The complete mitochondrial genome of Hydrotaea (Ophyra) chalcogaster (Diptera: Muscidae).

ABSTRACT: Hydrotaea (Ophyra) chalcogaster (Diptera: Muscidae) is a significant flesh fly in forensic entomology. In this study, the complete mitochondria gene (mitogenome) of H. chalcogaster was sequenced and annotated for the first time, and the full-length was a 15,279 bp fragment, consisting of A (39.6%), G (9.1%), T (39,0%), and C (12.3%), which is the classical structure for insect mitogenome. Phylogenetic analyses showed that H. chalcogaster clearly separated from the Muscinae subfamily. This work provides support for further study of the use of mitochondrial genome in the species identification, and enriches the databases of the Muscidae species.

Project description:Graphomya rufitibia (Diptera: Muscidae) was distributed worldwide. In this study, the complete mitogenome of G. rufitibia was sequenced and annotated, and the full-length was a 15,374 bp fragment, consisting of A (40.64%), G (8.96%), T (36.80%), and C (13.59%). The mitogenome is composed of 13 protein-coding genes, 22 transfer RNA genes, two ribosomal RNA genes, and a non-coding AT-rich region. Most PCGs used the canonical putative codon to start and stop, 21 tRNAs are folded into the classic clover-leaf structure, with the one exception of tRNA-Ser (AGN). A total length of 11,182 bp encoding 3727 amino acids, and account for 72.7% of the whole mitogenome. Phylogenetic analyses showed that G. rufitibia belongs to family Mydaeinae.

The complete mitochondrial genome of Muscina pascuorum (Diptera: Muscidae).

Project description:Muscina pascuorum (Diptera: Muscidae) represents an important hygiene pest. The complete mitochondrial genome (mitogenome) of M. pascuorum was first sequenced and annotated in this study. The full length of mitogenome was 14, 940 bp, consisting of 13 protein-coding genes (PCGs), two ribosomal RNA (rRNA), 22 transfer RNA (tRNA), and one AT-rich region. The nucleotide content of these flies was 40.0% A, 13.2% C, 9.1% G, and 37.6% T. This study illustrates that the arrangement of the genes was identical to classical metazoans. Besides, the phylogenetic analyses indicated that the branch of M. pascuorum was clustered separately from the common three Muscina spp in the tree. This genome provides an essential reference for understanding the phylogenetic relationships of Muscidae.

The complete mitochondrial genome of Fannia scalaris (Diptera: Muscidae).

Project description:Fannia scalaris (Fabricius, 1794) is closely related to human life in ecological habits, which can lead to health concerns since they feed on various contamination sources. In this study, we first present the mitochondrial genome (mitogenome) of F. scalaris (GenBank No. MT017706). The length of this mitogenome was composed of 15,040 base pairs, including 13 protein-coding genes, two ribosomal RNA, 22 transfer RNA, and an AT-rich region. It consisted of A 39.3%, G 9.1%, C 13.0%, and T 38.6%. The arrangement of the genes was consistent with that of the ancestral metazoan. Furthermore, phylogenetic relationship indicated that F. scalaris was obviously separated from the muscid flies. This study provides useful genetic data in order to further understand the evolutionary relationship of the Muscidae.

The complete mitochondrial genome of Neomyia cornicina (Diptera: Muscidae).

Project description:Neomyia cornicina (Fabricius, 1781) (Diptera: Muscidae) is considered to be an important dung-degrading species in Japan. In this study, we report the first mitochondrial genome (mitogenome) of N. cornicina. The complete mitogenome of N. cornicina was 17,254 bp in length (GenBank accession No. MW592695), containing 13 protein-coding genes (PCGs), 22 transfer RNA (tRNA) genes, 2 ribosomal RNA (rRNA) genes, and a non-coding AT-rich region. Its nucleotide composition was A (41.0%), G (8.4%), C (11.8%), and T (38.8%). Phylogenetic analysis indicated that N. cornicina is closely related to the species of Eudasyphora canadiana. This mitogenome contributes useful information for further understanding of the phylogenetic relationship and species identification within Muscidae species.

The complete mitochondrial genome of the bazaar fly, Musca sorbens Wiedemann (Diptera: Muscidae).

Project description:The bazaar fly, Musca sorbens (Diptera: Muscidae) Wiedemann, 1830 is a world-wide species with sanitary, medical, and veterinary importance. The complete mitochondrial genome of M. sorbens is sequenced to better understand the mitogenomic characteristics and phylogeny of this species. The circular mitogenome is 16,120 bp in length, contains 13 protein-coding genes (PCGs), 22 tRNA genes (tRNAs), two rRNA genes, and an AT-rich control region. The mitogenome comprises an A + T content of 77.4%. All PCGs start with "ATN" codons except for COI which starts with TCG, and terminate with the common stop codons TNN. A phylogenetic tree, including six Muscidae species, is reconstructed based on the whole mitogenome sequences. The interspecific distances of mitogenomes between the six Muscidae species range from 0.059 to 0.168.

Sequencing and analyzing complete mitochondrial DNA of Muscina angustifrons (Insecta, Diptera, Muscidae).

Project description:In this study, the complete mitochondrial genome sequenced and analyzed from a fly, Muscina angustifrons which collected from South Korea. The size of mitochondrial genome is 16,316 bp with 40.9% A, 12.3% C, 8.4% G and 38.4% T distribution. Furthermore, phylogenetic relationships of the superfamily Muscoidea evaluated due to mitochondrial protein coding genes. The results showed that the family Muscidae is a paraphyletic group and the closest species to M. angustifrons is M. levida. This is the third complete mitochondrial genome for the genus Muscina and the first genus record from South Korea.

Phylogeny and Evolutionary Timescale of Muscidae (Diptera: Calyptratae) Inferred from Mitochondrial Genomes.

Project description:House flies (Muscidae) comprise the most species-rich family of the muscoid grade with over 5000 described species worldwide, and they are abundant in various terrestrial and aquatic ecosystems. The high number of species, varied appearances, complex feeding habits, and wide distributions have hindered researchers from understanding their phylogeny and evolutionary history. Here, we newly sequenced fifteen mitochondrial genomes and reconstructed the phylogenetic relationships and divergence time among eight subfamilies of Muscidae (Diptera). The best phylogenetic tree, which was inferred by IQ-Tree, recovered the monophyly for seven out of eight subfamilies (except for Mydaeinae). Based on phylogenetic analyses and morphological characteristics, we prefer the subfamily status of Azeliinae and Reinwardtiinae, and separate Stomoxyinae from Muscinae. Genus Helina Robineau-Desvoidy, 1830 was synonymized with Phaonia Robineau-Desvoidy, 1830. The divergence time estimation indicated Muscidae originated at 51.59 Ma (early Eocene). Most subfamilies had originated around 41 Ma. We provided a mtgenomic viewpoint on the phylogenetic relationships and divergence time estimation of Muscidae.

Project description:The complete mitochondrial genome of Limnophyes minimus (Meigen 1818) was sequenced and annotated, and its general features and base composition were analyzed. The phylogenetic relationships of the families Chironomidae, Simuliidae, Sciaridae and Culicidae based on 25 metagenomes were reconstructed using maximum likelihood (ML) methods based on the concatenated nucleotide sequences, the phylogenetic analysis showed that L. minimus belongs family Chironomidae, which is consistent with the traditional morphological classification.

The complete mitochondrial genome of Sarcophaga brevicornis (Diptera: Sarcophagidae).

The complete mitochondrial genome (mitogenome) of S. brevicornis was first sequenced and assembled in this study. The length of circular mitogenome was 15,152 bp, which showed in a typical arthropod genome, including 13 protein-coding genes (13 PCGs), two ribosomal RNA (two rRNA) genes, 22 transfer RNA (22 tRNA) genes, and an AT-rich region. Its nucleotide composition was A 39.0%, C 12.7%, G 10.6%, and T 37.7%. Furthermore, phylogenetic relationships of S. brevicornis and the published sarcophagid species were evaluated based on 13 PCGs. The results indicated that S. brevicornis was clearly separated from the other sarcophagid species, but it was closed to the species of Sarcophaga similis. This study provided a significant database reference for genetic structure and phylogenetic analysis of Sarcophagidae.

Project description:The mitochondrial genome of Silba sp. was sequenced and annotated as the first representative of family Lonchaeidae. The complete mitochondrial genome of Silba sp. is 16,008 bp totally, consisting of 13 protein-coding genes, two rRNAs, and 22 transfer RNAs, which has a similar gene structure with other published species of Diptera. The nucleotide composition biases toward A and T is 77.2% of the entirety. All PCGs start with ATN codons, except COI and ND1, and end with TAA or incomplete stop codon. Phylogenetic analyses based on 11 Diptera species supported the monophyly of superfamily Tephritoidea.
