and contains 13 protein-coding genes, 2 ribosomal RNA genes, 22 transfer RNA genes, and a non-coding region, which is the typical gene arrangement found in the available Hylinae mitogenomes. Phylogenetic analysis of six Hyla spp. mitochondrial genomes revealed that H. suweonensis is closer to H. tsinlingensis.

Sequencing of complete mitochondrial genome for Tsinling Tree Toad (Hyla tsinlingensis).

Project description:The complete mitochondrial genome sequence of Hyla tsinlingensis was determined in this research. The length of mitogenome is 17850 bp, including 13 protein-coding genes, 22 tRNA genes, 2 rRNA genes, 1 OL and 1 control region. The phylogentic tree was reconstructed using the BI method based on concatenated nucleotide sequences of mtDNA genes (12S ribosomal small subunit gene/12S rRNA; NADH dehydrogenase subunit 1 gene/ND1, including adjacent transfer RNAs and the partial 16S ribosomal large subunit gene). The phylogenetic tree was split into two clades, Clade A and Clade B. The H. tsinlingensis which we determined clustered into Clade A.

Complete mitochondrial genome sequence of the high-altitude Brazilian tree frog Bokermannohyla alvarengai (Anura, Hylidae).

Project description:The first complete mitochondrial genome (mtDNA) for the genus Bokermannohyla (Hylidae) is presented. The mtDNA contains 17,325 bp and is similar in size, gene content, and gene location to other hylid mitochondrial genomes described, with 2 rRNA, 22 tRNA, and 13 PCGs. The control region (D-loop) is shorter than in mtDNAs of hylids from Asia. A phylogenetic tree based on homologous genes did not corroborate the monophyly of Hylidae neither the recently proposed monophyly of Hyla and Dryophytes.

Complete mitochondrial genome of Dryophytes suweonensis (Anura Hylidae).

Project description:Dryophytes suweonensis is an endangered species with fragmented and declining populations from the Korean peninsula. We described 17,448 bp of D. suweonensis mtDNA, which had a shorter D-loop than other closely related species. The variation in nucleotide composition was similar to that of Hyla tsinlingensis but was larger than the one of its sister clade, D. japonicus.

Characterization of Two Mitogenomes of Hyla sanchiangensis (Anura: Hylidae), with Phylogenetic Relationships and Selection Pressure Analyses of Hylidae.

Project description:Hyla sanchiangensis (Anura: Hylidae) is endemic to China and is distributed across Anhui, Zhejiang, Fujian, Guangdong, Guangxi, Hunan, and Guizhou provinces. The mitogenomes of H. sanchiangensis from two different sites (Jinxiu, Guangxi, and Wencheng, Zhejiang) were sequenced. Phylogenetic analyses were conducted, including 38 mitogenomes of Hylidae from the NCBI database, and assessed the phylogenetic relationship of H. sanchiangensis within the analyzed dataset. Two mitogenomes of H. sanchiangensis showed the typical mitochondrial gene arrangement with 13 protein-coding genes (PCGs), two ribosomal RNA genes (12S rRNA and 16S rRNA), 22 transfer RNA (tRNA) genes, and one non-coding control region (D-loop). The lengths of the 12S rRNA and 16S rRNA genes from both samples (Jinxiu and Wencheng) were 933 bp and 1604 bp, respectively. The genetic distance (p-distance transformed into percent) on the basis of the mitogenomes (excluding the control region) of the two samples was calculated as 4.4%. Hyla sanchiangensis showed a close phylogenetic relationship with the clade of (H. annectans + H. tsinlingensis), which was supported by ML and BI analyses. In the branch-site model, five positive selection sites were found in the clade of Hyla and Dryophytes: Cytb protein (at position 316), ND3 protein (at position 85), and ND5 protein (at position 400) have one site, respectively, and two sites in ND4 protein (at positions 47 and 200). Based on the results, we hypothesized that the positive selection of Hyla and Dryophytes was due to their experience of cold stress in historical events, but more evidence is needed to support this conclusion.

Project description:In this study, the complete mitochondrial genome of Duttaphrynus himalayanus was sequenced adopting Illumina high-throughput sequencing method. The complete mitogenome of the species was 17,172 bp in length, including 13 protein-coding genes, 22 transfer RNA (tRNA) genes, two ribosomal RNA genes, and a non-coding control region (CR). The overall base composition of mitogenome was 29.7% A, 29.6% T, 26.0% C, and 14.7% G. Most mitochondrial genes are encoded on the heavy strand, only ND6 and eight tRNA genes on the light strand. The complete mitogenome of D. himalayanus can provide an important data for future studies on phylogenetic relationship and population genetics of this species.

The mitogenome size is 17,485 bp, containing 13 protein-coding genes, 2 rRNA genes, 23 tRNA genes and a control region (D-loop). The base composition of the whole genome is 28.1% A, 32.6% T, 24.4% C and 14.8% G. As observed in this genus before, this mitogenome also appears a tandem duplication of tRNAMet gene. This study will provide fundamental data for further research to resolve population genetics of this species and systematic problems of the genus Vibrissaphora.

Complete mitochondrial genome sequence of the Asiatic Toad Bufo Gargarizans (Amphibia, Anura, Bufonidae).

Project description:The Asiatic toad Bufo gargarizans belongs to Bufonidae. This species is known from the Russian Far East, central, northern and north-eastern China, the Democratic People's Republic of Korea and Japan. In this study, the complete mitochondrial genome of B. gargarizans was sequenced. The mitogenome was 17,407 bp in length, consisting of 13 protein-coding genes, 22 transfer RNA (tRNA) genes, two ribosomal RNA (rRNA) genes, and a non-coding control region. As in other vertebrates, most mitochondrial genes are encoded on the heavy strand, except for ND6 and eight tRNA genes which are encoded on the light strand. The overall base composition of the B. gargarizans is 28.9% A, 28.2% T, 27.5% C, and 15.3% G. Phylogenetic analysis showed B. gargarizans was closely related to B. bankorensis and B. tibetanus. The complete mitogenome of B. gargarizans can provide an important data for the studies on phylogenetic relationship and population genetics to further explore the taxonomic status of this species.

The complete mitochondrial genome of the Baishanzu horned toad Boulenophrys baishanzuensis (Anura: Megophryidae).

Project description:The mitochondrial genome (mitogenome) of Boulenophrys baishanzuensis (Anura: Megophryidae) was sequenced by the Illumina platform. The assembled circular mitogenome of B. baishanzuensis had a total length of 17,040 bp, with a GC content of 41.25%. It consisted of 13 protein-coding genes (PCGs), two rRNA genes, 22 tRNA genes, and a D-loop region. The majority of the PCGs were encoded by the H-strand, while one PCG (nad6) and eight tRNA genes (tRNA-Gln, tRNA-Ala, tRNA-Asn, tRNA-Cys, tRNA-Tyr, tRNA-Ser2, tRNA-Glu, and tRNA-Pro) were encoded in the L-strand. Phylogenetic analysis revealed that the newly sequenced species formed a clade with other Boulenophrys species, while the genus Boulenophrys itself formed a sister group with the genus Atympanophrys.
