Complete mitochondrial genome of a cave dwelling Desmopachria (Insecta: Coleoptera: Dytiscidae) from the Eastern Amazon.

ABSTRACT: Coleoptera presents most of the cave fauna biodiversity, with several troglobite species belonging to the aquatic family Dytiscidae. However, very little is known on both genetic and genomic diversity traits of Neotropical cave beetles. Thus, here we present the complete mitochondrial genome sequence of five specimens of Desmopachria collected in a ferruginous cave from Serra dos Carajás in Parauapebas (Pará, Brazil, Eastern Amazon). Besides the general characteristics of the mitogenome of the analyzed specimens, we present their phylogenetic position within the family, considering the available genome sequences of different subfamilies within Dytiscidae.

The complete mitochondrial genome of the critically endangered diving beetle Dytiscus sharpi (Coleoptera: Dytiscidae).

Project description:The diving beetles Dytiscus sharpi is one of the most critically endangered species in Japan, caused by excessive capture and destruction of their habitats. Here, the complete mitochondrial genome sequence (mitogenome) of D. sharpi is described. The entire mitogenome sequence was 19,476 bp long, containing 13 protein-coding genes, 2 ribosomal RNA genes, 22 transfer RNA genes, and 1 control region. The average A + T content of mitogenome was 81.3%. This is the first description of a complete mitochondrial genome of Dytiscus, and the second of Dytiscinae. The primary genetic data obtained in this study is expected to contribute to conservation genetic studies using various genetic analysis methods, including environmental DNA.

Project description:The flightless intertidal beetle genus Aegialites (family Salpingidae) is distributed along the Northern Pacific coasts, from California to Alaska and from Northern Japan to Kamchatka. Systematics of Aegialites and its phylogenetic relationships to other members of Salpingidae are unclear, and little genetic information is available. We here present the first complete mitochondrial genome of this genus, represented by Aegialites californicus (Motchoulsky, 1845) from Sonoma County, California, U.S.A. The complete mitochondrial genome of A. californicus is 15,899 bp long and comprises 13 protein-coding (PCG), two ribosomal RNA (rRNA) and 22 transfer RNA (tRNA) genes. The phylogenetic analysis places A.californicus as sister to other members of family Salpingidae. The mitochondrial genome sequence of A. californicus will contribute to future phylogenetic and taxonomic studies of genus Aegialites, family Salpingidae and superfamily Tenebrionoidea.

Complete mitochondrial genome of the dark mealworm Tenebrio obscurus Fabricius (Insecta: Coleoptera: Tenebrionidae).

Project description:The dark meal worm Tenehrio obscurus (Fabricius) is frequently found as a pest of stored grain, but it is an important resource insect like the yellow meal worm (Tenebrio molitor), larvae of which has high-protein content. The mitochondrial genome of T. obscurus is a circular molecule of 15,771 bp (GenBank accession number MG739327), with A + T bias of 72.69%. It comprises 13 protein-coding, 22 tRNA and two rDNA genes. The protein-coding genes had typical ATN (Met) as the initiation codon and were terminated by the typical stop codon TAN, except for COX2, COX3, nadh5 and nadh4. The phylogenetic tree based on maximum-likelihood method showed that the phylogenetic position of T. obscurus is closely related to T. molitor.

The complete mitochondrial genome of a cave-dwelling loach Triplophysa baotianensis (Teleostei: Nemacheilidae).

Project description:Triplophysa baotianensis belong to the genus Triplophysa (Teleostei, Nemacheilidae), endemic to Guizhou Province, Southwestern China. In this study, the complete mitochondrial genome of T. baotianensis was sequenced and reported for the first time. The circular mitogenome was 16,576 bp in length and consisted of 13 protein-coding genes, 2 ribosomal RNA (rRNA) genes, 22 transfer RNA (tRNA) genes, and 1 non-coding control region. The overall base composition was 30.79% A, 27.62% T, 25.46% C, and 16.13% G with 41.59% GC content. Phylogenetic analysis using mitochondrial genomes of 40 species showed that all Triplophysa species clustered as one monophyletic clade, and T. baotianensis was the closest to (T. nasobarbatula + (T. rosa + T. xiangxiensis)).

Complete mitochondrial genome of the diving beetle, Cybister brevis Aube, 1838 (Coleoptera, Dytiscidae) from Jeju Island.

Project description:The mitochondrial genome of Cybister brevis Aubé, 1838, is 16,198 bp long and includes 37 genes that are highly conserved in the family Dytiscidae. Phylogenetic analysis revealed that all genera in Dytiscidae, except Hydroporus and Oreodytes, are monophyletic. The Hydroporini tribe was found to be polyphyletic and closely associated with the Hygrotini, Bidessini, and Hyphydrini tribes. Dytiscinae was found to be a highly divergent polyphyletic group comprising three distinct clades. This study also revealed that C. brevis is closely related to Cybister japonicus and that the tribe Cybistrini diverged relatively early compared to other tribes in Hydroporinae. These findings are consistent with those of previous studies and provide new insights into the phylogeny of the Dytiscidae family, which has previously shown discrepancies between morphological characteristics and molecular data. The genome-level analyses conducted in this study serve as a valuable foundation for future investigations into the Dysticidae evolutionary history.

Complete mitochondrial genome and phylogenetic analysis of Mastax latefasciata Liebke 1931 (Insecta: Coleoptera: Carabidae).

Specimens of adult Mastax latefasciata Liebke, 1931 were collected from Yājì Hill, Huáihuà City, Húnán Province, China. The complete mitochondrial genome (GenBank accession number ON674050.1) of M. latefasciata was sequenced, annotated, and characterized. The results showed that it was a circular DNA molecule of 16,735 bp with 81.07% AT content and comprised 13 protein-coding genes (PCG), 22 tRNA genes, 2 rRNA genes, and 1 control region. The PCGs were initiated using typical ATN (Met) and TTG (Met) start codons and terminated using typical TAN stop codons. The phylogenetic position of Mastax within the Carabidae was first evaluated using complete mitogenomes, and the results showed that it was close to Cicindela anchoralis and Manticora tibialis.

Complete mitochondrial genome of Morphostenophanes yunnanus (Zhou, 2020) (Insecta: Coleoptera: Tenebrionidae) and phylogenetic analysis.

Project description:Morphostenophanes yunnanus (Zhou, 2020) is widely distributed from central to eastern Yunnan with distinct geographical variations in morphology. Beetles were collected in Manwan Town, and a mitochondrial genome sequence (GenBank accession number MZ298928) of this species was sequenced using the MGI-SEQ 2000 platform, assembled using NOVOPlasty v4.3.1, and characterized. The mitogenome was a circular DNA molecule of 15,690 bp with 64.710% AT content, which comprised 13 protein-coding genes, 22 tRNA genes, two rRNA genes, and one control region. The protein-coding genes showed the typical ATN (Met) and TTG (Met) start codons, except nad1 and cox1 (TTG as start codon), and were terminated by typical TAN stop codons. The maximum-likelihood polygenetic tree was generated using protein sequences of thirteen protein-coding regions of seventeen mitogenomes with mtREV + G + F + I with 1000 replicates under the Bayesian information criterion using MEGA 11, which showed that M. yunnanus was the most closely related to M. sinicus. This study provides essential genetic and molecular data for phylogenetic analyses of the genus Morphostenophanes.

Project description:The complete mitochondrial genome of Blaps rynchopetera Fairmaire (GenBank accession number MN267802) from Dali city consists of a circular DNA molecule of 16,766 bp (with 72.01% A + T content), which is longer than that of the mitogenome of B. rynchopetera from Kunming. The mitogenome comprises 13 protein-coding, 22 tRNA, and 2 rDNA genes. The protein-coding genes have typical ATN (Met) initiation codons, and are terminated by typical TAN stop codons. A phylogenetic tree generated by the neighbour-joining method showed that B. rynchopetera is closely related to Platydema sp. PLA01, which has the potential for medicinal development.
