Complete mitochondrial genome of a leaf-mining beetle, Podagricomela nigricollis (Coleoptera: Chrysomelidae).

ABSTRACT: Podagricomela nigricollis is a citrus pest that distributes in South China. Currently, there was no complete mitochondrial genome of Podagricomela species available in GenBank. Here, we reported the complete circular mitogenome of P. nigricollis. It had a total length of 16,756 bp, including 13 protein-coding genes (PCGs), 22 tRNA genes, two rRNA genes, and one A + T-rich region. Among the 13 PCGs, only four (NAD5, NAD4, NAD4l, NAD1) located on the L-strand, whereas the other nine (NAD2, COX1, COX2, ATP8, ATP6, COX3, NAD3, NAD6, COB) located on the H-strand. Phylogenetic analysis using nucleotide sequences of the 13 PCGs indicated that P. nigricollis were clustered with six Galerucinae species, which was consistent with previous morphological classification.

Project description:The complete circular mitochondrial genome of Agonita chinensis was 16,395 bp in length, which contained two ribosomal RNA genes, 22 transfer RNAs, 13 protein-coding genes (PCGs) and one non-coding AT-rich region with the length of 2001 bp. All of the 22 tRNA genes displayed a typical clover-leaf structure, with the exception of tRNASer (TCT). Twelve PCGs were initiated by ATN codons, except that nad1 started with TTG. Only four PCGs used the typical stop codon 'TAA' and 'TGA', while nine PCGs terminated with incomplete stop codons (TA or T). Phylogenetic analysis based on 13 PCGs of Chrysomelidae mitogenomes showed that A. chinensis was closely related to Cassida viridis.

Project description:The complete circular mitochondrial genome (mitogenome) of Rhadinosa nigrocyanea was 17,965 bp in length, which contained 2 ribosomal RNA genes, 22 transfer RNAs, 13 protein-coding genes (PCGs) and 1 non-coding AT-rich region with the length of 3002 bp. All of the 22 tRNA genes displayed a typical clover-leaf structure, with the exception of tRNASer (TCT). 12 PCGs were initiated by ATN codons, except for ND1 started with TTG. Only six PCGs used the typical stop codon 'TAA' and 'TGA', while seven PCGs terminated with incomplete stop codons (TA or T). Phylogenetic analysis showed that R. nigrocyanea grouped with Cassidinae species, sister to Clytrinae + Cryptocephalinae.

All 22 tRNA genes displayed a typical clover-leaf structure except for tRNASer (AGN). All 13 PCGs initiated with ATN codons. Phylogenetic analysis based on 13 PCGs of Chrysomelidae mitogenomes showed that C. bowringi was closely related to Agonita chinensis and Rhadinosa nigrocyanea.

Mitochondrial genome of a leaf-mining beetle Prionispa champaka Maulik (Coleoptera: Chrysomelidae: Cassidinae).

Project description:Prionispa champaka is a leaf-mining species which feeds on Pollia spp. and widely distributes in southern China. The complete mitogenomic sequence of P. champaka (Chrysomelidae: Cassidinae) was obtained and annotated, with a length of 20,494 bp. It was longer than those of other Chrysomelid species (not including Bruchinae) because of its much longer non-coding sequences. Gene arrangement and content of P. champaka was identical to the most common type in insects, and it was also biased toward AT (accounting for 78.4%). Phylogenetic analysis based on mitochondrial PCGs indicated that P. champaka was closely clustered with 5 other Cassidinae species, supporting the traditional morphological classification within Cassidinae.

Complete mitochondrial genome of coconut hispine beetle Brontispa longissima (Coleoptera: Chrysomelidae: Cassidinae).

Project description:In the present study, we determined complete mitogenome of Brontispa longissima, the first for the tribe Cryptonychini. This mitochondrial genome contains 15,696 bp, with an A+T content of 72.2% (GenBank accession no. MN052901). All of the 22 tRNA genes displayed a typical clover-leaf structure, with the exception of tRNASer (TCT). 12 PCGs were initiated by ATN codons, and ND1 started with TTG. Eight PCGs used the typical stop codon 'TAA' and 'TGA', while five PCGs terminated with incomplete stop codons "T". Phylogenetic analyses performed using the mitogenomes for the B. longissima and other seven species from subfamily Cassidinae.

Midgut bacterial diversity of a leaf-mining beetle, Dactylispa xanthospila (Gestro) (Coleoptera: Chrysomelidae: Cassidinae).

Project description:Microorganisms play an essential role in the growth and development of numerous insect species. In this study, the total DNA from the midgut of adults of Dactylispa xanthospila were isolated and bacterial 16S rRNA sequenced using the high-throughput Illumina MiSeq platform. Then, the composition and diversity of the midgut bacterial community were analysed with QIIME2. The results showed the midgut bacteria of D. xanthospila belong to 30 phyla, 64 classes, 135 orders, 207 families and 369 genera. At the phylum level, Proteobacteria, Bacteroidetes and Firmicutes were the dominant bacteria, accounting for 91.95%, 3.44% and 2.53%, respectively. The top five families are Enterobacteriaceae (69.51%), Caulobacteraceae (5.24%), Rhizobiaceae (4.61%), Sphingomonadaceae (4.23%) and Comamonadaceae (2.67%). The bacterial community's primary functions are carbohydrate metabolism, amino acid metabolism and cofactor and vitamin metabolism, which are important for the nutritional requirements of plant-feeding insects.

Chromosome-level genome assembly of Monolepta hieroglyphica, two-spotted leaf beetle (Coleoptera: Chrysomelidae).

Project description:Monolepta hieroglyphica, in view of its wide-ranging host and highly polyphagous characteristics, has become an important agricultural pest in East and Southeast Asian countries. To better understand its biology and develop control strategies, we present a high-quality chromosome-level genome assembly of M. hieroglyphica, with contig N50 of 18.62 Mb and scaffold N50 of 241.916 Mb. Using 156x Hi-C data, we anchored a total length of 2,313 Mb (91.9% of the genome) to nine chromosomes. We predicted 46,561 genes using an integrated approach. The completeness of the final reported genome and proteome were assessed by BUSCO and show high values of 99.4% and 95.2%, respectively. The high-quality chromosome-level genome assembly of M. hieroglyphica will be a useful molecular resource for the communities of both beetle and crop protection.

Characterization of the Complete Mitochondrial Genome of a Flea Beetle Luperomorpha xanthodera (Coleoptera: Chrysomelidae: Galerucinae) and Phylogenetic Analysis.

Project description:In this study, the mitochondrial genome of Luperomorpha xanthodera was assembled and annotated, which is a circular DNA molecule including 13 protein-coding genes (PCGs), 22 transfer RNA (tRNA) genes, 2 ribosomal RNA genes (12S rRNA and 16S rRNA), and 1388 bp non-coding regions (A + T rich region), measuring 16,021 bp in length. The nucleotide composition of the mitochondrial genome is 41.3% adenine (A), 38.7% thymine (T), 8.4% guanine (G), and 11.6% cytosine (C). Most of the protein-coding genes presented a typical ATN start codon (ATA, ATT, ATC, ATG), except for ND1, which showed the start codon TTG. Three-quarters of the protein-coding genes showed the complete stop codon TAR (TAA, TAG), except the genes COI, COII, ND4, and ND5, which showed incomplete stop codons (T- or TA-). All the tRNA genes have the typical clover-leaf structure, except tRNASer1 (AGN), which has a missing dihydrouridine arm (DHU). The phylogenetic results determined by both maximum likelihood and Bayesian inference methods consistently supported the monophyly of the subfamily Galerucinae and revealed that the subtribe Luperina and genus Monolepta are polyphyletic groups. Meanwhile, the classification status of the genus Luperomorpha is controversial.

Underwater Attachment of the Water-Lily Leaf Beetle Galerucella nymphaeae (Coleoptera, Chrysomelidae).

Project description:While the reversible attachment of artificial structures underwater has moved into the focus of many recent publications, the ability of organisms to walk on and attach to surfaces underwater remains almost unstudied. Here, we describe the behaviour of the water-lily leaf beetle Galerucella nymphaeae when it adheres to surfaces underwater and compare its attachment properties on hydrophilic and hydrophobic surfaces underwater and in the air. The beetles remained attached to horizontal leaves underwater for a few minutes and then detached. When the leaf was inclined, the beetles started to move upward immediately. There was no difference in the size of the tarsal air bubble visible beneath the beetles' tarsi underwater, between a hydrophilic (54° contact angle of water) and a hydrophobic (99°) surface. The beetles gained the highest traction forces on a hydrophilic surface in the air, the lowest on a hydrophobic surface in air, and intermediate traction on both surfaces underwater. The forces measured on both surfaces underwater did not differ significantly. We discuss factors responsible for the observed effects and conclude that capillary forces on the tarsal air bubble might play a major role in the adhesion to the studied surfaces.

The complete mitochondrial genome of Downesia tarsata (Coleoptera: Chrysomelidae: Cassidinae).

Project description:Downesia species are leaf-mining beetles mainly feed on Poaceae plants in the tropical and subtropical areas in Asia. In this study, we firstly sequenced and reported the complete mitochondrial genome for the genus. The complete mitogenome of Downesia tarsata is 18,557 bp in length, including 13 protein-coding genes (PCG), 22 transfer RNA (tRNA), two ribosomal RNA (rRNA), and one AT-rich region. Phylogenomic analysis indicated that D. tarsata is closely related to Agonita chinensis, and the two species belong to the same tribe of Gonophorini. The complete mitochondrial genome of D. tarsata could help clarify the phylogenetic relationship among Cassidinae species.
