The complete mitochondrial genome of the Basidiomycete edible fungus Pleurotus eryngii.

ABSTRACT: The complete mitochondrial genome of the edible fungus Pleurotus eryngii (oyster mushroom) was determined using Illumina sequencing. This mitogenome is a circular molecule of 72,650 bp in length with a GC content of 26.28%. Gene prediction showed that 40 putative protein-coding genes, the small ribosomal RNA subunits (rns), and 23 tRNAs were located on the same strand. The mitogenome of P. eryngii has a similar structure to that of P. ostreatus in both gene content and gene order. The mitogenome information of P. eryngii should contribute to our understanding of the diversity and evolution of Pleurotaceae and Agaricales.

Project description:Pleurotus cornucopiae is a commercial edible and medicinal fungus. Herein, we determined and analyzed its complete mitochondrial genome. The mitogenome length was 72,134 bp with a GC content of 26.7%, contained 14 conserved protein coding genes, two rRNA genes (rnl and rns), ribosomal protein gene rps3 and 24 tRNA genes. Pleurotus cornucopiae has a similar gene content and gene order of the mitogenome as P. ostreatus and P. eryngii. A phylogenetic analysis based on complete mitogenome in related fungi showed that P. cornucopiae is a member of the order Agaricales, forming a clade with P. ostreatus and P. eryngii, with P. ostreatus as a sister taxa. The mitochondrial genome sequence of P. cornucopiae appeared a promising tool for further studies of the taxonomy and evolution of Pleurotaceae and Agaricales.

It contains 14 putative protein-coding genes, the ribosomal RNA subunits and 23 tRNAs, all located on the same strand. The evolutionary relationships between P. portentosus and other 23 representative basidiomycete species were revealed based on sequences at the 14 concatenated mitochondrial protein-coding genes.

Project description:The complete mitochondrial genome of the edible fungus Thelephora ganbajun was determined using Illumina sequencing. This mitogenome is a circular molecule of 52,857 bp in length with a GC content of 25.73%. Gene prediction showed that the mitogenome codes 28 tRNAs, 2 pseudo-tRNAs, and 21 known and 7 hypothetical proteins. The evolutionary relationships between Th. ganbajun and other representative species based on the mitogenome are consistent with those based on nuclear genes. The mitogenome information of Th. ganbajun should contribute to our understanding of the diversity and evolution of Thelephorales.

Project description:Pleurotus giganteus (Berk.) Karunarathna & K.D. Hyde 2011 is one of the largest edible mushrooms integrating medicinal value and edible value. The complete mitochondrial genome of the edible fungus P. giganteus was published in this paper. It was determined using Pacbio and Illumina sequencing. The circular molecule is 102,950 bp in length, consisting of 30 protein-coding genes (PCGs), two ribosomal RNA (rRNA) genes, and 24 transfer RNA (tRNA) genes. The base composition of the whole mitogenome is A (37.3%), T (37.7%), G (12.2%), and C (12.8%). The phylogenetic tree shows P. giganteus was the basal taxon in Pleurotus and closely related to Pleurotus citrinopileatus Singer 1990.

Identification of the Antidepressant Function of the Edible Mushroom Pleurotus eryngii.

Project description:Pleurotus eryngii produces various functional molecules that mediate physiological functions in humans. Recently, we observed that P. eryngii produces molecules that have antidepressant functions. An ethanol extract of the fruiting body of P. eryngii was obtained, and the extract was purified by XAD-16 resin using an open column system. The ethanol eluate was separated by HPLC, and the fraction with an antidepressant function was identified. Using LC-MS, the molecular structure of the HPLC fraction with antidepressant function was identified as that of tryptamine, a functional molecule that is a tryptophan derivative. The antidepressant effect was identified from the ethanol extract, XAD-16 column eluate, and HPLC fraction by a serotonin receptor binding assay and a cell-based binding assay. Furthermore, a forced swimming test (FST) showed that the mice treated with purified fractions of P. eryngii exhibited decreased immobility time compared with nontreated mice. From these results, we suggest that the extract of P. eryngii has an antidepressant function and that it may be employed as an antidepressant health supplement.

Genome-wide gene expression patterns in dikaryon of the basidiomycete fungus Pleurotus ostreatus.

Project description:Dikarya is a subkingdom of fungi that includes Ascomycota and Basidiomycota. The gene expression patterns of dikaryon are poorly understood. In this study, we bred a dikaryon DK13×3 by mating monokaryons MK13 and MK3, which were from the basidiospores of Pleurotus ostreatus TD300. Using RNA-Seq, we obtained the transcriptomes of the three strains. We found that the total transcript numbers in the transcriptomes of the three strains were all more than ten thousand, and the expression profile in DK13×3 was more similar to MK13 than MK3. However, the genes involved in macromolecule utilization, cellular material synthesis, stress-resistance and signal transduction were much more up-regulated in the dikaryon than its constituent monokaryons. All possible modes of differential gene expression, when compared to constituent monokaryons, including the presence/absence variation, and additivity/nonadditivity gene expression in the dikaryon may contribute to heterosis. By sequencing the urease gene poure sequences and mRNA sequences, we identified the monoallelic expression of the poure gene in the dikaryon, and its transcript was from the parental monokaryon MK13. Furthermore, we discovered RNA editing in the poure gene mRNA of the three strains. These results suggest that the gene expression patterns in dikaryons should be similar to that of diploids during vegetative growth.

Complete mitochondrial genome of the edible Basidiomycete mushroom Thelephora aurantiotincta (Aphyllophorales: Thelephoraceae) from China.

Project description:The complete mitochondrial genome of Thelephora aurantiotincta, an edible Basidiomycete mushroom species with ecological and economic value is reported in this study. The whole genome is a circular molecule 50,672 bp in length and encodes 42 genes as follows: 15 protein-coding genes, two rRNA genes and 25 tRNA genes. The A, T, C, G contents in the genome are 35.60%, 35.31%, 13.89%, and 15.20%, respectively. Phylogenetic analysis revealed a close relationship between T. aurantiotincta and T. ganbajun. This is the first complete mitochondrial genome for T. aurantiotincta that will be useful for providing basic genetic information for this important species.

Olive Mill Waste Enhances α-Glucan Content in the Edible Mushroom Pleurotus eryngii.

Project description:Mushroom polysaccharides are edible polymers that have numerous reported biological functions; the most common effects are attributed to β-glucans. In recent years, it became apparent that the less abundant α-glucans also possess potent effects in various health conditions. Here we explore several Pleurotus species for their total, β and α-glucan content. Pleurotus eryngii was found to have the highest total glucan concentrations and the highest α-glucans proportion. We also found that the stalks (stipe) of the fruit body contained higher glucan content then the caps (pileus). Since mushrooms respond markedly to changes in environmental and growth conditions, we developed cultivation methods aiming to increase the levels of α and β-glucans. Using olive mill solid waste (OMSW) from three-phase olive mills in the cultivation substrate. We were able to enrich the levels mainly of α-glucans. Maximal total glucan concentrations were enhanced up to twice when the growth substrate contained 80% of OMSW compared to no OMSW. Taking together this study demonstrate that Pleurotus eryngii can serve as a potential rich source of glucans for nutritional and medicinal applications and that glucan content in mushroom fruiting bodies can be further enriched by applying OMSW into the cultivation substrate.

This mitogenome is a circular molecule of 93,439 bp with a GC content of 29.28% and contains 15 protein-coding, two rRNA (rnl and rns), and 24 tRNA genes. Phylogenetic analysis revealed that D. tabescens is genetically closest to Agrocybe aegerita. Desarmillaria tabescens mitogenome can contribute to our understanding of the phylogeny and evolution of this species.

Project description:The complete mitochondrial genome of the widely cultivated edible fungus Lentinula edodes was determined using the next-generation sequencing technology. The circular molecule is 116,819 bp in length with a GC content of 30.75%. Conserved genes including 13 putative protein-coding genes and 24 tRNAs were located on the same strand. We detected 14 introns invading 4 genes, including cob, cox1, nad1, and nad5. The phylogenetic analysis confirmed that L. edodes was a number of Agaricales. This mitochondrial genome may open new avenues for understanding the phylogeny and evolution of Omphalotaceae and Agaricales.
