Complete mitochondrial genome of the whitetip reef shark Triaenodon obesus from the British Indian Ocean Territory Marine Protected Area.

ABSTRACT: We present the first mitochondrial genome of Trianenodon obesus from the Chagos Archipelago in the British Indian Ocean Territory (BIOT) Marine Protected Area. The mitogenome was 16,702 bp in length and consisted of 13 protein-coding genes (PCGs), 22 tRNA genes, 2 rRNA genes, and a non-coding control region (D-loop). GC content was at 38.9%. The control region was 1064 bp in length. This mitogenome for the BIOT MPA T. obesus differed from the previously published T. obesus genome by 15 bp and the differences include a 2 bp insertion and 13 single nucleotide polymorphisms distributed across the mitogenome in the BIOT MPA sequence. Whole mitogenome sequence of T. obesus from the Chagos archipelago presented here fills existing gaps in genetic information on marine species from the BIOT MPA and provides additional tools for species specific assessments as to the effectiveness of MPA management. In addition, methods presented here lay the framework for genetic studies in remote locations with limited infrastructure.

Project description:BACKGROUND:The spread of carbapenemase-producing Enterobacteriaceae (CPE) in the Southwest Indian Ocean area (SIOA) is poorly documented. Reunion Island is a French overseas territory located close to Madagascar and connected with Southern Africa, Indian sub-continent and Europe, with several weekly flights. Here we report the results of the CPE surveillance program in Reunion Island over a six-year period. METHODS:All CPE were collected between January 2011 and December 2016. Demographics and clinical data of the carrier patients were collected. We determined their susceptibility to antimicrobials, identified the carbapenemases and ESBL by PCR and sequencing, and explored their genetic relationship using pulsed-field gel electrophoresis and multi-locus sequence typing. RESULTS:A total of 61 CPEs isolated from 53 patients were retrieved in 6 public or private laboratories of the island. We found that 69.8% of CPE patients were linked to a foreign country of SIOA and that almost half of CPE cases (47.2%) reached the island through a medical evacuation. The annual number of CPE cases strongly increased over the studied period (one case in 2011 vs. 21 cases in 2016). A proportion of 17.5% of CPE isolates were non-susceptible to colistin. blaNDM was the most frequent carbapenemase (79.4%), followed by blaIMI (11.1%), and blaIMP-10 (4.8%). Autochtonous CPE cases (30.2%) harboured CPE isolates belonging to a polyclonal population. CONCLUSIONS:Because the hospital of Reunion Island is the only reference healthcare setting of the SIOA, we can reasonably estimate that its CPE epidemiology reflects that of this area. Mauritius was the main provider of foreign CPE cases (35.5%). We also showed that autochthonous isolates of CPEs are mostly polyclonal, thus unrelated to cross-transmission. This demonstrates the local spread of carbapenemase-encoding genes (i.e. blaNDM) in a polyclonal bacterial population and raises fears that Reunion Island could contribute to the influx of NDM-carbapenemase producers into the French mainland territory.

Project description:AimUnderstanding the drivers of the structure of coral reef fish assemblages is vital for their future conservation. Quantifying the separate roles of natural drivers from the increasing influence of anthropogenic factors, such as fishing and climate change, is a key component of this understanding. It follows that the intrinsic role of historical biogeographical and geomorphological factors must be accounted for when trying to understand the effects of contemporary disturbances such as fishing.LocationComoros, Madagascar, Mozambique and Tanzania, Western Indian Ocean (WIO).MethodsWe modeled patterns in the density and biomass of an assemblage of reef-associated fish species from 11 families, and their association with 16 biophysical variables.ResultsCanonical analysis of principal coordinates revealed strong country affiliations of reef fish assemblages and distance-based linear modeling confirmed geographic location and reef geomorphology were the most significant correlates, explaining 32% of the observed variation in fish assemblage structure. Another 6%-8% of variation was explained by productivity gradients (chl_a), and reef exposure or slope. Where spatial effects were not significant between mainland continental locations, fishing effects became evident explaining 6% of the variation in data. No correlation with live coral was detected. Only 37 species, predominantly lower trophic level taxa, were significant in explaining differences in assemblages between sites.Main conclusionsSpatial and geomorphological histories remain a major influence on the structure of reef fish assemblages in the WIO. Reef geomorphology was closely linked to standing biomass, with "ocean-exposed" fringing reefs supporting high average biomass of ~1,000 kg/ha, while "lagoon-exposed fringing" reefs and "inner seas patch complex" reefs yielded substantially less at ~500kg/ha. Further, the results indicate the influence of benthic communities on fish assemblages is scale dependent. Such insights will be pivotal for managers seeking to balance long-term sustainability of artisanal reef fisheries with conservation of coral reef systems.
