The complete mitogenome of ghost jellyfish Cyanea nozakii (Cnidaria, Semaeostomeae, Cyaneidae).

ABSTRACT: The cnidarian jellyfishes are impressive organisms to show animal mitochondrial genomic diversities. Their mitogenome structure is linear and tRNA content has one or two in numbers, which is highly different than other metazoans. In this study, a complete mitogenome of the ghost jellyfish Cyanea nozakii (Cnidaria, Semaeostomeae, Cyaneidae) was sequenced and analyzed. The mitgenome is 17,381 bp long with 38.5% A, 16.0% C, 13.9% G, and 31.6% T nucleotide distributions. In addition, phylogenetic relationship of C. nozakii in the class Scyphozoa was investigated by using mitochondrial protein coding genes. Due to results, C. nozakii was positioned in the paraphyletic order Semaeostomeae. This is the first complete mitogenome from the genus Cyanea.

Project description:In this study, the complete mitogenome sequence of Galaxy Coral, Galaxea fascicularis (Cnidaria: Oculinidae), has been sequenced by next-generation sequencing method. The overall base composition of Galaxea fascicularis mitogenome is 24.9% for A, 14.0% for C, 24.3% for G and 36.8% for T and has low GC content of 38.3%. The assembled mitogenome, consisting of 18 751 bp, has unique 13 protein-coding genes (PCGs), three transfer RNAs and two ribosomal RNAs genes. The G. fascicularis mitogenome has the common mitogenome gene organization and feature of scleractinian coral. Among 13 PCGs, ND5 gene is interrupted by group I intron (12 022 bp). There are 12 genes embedded in ND5 group I intron (tRNA-Leu, ND1, CYTB, ND2, ND6, ATP6, ND4, 12S rRNA, COX3, COX2, ND4L and ND3). The complete mitogenome provides essential and important DNA molecular data for further phylogenetic and evolutionary analysis for stony coral.

Evaluation of the complete nuclear rDNA unit sequence of the jellyfish Cyanea nozakii Kishinouye (Scyphozoa: Semaeostomeae) for molecular discrimination.

Project description:The harmful jellyfish Cyanea nozakii Kishinouye has frequently occurred on Korean coasts, and its blooms have caused serious ecological and economic damages. DNA sequences of the C. nozakii for molecular detection and discrimination are relatively scarce. In this study, we determined the complete sequence of a single unit of tandemly repeated ribosomal DNA (rDNA) of the Korean C. nozakii and characterized the molecular features of the rDNA. The complete rDNA contained 8,003 bp (48.4% GC) with the same gene arrangement (18S, ITS1, 5.8S, ITS2, 28S, and IGS) to the typical eukaryotes. Dot plot analysis showed that the coding regions (18S, 5.8S, and 28S) were highly conserved, while the non-coding regions (ITS1, ITS2, and IGS) were more variable and parsimony-informative. The IGS contained a putative transcription termination signal (poly(T) tract) and four repeats of block minisatellites. Phylogenetic analyses using 18S and 28S rDNA revealed well-resolved relationships of C. nozakii within the order Semaeostomeae, separating it from other Cyanea species. The complete rDNA sequence provides various options for the selection of jellyfish taxonomic markers and may be useful for discriminating between species of C. nozakii and phylogeny reconstruction with close relatives.

Next-generation sequencing yields the complete mitogenome of massive coral, Porites lutea (Cnidaria: Poritidae).

Project description:In this study, the complete mitogenome sequence of massive coral, Porites lutea (Cnidaria: Poritidae), has been sequenced by next-generation sequencing method. The overall base composition of Porites lutea mitogenome is 26.0% for A, 13.3% for C, 23.0% for G and 37.8% for T and have high AT content of 63.7%. The assembled mitogenome, consisting of 18 646 bp, has unique 13 protein-coding genes (PCGs), seven transfer RNAs and two ribosomal RNAs genes. The Porites lutea mitogenome has the common mitogenome gene organization and feature of scleractinian coral. Among 13 PCGs, ND5 and COX1 genes are interrupted by group I intron (11 130 and 971 bp, respectively). There are 13 genes embedded in ND5 group I intron (tRNA-Glu, ND1, CYTB, tRNA-Met, ND2, ND6, ATP6, ND4, 12S rRNA, COX3, COX2, ND4L and ND3), and two genes embedded in COX1 group I intron (tRNA-Ile and tRNA-Pro). The complete mitogenome provides essential and important DNA molecular data for further phylogenetic and evolutionary analysis for stony coral.

Jellyfish Cyanea capillata nematocyst venoms LC-MSMS

Project description:The scyphozoan jellyfish Cyanea capillata are common blooming species in China.The venomous characteristics of jellyfish Cyanea capillata is largely attributed to their complex and elaborate venom delivery system: the stinging cells, or nematocytes. These specialized cells synthesize and secrete unique intracellular organelles called nematocysts. Nematocysts with heterogeneous sizes and morphologies may contain different toxic components that exert diverse pharmacological and physiological activities such as hemolysis, cytolysis and proteolysis for defense and prey capture. We modified the previous method of nematocyst preparation using density centrifugation to purify undischarged nematocysts from jellyfish Cyanea capillata tentacles. Ultimately, mastigophore-type nematocysts were successfully purified.Then we used purified nematocysts for proteomic study to identify sting-related toxins in Cyanea capillata nematocysts.

The complete mitochondrial genome of the hydrozoan jellyfish Spirocodon saltatrix (Cnidaria; Hydrozoa; Anthoathecata) with phylogeny analysis.

Project description:In the present study, we sequenced and analyzed the complete mitochondrial genome of the hydrozoan jellyfish Spirocodon saltatrix. The mitochondrial genome was a linear form (15,752 bp long, 70.4% AT), consisting of 13 protein coding genes (cox1, cox2, atp8, atp6, cox3, nad2, nad5, nad6, nad3, nad4L, nad1, nad4, and cytB), two tRNAs (tRNA-Met and tRNA-Trp), and two rRNAs (12S and 16S). Mitochondrial gene arrangement of the S. saltatrix was completely identical to already-known mitochondrial genomes of hydrozoans. Molecular phylogenetic analysis using 13 protein-coding genes showed that S. saltatrix was closely related to the hydrozoan Clava multicornis.

Preparation and Neutralization Efficacy of Novel Jellyfish Antivenoms against Cyanea nozakii Toxins.

Project description:Jellyfish stings are a common issue globally, particularly in coastal areas in the summer. Victims can suffer pain, itching, swelling, shock, and even death. Usually, hot water, vinegar, or alumen is used to treat the normal symptoms of a jellyfish sting. However, a specific antivenom may be an effective treatment to deal with severe jellyfish stings. Cyanea nozakii often reach a diameter of 60 cm and are responsible for hundreds of thousands of stings per year in coastal Chinese waters. However, there has been no specific C. nozakii antivenom until now, and so the development of this antivenom is very important. Herein, we collected C. nozakii antisera from tentacle extract venom immunized rabbits and purified the immunoglobulin (IgG) fraction antivenom (AntiCnTXs). Subsequently, two complete procedures to produce a refined F(ab')2 type of antivenom (F(ab')2-AntiCnTXs) and Fab type of antivenom (Fab-AntiCnTXs) by multiple optimizations and purification were established. The neutralization efficacy of these three types of antivenoms was compared and analyzed in vitro and in vivo, and the results showed that all types of antibodies displayed some neutralization effect on the lethality of C. nozakii venom toxins, with the neutralization efficacy as follows: F(ab')2-AntiCnTXs ≥ AntiCnTXs > Fab-AntiCnTXs. This study describes the preparation of novel C. nozakii jellyfish antivenom preparations towards the goal of developing a new, effective treatment for jellyfish stings.

Project description:In this study, we analyzed the complete mitochondrial genome of the hydrozoan jellyfish Blackfordia virginica. The genome was a linear form (15,109 bp long, 73.6% AT), including 13 protein-coding genes (cox2, atp8, atp6, cox3, nad2, nad5, nad6, nad3, nad4L, nad1, nad4, cytB, and cox1), 2 tRNAs (tRNA-Met and tRNA-Trp), and 2 rRNAs (12S and 16S RNA). The genome structure of the B. virginica was completely identical to mitochondrial genomes of other hydrozoans that belonged to Leptothecata and Anthoathecata. Molecular phylogenetic analysis within hydrozoan species showed that B. virginica was the closest to the hydrozoan Laomedea flexuosa.

Project description:In this study, we sequenced and analyzed the complete mitochondrial genome of Cladonema multiramosum Zhou et al., 2022 from Fujian, China. The length of the linear mitochondrial genome is 15164 bp, containing 13 protein-coding genes (cox2, atp8, atp6, cox3, nad2, nad5, nad6, nad3, nad4L, nad1, nad4, cob, cox1), two tRNAs (trnM and trnW) and 2 rRNAs (12S and 16S). The arrangement of mitogenomes show some little differences in different hydrozoan groups. The phylogenetic analysis of 13 protein coding genes (PCGs) in Cnidarians showed that C. multiramosum was closely related to Cladonema pacificum.

The mitogenome is a circular molecule 17,024 bp in length, including 13 protein-coding genes (including Cox 1, Cox2, Atp 8, Atp 6, ND5, ND 6, ND3, ND41, ND1,ND4, Cytb), 6 tRNAs (tRNA-Trp, tRNA-Met, tRNA-Val, tRNA-Arg, tRNA-Glu, tRNA-Asn), 2 rRNA genes (small subunit RNA and large subunit RNA), and 1 putative control region. The phylogenetic tree in the related species showed that giant jellyfish is close to Hydraoligaclis.

The mtgenome was a complete linear form (15,047 bp in length, 30.9% A, 42.1% T, 12.5% C, and 14.5% G), including 13 protein coding genes (PCGs) (cox1, cox2, cox3, atp6, atp8, nad1, nad2, nad3, nad4, nad4L, nad5, nad6, and cytb), 2 tRNAs (tRNAMet and tRNATrp), and 2 rRNAs (12S and 16S rRNA). The genome structure of the T. lata was completely identical to those of other species within the subclass Hydroidolina. In addition, our molecular phylogenetic analysis using 13 PCGs within hydrozoans showed that T. lata was the closest to Turritopsis dohrnii.
