Impacts of Citric Acid and Malic Acid on Fermentation Quality and Bacterial Community of Cassava Foliage Silage.

ABSTRACT: The microbiota and fermentation quality of cassava foliage (CF) ensiled in the absence of additive (CK), or the presence of citric acid (CA), malic acid (MA), and their combination with a Lactobacillus plantarum strain (CAL and MAL)were investigated. These additives reduced (P < 0.05) the pH, butyric acid, and ammonia-N contents but increased (P < 0.05) the lactic acid content, and CAL and MAL showed similar remarkable effects. Paenibacillus (mean, 27.81%) and Bacillus (mean, 16.04%) were the predominant strains in CF silage. The addition of CA or MAL increased the abundance of Paenibacillus (25.81-52.28% and 47.97%, respectively), and the addition of MA increased the abundance of Bacillus (15.76-32.48%) compared with the CK group. Moreover, CAL and MAL increased the abundances of the potentially desirable bacteria Cellulosimicrobium (CAL 0-12.73%), Hyphomicrobium (0-7.90% and 8.94%), and Oceanobacillus (0-8.37% and 3.08%) compared with the CK group. These findings suggested that CA and MA could enhance the silage quality of CF, and their combinations with Lactobacillus plantarum were more effective.

Project description:Sugarcane tops silage (STS), as a source of roughage for ruminants, is rich in water-soluble carbohydrate (WSC) content, which significantly affects silage quality. Citric acid (CA) is a low-cost natural antimicrobial agent that can inhibit undesirable microbes and improve silage quality. The objectives of this study were to investigate the effects of CA on the chemical composition, fermentation quality, microbial communities, and metabolic pathways of STS with high and low WSC contents before or after aerobic exposure. Fresh sugarcane tops with low-WSC [143.05 g/kg dry matter (DM)] and high-WSC (249.99 g/kg DM) contents were treated with and without CA and then ensiled for 125 days, followed by aerobic exposure for 4, 8, and 16 days. The results showed that high-WSC STS had lower crude protein (CP) content and higher DM, neutral detergent fiber (NDF), and acid detergent fiber (ADF) contents, whether treated with CA or not. CA-inoculated silage exhibited decreased DM loss and enterobacteria (EB) counts compared to the control. High-WSC STS treated with CA had higher WSC content and lower yeast count than those without CA inoculation. During the 0-16 days of aerobic exposure, the propionic acid and butyric acid contents in CA-inoculated silage were almost unchanged and ranged from 0 to 1 g/kg DM. Meanwhile, the ethanol content was almost unchanged and ranged from 0 to 1 g/kg DM only in low-WSC STS, irrespective of CA addition. Before aerobic exposure, CA inoculation decreased the abundances of undesirable microbes (e.g., Clostridium_sensu_stricto_12 and Paecilomyces) and animal pathogens, while amino acid metabolism was lower in high-WSC STS regardless of CA treatment. After aerobic exposure, CA inoculation increased the abundance of bacteria with antibacterial effects, including Paenibacillus and Bacillus. Moreover, the metabolism of energy and nucleotides was lower in high-WSC STS treated with CA, and the animal pathogens was lower in low-WSC STS treated with CA. In conclusion, CA inoculation could be effective in decreasing nutrients loss, improving fermentation quality, inhibiting harmful microorganisms, and modulating the metabolic pathways of microorganisms in STS with high and low WSC contents prior to and after aerobic exposure.

Project description:To better understand the ensiling characteristics of sorghum stalk, the dynamic changes of fermentation parameters, nutrient components and bacterial community of sorghum stalk silage were analyzed by intermittently sampling on day 0, 1, 3, 7, 14, 28, and 56 of ensiling duration. The results showed that high-moisture sorghum stalk was well preserved during ensiling fermentation, with the DM loss of 4.10% and the little difference between the nutrients of sorghum stalk before and after ensiling. The pH value of silage declined to its lowest value of 4.32 by Day 7 of ensiling, and other fermentation parameters kept steady since Day 28 of ensiling. The amplicon sequencing analysis revealed that the alpha diversity parameters of silage bacterial community including Shannon index, observed features, Pielou evenness and Faith PD gradually declined (P < 0.01) with ensiling duration. Principal coordinate analysis (PCoA) revealed that bacterial profiles of raw material would experience a succession becoming a quite different community during ensiling fermentation. Taxonomic classification revealed a total of 10 and 173 bacterial taxa at the phylum and genus level, respectively, as being detected with relative abundances higher than 0.01% and in at least half samples. LEfSe analysis revealed that 26 bacterial taxa were affected by sampling timepoint (P < 0.05 and LDA score > 4). When focusing on the dynamic trend of silage bacterial taxa, lactic acid bacteria successfully dominated in the bacterial community on Day 1 of ensiling, and the bacterial community almost came to a plateau by Day 28 of ensiling, with Lactobacillus and Leuconostoc as the dominant genera. In a word, the succession of fermentation parameters, nutrient components and bacterial community indicate a successful dominance establishment of LAB and a fast advent of fermentation plateau, suggesting that high-moisture sorghum stalk can be ensiled directly, but the pH of mature silage is a little high.

Project description:To investigate the effects of wilting and lactic acid bacterial inoculants on the fermentation quality and bacterial community of Moringa oleifera leaf silage, fresh and wilted M. oleifera leaves were ensiled with or without Lactobacillus farciminis LF or Lactococcus lactis LL for 1, 7, 14, 30, and 60 days. The results showed that wilting, inoculants, and their interaction exerted significant (P < 0.05) effects on the fermentation characteristics covering dry matter loss, pH value, lactic acid bacterial number, the ratio of lactic acid to acetic acid, and the relative abundances of bacteria, like for species of Lactobacillus, Lactococcus, Pediococcus, Enterococcus, Leuconostoc, and Enterobacter Both LF and LL improved the fermentation quality of wilted and unwilted M. oleifera leaf silage by accelerating lactic acid production and pH decline, decreasing dry matter loss, and inhibiting yeast and coliform bacterial growth through the whole fermentation process. During ensiling, the abundances of Lactococcus, Enterococcus, and Leuconostoc spp. increased from day 1 to day 7 and then declined sharply from day 7 to day 14. Members of these genera and Enterobacter were inhibited, whereas Lactobacillus spp. were enhanced by these two lactic acid bacterial inoculants. The relative abundances of Enterobacter, Enterococcus, and Pediococcus spp. in inoculated silages were relatively low during the whole ensiling process. A lower abundance of Enterobacter spp. was observed in wilted silages than in unwilted silages. In summary, wilting and lactic acid bacterial inoculants had an influence on bacterial community and the fermentation process; LF and LL improved the fermentation quality of wilted and unwilted M. oleifera leaf silage.IMPORTANCEMoringa oleifera leaf is a high-quality feed source for livestock and is increasingly used all over the world. Ensiling might be an effective method for preservation of the leaves. In the practice of silage making, lactic acid bacterial inoculants and wilting are commonly used to improve nutrition preservation. Monitoring the changes in a bacterial community during fermentation gives an insight into understanding and improving the ensiling process. Our results suggest that wilting and lactic acid bacterial inoculants had an influence on the bacterial community and fermentation process of M. oleifera leaf silage. Wilting showed positive effects on silage fermentation by decreasing the abundance of Enterobacter spp., while LF and LL improved the fermentation quality by inhibiting Enterobacter spp. and enhancing Lactobacillus spp. Both LF and LL accelerated the ensiling process from cocci (like Lactococcus, Enterococcus, and Leuconostoc spp.) to lactobacilli.
