Complete mitochondrial genome of Spodoptera littoralis (Lepidoptera: Noctuidae) from Egypt.

ABSTRACT: The polyphagous cotton leafworm (Spodoptera littoralis) is one of the most destructive herbivorous insects worldwide. The present study reports the complete mitochondrial genome of S. littoralis collected from Egypt. The circular-mapping mitogenome was 15,408 bp in length with an overall A + T content of 81.1%, encoding a common set of 37 genes, including 13 protein-coding genes (PCGs), 22 tRNA genes, and two rRNA genes. Most PCGs were found to use conventional ATN as the start codon and TAN as the stop codon. The phylogenetic tree based on the nucleic acid sequences of 13 shared PCGs of 29 Noctuidae species revealed that S. littoralis and Spodoptera litura are sister species. The data in this study will be helpful to understand geographical genetic variations, phylogenetic relationships, and species identification of S. littoralis.

Transcriptome Analysis of Gene Families Involved in Chemosensory Function in Spodoptera littoralis (Lepidoptera: Noctuidae).

Project description:BackgroundDeciphering the molecular mechanisms mediating the chemical senses, taste, and smell has been of vital importance for understanding the nature of how insects interact with their chemical environment. Several gene families are implicated in the uptake, recognition, and termination of chemical signaling, including binding proteins, chemosensory receptors and degrading enzymes. The cotton leafworm, Spodoptera littoralis, is a phytophagous pest and current focal species for insect chemical ecology and neuroethology.ResultsWe produced male and female Illumina-based transcriptomes from chemosensory and non-chemosensory tissues of S. littoralis, including the antennae, proboscis, brain and body carcass. We have annotated 306 gene transcripts from eight gene families with known chemosensory function, including 114 novel candidate genes. Odorant receptors responsive to floral compounds are expressed in the proboscis and may play a role in guiding proboscis probing behavior. In both males and females, expression of gene transcripts with known chemosensory function, including odorant receptors and pheromone-binding proteins, has been observed in brain tissue, suggesting internal, non-sensory function for these genes.ConclusionsA well-curated set of annotated gene transcripts with putative chemosensory function is provided. This will serve as a resource for future chemosensory and transcriptomic studies in S. littoralis and closely related species. Collectively, our results expand current understanding of the expression patterns of genes with putative chemosensory function in insect sensory and non-sensory tissues. When coupled with functional data, such as the deorphanization of odorant receptors, the gene expression data can facilitate hypothesis generation, serving as a substrate for future studies.

Project description:The fall armyworm, Spodoptera frugiperda, is a serious pest in large numbers on more than 350 plant species in the world. We have determined a 15,388 bp mitogenome of S. frugiperda which includes 13 protein-coding genes, two ribosomal RNA genes, and 22 transfer RNAs. The base composition was AT-biased (81.3%). Phylogenetic trees present that Korean S. frugiperda placed in basal position of S. frugiperda clade. S. frugiperda mitochondrial genome can be used for understanding recent active migration of S. frugiperda.

Characterization of the mitochondrial genome of the lawn cutworm Spodoptera depravata (Lepidoptera: Noctuidae).

Project description:The lawn cutworm, Spodoptera depravata, is one of the most important pests that causes economic damage to grass crops. This study reports the complete mitochondrial genome of an S. depravata sample collected in China. The genome is a circular molecule 15,460 bp in length with an overall A + T content of 81.6%. It contains 13 protein-coding genes, 22 transfer RNA genes, and two ribosomal RNA genes. The gene content and organization of the mitogenome of S. depravata are identical to those of other Spodoptera species. Maximum-likelihood phylogenetic analysis based on mitogenomes showed a close evolutionary relationship between S. depravata and S. exempta. This study provides new molecular data for the identification and further phylogenetic analyses of Spodoptera species.

Impact of transgenerational host switch on gut bacterial assemblage in generalist pest, Spodoptera littoralis (Lepidoptera: Noctuidae).

Project description:Diet composition is vital in shaping gut microbial assemblage in many insects. Minimal knowledge is available about the influence of transgenerational diet transition on gut microbial community structure and function in polyphagous pests. This study investigated transgenerational diet-induced changes in Spodoptera littoralis larval gut bacteriome using 16S ribosomal sequencing. Our data revealed that 88% of bacterial populations in the S. littoralis larval gut comprise Proteobacteria, Firmicutes, Actinobacteria, and Bacteroidetes. The first diet transition experiment from an artificial diet (F0) to a plant diet (F1), cabbage and cotton, caused an alteration of bacterial communities in the S. littoralis larval gut. The second transgenerational diet switch, where F1 larvae feed on the same plant in the F2 generation, displayed a significant variation suggesting further restructuring of the microbial communities in the Spodoptera larval gut. F1 larvae were also challenged with the plant diet transition at the F2 generation (cabbage to cotton or cotton to cabbage). After feeding on different plant diets, the microbial assemblage of F2 larvae pointed to considerable differences from other F2 larvae that continued on the same diet. Our results showed that S. littoralis larval gut bacteriome responds rapidly and inexplicably to different diet changes. Further experiments must be conducted to determine the developmental and ecological consequences of such changes. Nevertheless, this study improves our perception of the impact of transgenerational diet switches on the resident gut bacteriome in S. littoralis larvae and could facilitate future research to understand the importance of symbiosis in lepidopteran generalists better.

The complete mitochondrial genome of the Sideridis albicosta (Lepidoptera: Noctuoidea: Noctuidae).

Project description:The complete mitochondrial genome (mitogenome) of Sideridis albicosta is described in this study. The circular molecule is 15,320 bp in length and contains 13 protein-coding genes (PCGs), two rRNA genes, 22 tRNA genes, and one AT-rich region. Thirteen PCGs is 11,252 bp in total, encoding 3723 amino acids. All PCGs start with ATN, except for COI gene starting with CGA; 10 of the 13 PCGs use the typical stop codon TAA, whereas COI, COII, and ND4 stop with a single T. The rrnL and rrnS genes are 1377 bp and 783 bp in length, respectively. The 328 bp AT-rich region contains several structures characteristic of the lepidopterans. New phylogenetic analyses upon mitogenomics would provide us further insights on the taxonomy and phylogeny of Noctuoidea.

Toxicity and biochemical impact of methoxyfenozide/spinetoram mixture on susceptible and methoxyfenozide-selected strains of Spodoptera littoralis (Lepidoptera: Noctuidae).

Project description:Methoxyfenozide (M) is one of the selective insecticides used in integrated pest management (IPM) programs for lepidopteran pests. However, recent studies reported a development of M-resistance, which prompted us to look for alternatives. Here, we investigate the potency of a mixture of M with spinetoram (Sp) on M-resistant insects. In the laboratory, a selection pressure with M has carried out on Spodoptera littoralis (Lepidoptera: Noctuidae) strains. A dipping technique was used to evaluate the toxicity of a sublethal concentration of M and Sp. on S. littoralis larvae, and the same concentrations were used to assess the toxic impact of their combination on susceptible (SUS) and M-selected (MS) strains. The toxicity of M/Sp mixtures was computed using a combination index equation, and a potentiation effect was observed in the two tested strains. Synergism tests revealed that piperonyl butoxide had considerable synergistic effects on M toxicity in the MS strain. The results revealed that the M/Sp mixture's negative effect on both monooxygenases and esterases is most likely the cause of its potentiation effect on the SUS and MS strains. It was concluded that M/Sp mixtures are effective against M-resistant S. littoralis strains, so these can be used in IPM programs.

Monitoring Resistance and Biochemical Studies of Three Egyptian Field Strains of Spodoptera littoralis (Lepidoptera: Noctuidae) to Six Insecticides.

Project description:BackgroundSpodoptera littoralis (Boisd.) is a prominent agricultural insect pest that has developed resistance to a variety of insecticide classes. In this study, the resistance of three field strains of S. littoralis, collected over three consecutive seasons (2018 to 2020) from three Egyptian Governorates (El-Fayoum, Behera and Kafr El-Shiekh), to six insecticides was monitored.MethodsLaboratory bioassays were carried out using the leaf-dipping method to examine the susceptibility of the laboratory and field strains to the tested insecticides. Activities of detoxification enzymes were determined in an attempt to identify resistance mechanisms.ResultsThe results showed that LC50 values of the field strains ranged from 0.0089 to 132.24 mg/L, and the corresponding resistance ratio (RR) ranged from 0.17 to 4.13-fold compared with the susceptible strain. Notably, low resistance developed to spinosad in all field strains, and very low resistance developed to alpha-cypermethrin and chlorpyrifos. On the other hand, no resistance developed to methomyl, hexaflumeron or Bacillus thuringiensis. The determination of detoxification enzymes, including carboxylesterases (α- and β-esterase), mixed function oxidase (MFO) and glutathione-S-transferase (GST), or the target site of acetylcholinesterase (AChE), revealed that the three field strains had significantly different activity levels compared with the susceptible strain.ConclusionOur findings, along with other tactics, are expected to help with the resistance management of S. littoralis in Egypt.

Insights into the toxicity, biochemical activity, and molecular docking of Cymbopogon citratus essential oils and citral on Spodoptera littoralis (Lepidoptera: Noctuidae).

Project description:Insecticide resistance is a significant problem in insect management that can result from several processes including target-site change and increased activity of detoxifying enzymes. Spodoptera littoralis is one of the most resistant insect pests. For more effective insect management, alternatives to synthetic pesticides are encouraged. One of these alternatives is essential oils (EOs). Cymbopogon citratus EO and its main constituent citral were, therefore, considered in this study. The results revealed that C. citratus EO and citral exhibited significant larvicidal activity against S. littoralis, and the former was insignificantly more toxic than the latter. Additionally, treatments significantly affected the activity of detoxification enzymes. Cytochrome P-450 and glutathione-S-transferase were inhibited, while carboxylesterases, a-esterase and β-esterase, were induced. The molecular docking study indicated that citral bonded with the amino acids cysteine (CYS 345) and histidine (HIS 343) of cytochrome P-450. This result suggests that interaction with cytochrome P-450 enzyme is one key mechanism by which C. citratus EO and citral act in S. littoralis. The results of our study are hoped to contribute to a better understanding of the mechanism of action of essential oils at the biochemical and molecular levels and provide safer and more efficient pest management solutions for S. littoralis.

Molecular Oscillator Affects Susceptibility of Caterpillars to Insecticides: Studies on the Egyptian Cotton Leaf Worm-Spodoptera littoralis (Lepidoptera: Noctuidae).

Project description:The molecular oscillator is the core of the biological clock and is formed by genes and proteins whose cyclic expression is regulated in the transcriptional-translational feedback loops (TTFLs). Proteins of the TTFLs are regulators of both their own and executive genes involved in the control of many processes in insects (e.g., rhythmic metabolism of xenobiotics, including insecticides). We disrupted the clock operation in S. littoralis larvae by injecting the dsRNA of clock genes into their body cavity and culturing the larvae under continuous light. As a result, the daily susceptibility of larvae to insecticides was abolished and the susceptibility itself increased (in most cases). In the fat body, midgut, and Malpighian tubules (the main organs metabolizing xenobiotics) of the larvae treated with injected-dsRNA, the daily activity profiles of enzymes involved in detoxification-cytochrome P450 monooxygenases, Glutathione-S-transferase, and esterase-have changed significantly. The presented results prove the role of the molecular oscillator in the regulation of larvae responses to insecticides and provide grounds for rational use of these compounds (at suitable times of the day), and may indicate clock genes as potential targets of molecular manipulation to produce plant protection compounds based on the RNAi method.

Spodoptera littoralis genome mining brings insights on the dynamic of expansion of gustatory receptors in polyphagous noctuidae.

Project description:The bitter taste, triggered via gustatory receptors, serves as an important natural defense against the ingestion of poisonous foods in animals, and the increased host breadth is usually linked to an increase in the number of gustatory receptor genes. This has been especially observed in polyphagous insect species, such as noctuid species from the Spodoptera genus. However, the dynamic and physical mechanisms leading to these gene expansions and the evolutionary pressures behind them remain elusive. Among major drivers of genome dynamics are the transposable elements but, surprisingly, their potential role in insect gustatory receptor expansion has not been considered yet. In this work, we hypothesized that transposable elements and possibly positive selection would be involved in the highly dynamic evolution of gustatory receptor in Spodoptera spp. We first sequenced de novo the full 465 Mb genome of S. littoralis, and manually annotated the main chemosensory genes, including a large repertoire of 373 gustatory receptor genes (including 19 pseudogenes). We also improved the completeness of S. frugiperda and S. litura gustatory receptor gene repertoires. Then, we annotated transposable elements and revealed that a particular category of class I retrotransposons, the SINE transposons, was significantly enriched in the vicinity of gustatory receptor gene clusters, suggesting a transposon-mediated mechanism for the formation of these clusters. Selection pressure analyses indicated that positive selection within the gustatory receptor gene family is cryptic, only 7 receptors being identified as positively selected. Altogether, our data provide a new good quality Spodoptera genome, pinpoint interesting gustatory receptor candidates for further functional studies and bring valuable genomic information on the mechanisms of gustatory receptor expansions in polyphagous insect species.
