The complete mitochondrial genome of the hybrid of Hoplobatrachus chinensis (?)×H. rugulosus (?) and its phylogeny.

ABSTRACT: The complete mitochondrial genome sequence of the hybrid of Hoplobatrachus chinensis (?)?×?H. rugulosus (?) was obtained in this study. The circular mitochondrial genome was 20,282?bp in length (including extra ND5 genes). Compared with the complete mitogenome of the parents, the results indicated that the mitochondria of the hybrid tiger frog was consistent with a maternal inheritance. Phylogenetic analyses using concatenated nucleotide sequences of the 11 protein-coding genes with two different methods (maximum likelihood and MrBayes analysis) both highly supported a close relationship of the hybrid frogs with the Chinese tiger frog (=H. chinensis).

The complete mitochondrial genome of the hybrid was sequenced to be 16,621 bp in size following the female parent, M. terminalis. The genome contained 13 protein-coding genes, 22 transfer RNA genes, 2 ribosomal RNA genes, and 2 main non-coding regions (the control region and the origin of light strand replication). Sequence alignment between the mitochondrial genomes of the hybrid and its female parent showed that a total of 28 mutation sites were identified in 14 genes or regions. The genome information presented here may play an important role in further study on the genetic mechanisms of mitochondrial DNA in hybrids.

The genome sequence was 16 443 bp in size, and the gene order and contents were identical with those of congeneric species in the genus Takifugu. Comparing with the complete mitochondrial genome of its parents, the hybrid of T. fasciatus (♀) × T. flavidus (♂) was consistent with a maternal inheritance. All the initiation codons of protein-coding genes are ATG, except for COX1 that begins with GTG. The complete mitogenome of the hybrid of T. fasciatus (♀) × T. flavidus (♂) provides an important dataset for the exploration of mitochondrial inheritance mechanism.

The difference of total length is mainly caused by the difference of length in non-coding region. The nucleotide base composition is A = 29.15%, G = 15.62%, T = 26.91%, C = 28.32%, A + T = 56.06%, and C + G = 43.94%. The phylogenetic analysis using neighbour-joining (NJ) method showed that the hybrid offspring has a closer relationship to E. fuscoguttatus × E. lanceolatus.

Project description:In this work, we reported the complete mitochondrial DNA sequence of the hybrid of Leiocassis longirostris (♂)×Pelteobagrus fulvidraco (♀), which was obtained by artificial hybridization. The total length of the mitochondrial genome is 16,527 bp, with the base composition of 30.86% for A, 25.56% for T, 28.20% for C, and 15.38% for G. It contains two ribosomal RNA genes, 13 protein-coding genes, 22 transfer RNA genes, and a major non-coding control region (D-loop region). The arrangement of these genes is the same as that found in the teleosts. All the protein initiation codons are ATG, except for COX1 that begins with GTG. The complete mitogenome of the hybrid provides an important data set for the study in mitochondrial inheritance mechanism. Both the termination-associated sequence and critical central conserved sequences (CSB-D, CSB-E, and CSB-F) were also detected.
