The complete mitochondrial DNA of the Cuban gar (Atractosteus tristoechus).

ABSTRACT: The Cuban gar (Atractosteus tristoechus) is an endemic lepisosteid living in Cuba. Among gars, this species is one of the most threatened and has the smallest natural distribution range. Lepisosteids are air-breathing fishes belonging to the Holostean, a basal non-teleost clade of actinopterygians. Recent studies have indicated that these fishes could be a 'bridge between tetrapods and teleost biomedical models'. Herein, we sequenced and assembled the first complete mitochondrial genome of A. tristoechus. The total length of the mt genome is 16,290 bp, containing the typical 13 protein-coding genes, two ribosomal RNA (rRNA) genes, 22 transfer RNA (tRNA) genes, and a 537 bp length control region.

Project description:Lepisosteus platyrhincus is a member of the family Lepisosteidae living in the Western Hemisphere. It is a primitive air-breathing fish with the special intermediate position of phylogeny and between elasmobranchs and teleosts. Herein, we first sequenced and assembled the complete mitochondrial genome of Lepisosteus platyrhincus. The total length of mitochondrion is 16 320 bp with GC content of 42.43%, containing 13 protein-coding genes, two ribosomal RNA (rRNA) genes, 22 transfer RNA (tRNA) genes and a 564 bp control region. The accuracy of the fresh sequences was verified by phylogenetic analysis. This mitochondrial genome provides potentially important resources for addressing taxonomic issues and studying molecular evolution.

Complete mitochondrial DNA diversity in Iranians.

Project description:Due to its pivotal geographical location and proximity to transcontinental migratory routes, Iran has played a key role in subsequent migrations, both prehistoric and historic, between Africa, Asia and Europe. To shed light on the genetic structure of the Iranian population as well as on the expansion patterns and population movements which affected this region, the complete mitochondrial genomes of 352 Iranians were obtained. All Iranian populations studied here exhibit similarly high diversity values comparable to the other groups from the Caucasus, Anatolia and Europe. The results of AMOVA and MDS analyses did not associate any regional and/or linguistic group of populations in the Anatolia/Caucasus and Iran region pointing to close genetic positions of Persians and Qashqais to each other and to Armenians, and Azeris from Iran to Georgians. By reconstructing the complete mtDNA phylogeny of haplogroups R2, N3, U1, U3, U5a1g, U7, H13, HV2, HV12, M5a and C5c we have found a previously unexplored genetic connection between the studied Iranian populations and the Arabian Peninsula, India, Near East and Europe, likely the result of both ancient and recent gene flow. Our results for Persians and Qashqais point to a continuous increase of the population sizes from ∼24 kya to the present, although the phase between 14-24 kya is thought to be hyperarid according to the Gulf Oasis model. Since this would have affected hunter-gatherer ranges and mobility patterns and forced them to increasingly rely on coastal resources, this transition can explain the human expansion across the Persian Gulf region.

Complete mitochondrial DNA genome sequences from the first New Zealanders.

Project description:The dispersal of modern humans across the globe began ~65,000 y ago when people first left Africa and culminated with the settlement of East Polynesia, which occurred in the last 1,000 y. With the arrival of Polynesian canoes only 750 y ago, Aotearoa/New Zealand became the last major landmass to be permanently settled by humans. We present here complete mitochondrial genome sequences of the likely founding population of Aotearoa/New Zealand recovered from the archaeological site of Wairau Bar. These data represent complete mitochondrial genome sequences from ancient Polynesian voyagers and provide insights into the genetic diversity of human populations in the Pacific at the time of the settlement of East Polynesia.

The MRE11 GAR motif regulates DNA double-strand break processing and ATR activation.

Project description:The MRE11/RAD50/NBS1 complex is the primary sensor rapidly recruited to DNA double-strand breaks (DSBs). MRE11 is known to be arginine methylated by PRMT1 within its glycine-arginine-rich (GAR) motif. In this study, we report a mouse knock-in allele of Mre11 that substitutes the arginines with lysines in the GAR motif and generates the MRE11(RK) protein devoid of methylated arginines. The Mre11(RK/RK) mice were hypersensitive to ?-irradiation (IR) and the cells from these mice displayed cell cycle checkpoint defects and chromosome instability. Moreover, the Mre11(RK/RK) MEFs exhibited ATR/CHK1 signaling defects and impairment in the recruitment of RPA and RAD51 to the damaged sites. The M(RK)RN complex formed and localized to the sites of DNA damage and normally activated the ATM pathway in response to IR. The M(RK)RN complex exhibited exonuclease and DNA-binding defects in vitro responsible for the impaired DNA end resection and ATR activation observed in vivo in response to IR. Our findings provide genetic evidence for the critical role of the MRE11 GAR motif in DSB repair, and demonstrate a mechanistic link between post-translational modifications at the MRE11 GAR motif and DSB processing, as well as the ATR/CHK1 checkpoint signaling.

The result of phylogenetic analysis demonstrates that P. changtingensis is close to that of P. fasciatus.

The mitochondrial genome of B. crocodilinus is of 17,818 bp in length, with overall base composition of 32.11% A, 24.69% T, 16.22% G, and 26.98% C. The genome content includes 13 protein-coding genes, 22 tRNA genes, 2 rRNA genes, and 2 control regions. The result of phylogenetic analysis indicates that B. crocodilinus mitogenome is close to that of Ophisurus macrorhynchos, which are nested within the family Ophichthidae.

Complete sequence of mitochondrial DNA of Gracilaria tenuistipitata (Rhodophyta).

Project description:We sequenced and analysed the complete mitogenome of Gracilaria tenuistipitata. The complete G. tenuistipitata mitogenome is 25,819 bp long, containing 50 genes, including 24 protein-coding genes, one intron, two rRNA genes, 23 tRNA genes, and one unidentified open reading frame. Twenty-three of the 24 (95.83%) protein-coding genes ended with the TAA stop codon, and one (4.17%) with TAG (rps3 gene). All protein-coding genes in G. tenuistipitata were found to contain the start codon ATG. Phylogenetic analysis revealed that G. tenuistipitata clustered with G. chilensis. The complete mitochondrial genome sequence provided here should be useful for elucidation of Gracilaria evolution.
