The complete mitochondrial genome sequence of Cletus rubidiventris (Heteroptera: Coreidae).

ABSTRACT: Cletus rubidiventris is a crop pest, especially for rice. This study first reported the complete mitochondrial genome of this species. The total length of mitochondrial genome is 15,590 bp and including 13 PCGs, 22 tRNA genes, and 2 rRNA genes, with 31.8% T, 15.8% C, 41.6% A, and 10.8% G. The overall GC content of the genome is 27%. The mitochondrial genome order, nucleotide composition, and codon usage pattern is similar to C. punctiger. The phylogenetic tree shows that C. rubidiventris belong to the Coreidae.

The maximum likelihood (ML) phylogenetic tree confirmed that C. punctiger belonged to the Coreidae subfamily.

Project description:The complete 16,345-bp mitochondrial genome of the agriculturally destructive pod sucking pest, the giant coreid bug, Anoplocnemis curvipes (Hemiptera: Coreidae), was assembled from paired-end Illumina HiSeq 2500 reads. The A. curvipes mitochondrial genome consists of 13 protein coding genes (PCGs), 22 tRNAs, 2 rRNAs and a control region in the order and orientation typical among insects. PCG initiation codons (ATG, ATC, ATT and ATA) with termination codon (TAA) are used with the exception of TAG stop codons by Cytb and ND3. All tRNA genes fold into predicted cloverleaf secondary structures having requisite triplets on the anticodon loop, apart from tRNA-Ser1 (AGN) whose dihydrouridine (DHU) arm forms a simple loop. The phylogenetic analysis of hemipteran mitogenomes clusters to the family level and supports the monophyly of the five superfamilies in Pentatomomorpha of Hemiptera. The Coreoidea and Pyrrhocoroidea are sister groups, while Coreidae and Alydidae are sister groups to Rhopalidae. These analyses provide insight to mitogenomics and evolutionary relationships among pentatomoid insects.

Complete mitochondrial genome sequence and phylogenetic implications of Chorosoma macilentum (Heteroptera: Rhopalidae).

Project description:The complete mitogenome of Chorosoma macilentum Stål, 1858 was obtained using a next-generation sequencing approach, which is the first report in genus Chorosoma. The genome is 16,330 bp long with high A + T content. The phylogenetic analyses based on 13 protein-coding genes (PCGs) provide strong support for monophyly of each family within Pentatomomorpha. Chorosoma Macilentum was clustered together with other species within Rhopalidae, and grouped sibling species with Aeschyntelus notatus.

The first complete mitochondrial genome from the subfamily Phyllocephalinae (Heteroptera: Pentatomidae) and its phylogenetic analysis.

Project description:The true bug, Gonopsis affinis, is an important pest in China. Here, we determined the complete mitogenome of G. affinis, which is the first for the subfamily Phyllocephalinae. This 16,011-basepair (bp) mitogenome comprises of 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), two ribosomal RNA genes, and an A + T-rich region. The gene order and the orientation are similar to those of other sequenced Hemipteran species. All PCGs start with ATN codons, except COI, and end with TAA. The 22 tRNAs have a typical cloverleaf secondary structure except trnS-Ser (AGN), which lacks a dihydrouridine (DHU) arm. Phylogenetic analyses highly supported the monophyly of each family, and confirmed the reasonable placement of G. affinis.

Functional Immunomics of the Squash Bug, Anasa tristis (De Geer) (Heteroptera: Coreidae).

Project description:The Squash bug, Anasa tristis (De Geer), is a major piercing/sucking pest of cucurbits, causing extensive damage to plants and fruits, and transmitting phytopathogens. No genomic resources to facilitate field and laboratory studies of this pest were available; therefore the first de novo exome for this destructive pest was assembled. RNA was extracted from insects challenged with bacterial and fungal immunoelicitors, insects fed on different cucurbit species, and insects from all life stages from egg to adult. All treatments and replicates were separately barcoded for subsequent analyses, then pooled for sequencing in a single lane using the Illumina HiSeq2000 platform. Over 211 million 100-base tags generated in this manner were trimmed, filtered, and cleaned, then assembled into a de novo reference transcriptome using the Broad Institute Trinity assembly algorithm. The assembly was annotated using NCBIx NR, BLAST2GO, KEGG and other databases. Of the >130,000 total assemblies 37,327 were annotated identifying the sequences of candidate gene silencing targets from immune, endocrine, reproductive, cuticle, and other physiological systems. Expression profiling of the adult immune response was accomplished by aligning the 100-base tags from each biological replicate from each treatment and controls to the annotated reference assembly of the A. tristis transcriptome.

Characterization of the complete mitochondrial genome of Eysarcoris aeneus (Heteroptera: Pentatomidae), with its phylogenetic analysis.

Project description:In this study, the complete 16,065 bp mitochondrial genome of Eysarcoris aeneus was determined from China using next-generation sequencing technology. It consists of 13 protein-coding genes, 22 tRNAs, 2 ribosomal RNAs, and a control region. Among them, 23 genes were located on J-strand, and the other genes (nad5, nad4, nad4L, nad1, trnQ, trnC, trnY, trnF, trnH, trnP, trnL1, trnV, 12S rRNA, and 16S rRNA) were located on the N-strand. The phylogenetic tree was constructed using MrBayes method based on the 13 protein-coding genes (PCGs) showed the present species clustered within the Pentatomidae. The complete mitogenome data would be useful for further study of Pentatomoidea and Pentatomomorpha.

Complete mitochondrial genome sequence of Hemiculter bleekeri.

Project description:In our research, the complete mitochondrial DNA (mtDNA) of Hemiculter bleekeri was obtained from the dorsal myotome of the fish. The total length of the mitochondrial genome is 16,617 bp and deposited in GenBank with accession number KU198332. The gene arrangement was similar to other bony fishes, which contained 37 genes (13 protein-coding genes, 2 ribosomal RNA and 22 transfer RNAs) and a major non-coding control region. The mtDNA may provide some important genetic background information of this valuable fish. The G contents was relatively low in the total mtDNA with the GCskewthinsp;= -0.22. The negative GC-skew ranging from -0.51(ATP8) to -0.19(CO1), and the more positive AT-skew varying from -0.26(ATP8) to -0.03(CO1). The phylogenetic analysis demonstrated that H. Bleekeri was clustered together with Hemiculter leucisculus, which could be a sister species.

Complete mitochondrial genome sequence of Gymnopilus junonius.

Project description:Gymnopilus junonius, a well-known poisonous mushroom, is distributed worldwide. It contains a hallucinogenic alkaloid psilocybin and several other bioactive compounds. The mitochondrial genome, a circular DNA molecule of 161,145 bp, comprises 15 protein-coding genes, 24 transfer RNA genes, and 2 ribosomal RNA genes. The guanine-cytosine content was 31.56%. Based on the mitochondrial genome sequence, a phylogenetic tree was constructed to demonstrate the phylogenetic relationship. In this study, the phylogenetic positions of G. junonius and its related genera were determined.
