Complete mitochondrial genome of near threatened butter Catfish Ompok bimaculatus (Siluriformes: Siluridae).

ABSTRACT: The complete mitochondrial genome of Ompok bimaculatus was obtained, using illumina high-throughput NextSeq 500 with 2 × 150 bp sequencing of mitochondrial DNA. The genome of O. bimaculatus was 16,482 bp in length (GenBank Accession No. KY887474) comprised of 13 protein-coding genes, 22 tRNA genes, 2 rRNA genes and a control region i.e. D-loop. In present mitogenome, 9 SSR were identified and validated in silico and secondary structures of all the 22 tRNA were predicted. The arrangement of genes was found identical to other siluriformes fish mitogenomes available in NCBI database. Phylogenetic relationship with closely related species were established which provide useful insights into taxonomic status of the species.

The structural organization and gene order were equivalent to other bony fishes. The complete genome of S. cochinchinensis would provide the basic dataset for further research on phylogenetics and conservation genetics of catfishes.

Complete mitochondrial DNA sequence of the Japanese endemic catfish Silurus lithophilus (Siluriformes: Siluridae).

Project description:The Japanese endemic catfish Silurus lithophilus is distributed only in Lake Biwa, Lake Yogo, and their river drainages. There are four species of the genus Silurus in Japan, of which S. lithophilus has a limited distribution. This catfish needs to be collected for DNA data owing to the lack of information related to its conservation. Here, the complete mitochondrial genome of the S. lithophilus from Lake Biwa in Japan was analyzed using next-generation sequencing. The mitochondrial genome of S. lithophilus was identified as a 16,524 bp circular molecule containing 13 protein-coding genes (PCGs), 22 tRNA genes, and 2 rRNA genes, along with one A + T-rich control region. The AT content was 55.89%. The heavy (H)-strand was predicted to have 12 PCGs and 15 tRNA, and 2 rRNA genes, whereas the light (L)-strand was predicted to contain one PCG and seven tRNA genes. The start codons ATG and GTG were found in 13 PCGs. The stop codons TAA, TAG, and AGA were observed in all PCGs, except CytB. All tRNA genes formed typical cloverleaf secondary structures. The molecular phylogenetic relationship, inferred using 13 PCGs (based on the maximum likelihood), was consistent with that reported in previous studies, which predicted a sister relationship between S. lithophilus and S. asotus. The results also clearly indicated that S. lithophilus is a valid species.

Project description:In this study, the whole mitochondrial genome of Silurus grahami was reported to be 16,518 bp in length, including 13 protein-coding genes, two ribosomal RNAs, 22 transfer RNAs, and one control region. The phylogenetic analysis based on 13 protein-coding genes showed that S. grahami was sister to clade of S. meridionalis and S. lanzhouensis. A total of 81 bases differences were identified in COI barcoding region, which could be used for species identification in catfish.

The heavy (H)-strand was predicted to have 12 PCGs and 14 tRNA and two rRNA genes, while the light (L)-strand was predicted to contain one PCGs and eight tRNA genes. The molecular phylogenetic analysis showed that S. asotus from Lake Biwa is genetically similar to S. asotus from China.

Base composition is 30.7% A, 26.4% T, 28.5% C, and 14.4% G, and 42.9% GC content. Two start codon (ATG and GTG) and seven stop codon (TAA, ACT, CCT, TTA, CAT, AAT, and TAG) patterns were found in protein-coding genes. Control region presented the highest A + T (64%) and lowest G + C content (35.7%) among all mitochondrial regions.

Project description:Silurus asotus is a widely distributed species, and mainly lives in the middle and lower layers of rivers, lakes, and reservoirs. In this study, we report the complete mitochondrial genome of S. asotus, which was 17,385 bp in length, containing 13 protein-coding genes, 2 rRNA genes, 22 tRNA genes, and a control region. Phylogenetic analyses indicated that S. asotus has a close relationship with S. soldatovi. This research work will provide some useful information on further molecular evolution studies of Siluridae species.

Complete mitochondrial genome of the Korean torrent catfish Liobagrus andersoni (Siluriformes, amblycipitidae).

Project description:The Korean torrent catfish (Liobagrus andersoni), an endemic species in Korea, is a member of the family Amblycipitidae. Herein, we report the first sequencing and assembly of the complete mitochondrial genome of L. andersoni. The complete mitochondrial genome is 16,514 bp long, consisting of 13 protein-coding genes, 22 tRNA genes, two rRNA genes, and a control region. It has the typical vertebrate mitochondrial gene arrangement. Phylogenetic analysis using mitochondrial genomes of 21 species showed that L. andersoni was clustered with L. marginatoides and L. nigricauda. This mitochondrial genome provides potentially important resources for addressing taxonomic issues and studying molecular evolution.

The complete mitochondrial DNA sequence of L. hyeongsanensis was sequenced by next-generation sequencing (NGS) analysis. The mitochondrial genome was assembled with 16,529 bp in length and encoded 13 protein-coding genes (PCGs), 22 tRNAs, two rRNAs, and one control region (D-loop). Also, the gene structures such as gene order and content were totally identical with the congeneric species. Molecular phylogenetic analysis determined the taxonomical position of L. hyeongsanensis in species level among the genus Liobagrus.
