The complete mitochondrial genome of Beauveria lii (Hypocreales: Cordycipitaceae).

ABSTRACT: The mitochondrial genome of Beauveria lii, strain RCEF500, was sequenced on the NovaSeq 6000 and the Nanopore Sequencer, and annotated. The genome is 59,014 bp in length, encoding 15 conserved protein-coding genes (PCGs), 2 rRNA genes and 23 tRNA genes. The nucleotide composition of Beauveria lii mitochondrial genome was 38.23% of A, 35.81% of T, 11.61% of C, 14.36% of G, 25.97% of G + C content. Phylogenetic analysis confirmed B. lii as a member of Beauveria (Cordycipitaceae). The mitochondrial genome of B. lii will contribute to the understanding of phylogeny and evolution of the genus and family.

Selection of Beauveria bassiana (Hypocreales: Cordycipitaceae) strains to control Xyleborus affinis (Curculionidae: Scolytinae) females.

Project description:BackgroundXyleborus affinis Eichhoff (Coleoptera: Curculionidae) is an ambrosia beetle reported to affect avocado trees (Persea americana Mill.). The use of the entomopathogenic fungus (EPF) Beauveria bassiana (Bals.-Criv.) Vuill. for ambrosia beetle control represents an alternative to insecticides.MethodsThis study was designed in two stages to select B. bassiana strains with potential to control X. affinis females. In the first stage, 19 B. bassiana Mexican strains from EPF collection, isolated from Coleoptera (CHE-CNRCB, http://www.gob.mx/senasica/documentos/coleccion-de-hongos-entomopatogenos), were tested. Analyses included radial growth rate, conidial yield, spore germination, and germ tube length. Results were analysed by Principal Component Analysis (PCA) to identify clusters within favourable growth phenotypes. For the second stage, 10 selected strains were re-analysed for virulence-related metabolic characteristic, including cell wall-bound cuticle-degrading enzymes-Pr1-like proteases and β-N-acetyl glucosaminidases (NAGase) chitinases, conidial hydrophobicity and monopolar germination parameters. A second PCA analysis was run for those virulence parameters analysed, and upon results strains CHE-CNRCB 44, 171, 431 and 485 were selected and tested against X. affinis females. Females were treated with a 1 × 108 conidia mL-1 suspension (recommended rate), using a Potter Tower.ResultsAll strains showed insecticidal activity, inducing up to 58% mortality; about 30% dead beetles developed aerial mycelia (CHE-CNRCB 485) and the fastest mortality rate was t0 = 1.95 (CHE-CNRCB 44).ConclusionSince all selected strains showed virulence against X. affinis females, results indicated the possibility of selecting B. bassiana strains based on multiple metabolic attributes, as a preliminary test to perform bioassays against order-related target insects.

Larval Mortality and Ovipositional Preference in Aedes albopictus (Diptera: Culicidae) Induced by the Entomopathogenic Fungus Beauveria bassiana (Hypocreales: Cordycipitaceae).

Project description:Entomopathogenic fungi allow chemical-free and environmentally safe vector management. Beauveria bassiana (Balsamo-Crivelli) Vuillemin is a promising biological control agent and an important component of integrated vector management. We investigated the mortality of Aedes albopictus (Skuse) larvae exposed to five concentrations of B. bassiana using Mycotrol ESO and adult oviposition behavior to analyze the egg-laying preferences of wild Ae. albopictus in response to different fungal concentrations. We examined the mortality of mid-instars exposed to B. bassiana concentrations of 1 × 104, 1 × 105, 1 × 106, 1 × 107, and 1 × 108 conidia/ml every 24 h for 12 d. In the oviposition behavior study, the fungus was applied to wooden paddles at 1 × 105, 1 × 107, and 1 × 109 conidia/ml, and the paddles were individually placed into quad-ovitraps. Both experiments contained control groups without B. bassiana. Kaplan-Meier survival analysis revealed that larval mortality was concentration dependent. The median lethal concentration was 2.43 × 105 conidia/ml on d 12. The median lethal time was 3.68 d at 1 × 106 conidia/ml. Oviposition monitoring revealed no significant difference in egg count between the control and treatment paddles. We observed an inverse relationship between the concentration of B. bassiana and the percentage of paddles with eggs. We concluded that concentrations above 1 × 106 conidia/ml are larvicidal, and Ae. albopictus laid similar numbers of eggs on fungus-impregnated and control wooden substrates; however, they were more likely to oviposit on substrates without B. bassiana. With these findings, we suggest that B. bassiana-infused ovitraps can be used for mosquito population monitoring while also delivering mycopesticides to adult mosquitoes.

A phylogenetically-based nomenclature for Cordycipitaceae (Hypocreales).

Project description:The ending of dual nomenclatural systems for pleomorphic fungi in 2011 requires the reconciliation of competing names, ideally linked through culture based or molecular methods. The phylogenetic systematics of Hypocreales and its many genera have received extensive study in the last two decades, however resolution of competing names in Cordycipitaceae has not yet been addressed. Here we present a molecular phylogenetic investigation of Cordycipitaceae that enables identification of competing names in this family, and provides the basis upon which these names can be maintained or suppressed. The taxonomy presented here seeks to harmonize competing names by principles of priority, recognition of monophyletic groups, and the practical usage of affected taxa. In total, we propose maintaining nine generic names, Akanthomyces, Ascopolyporus, Beauveria, Cordyceps, Engyodontium, Gibellula, Hyperdermium, Parengyodontium, and Simplicillium and the rejection of eight generic names, Evlachovaea, Granulomanus, Isaria, Lecanicillium, Microhilum, Phytocordyceps, Synsterigmatocystis, and Torrubiella. Two new generic names, Hevansia and Blackwellomyces, and a new species, Beauveria blattidicola, are described. New combinations are also proposed in the genera Akanthomyces, Beauveria, Blackwellomyces, and Hevansia.

It has a total length of 50, 107 bp, the base composition of this mitogenome is as follows: A (35.9%), T (34.9%), C (14.4%), and G (14.8%). The mitogenome contains 26 protein-coding genes, two ribosomal RNA (rRNA) genes, and 25 transfer RNA (tRNA) genes. The taxonomic status of the A. alternata mitogenome exhibits a closest relationship with Phaeosphaeria nodorum.

The complete mitochondrial genome of Ilyonectria sp. (Hypocreales: Hypocreomycetidae).

Project description:In the present study, we presented the complete mitochondrial genome (mitogenome) of Ilyonectria sp. The complete mitogenome of Ilypnectria sp. was composed of circular DNA molecules, with a total length of 34, 584 bp. The base composition of this mitogenome is as follows: A (35.31%), T (35.92%), G (15.93%), and C (12.83%). The mitogenome contains 18 protein-coding genes, 2 ribosomal RNA genes (rRNA), and 25 transfer RNA (tRNA) genes. The taxonomic status of the Ilyonectria sp. mitogenome exhibits a closest relationship with Ilyonectria destructans.

The complete mitochondrial genome of Cladobotryum mycophilum (Hypocreales: Sordariomycetes).

Project description:Cladobotryum mycophilum is the causal agent of cobweb disease in many important mushroom crops. In this study, we report the complete mitochondrial genome of C. mycophilum for the first time. The genome is 78,729 bp long and comprises 52 protein-coding genes (PCGs), 2 ribosomal RNA genes (rRNA), and 26 transfer RNA (tRNA) genes. The nucleotide composition of C. mycophilum mitochondrial genome is as follows: A (38.06%), T (34.68%), C (12.19%), and G (15.07%). Phylogenetic analysis revealed that C. mycophilum had a close relationship with Cladobotryum varium from Hypocreaceae. This study provided a basis for studies of the mitochondrial evolution of Hypocreaceae.

Biocontrol of Xyleborus affinis (Curculionidae: Scolitinae) Females and Progeny by Beauveria bassiana (Hypocreales: Cordycipitaceae) in a Sawdust Artificial Diet Model.

Project description:The ambrosia beetle Xyleborus affinis, recently reported affecting avocado trees in Mexico, represents one of the most widespread insects worldwide. Previous reports have shown that Xyleborus genera members are susceptible to Beauveria bassiana and other entomopathogenic fungus strains. However, their effect on borer beetles' progeny has not been fully investigated. The aim of the present study was to determine the insecticidal activity of B. bassiana on X. affinis adult females and their progeny in an artificial sawdust diet bioassay model. The B. bassiana strains CHE-CNRCB 44, 171, 431, and 485 were individually tested on females at concentrations ranging from 2 × 106 to 1 × 109 conidia mL-1. After 10 d of incubation, diet was evaluated to count laid eggs, larvae, and adults. Insect conidia loss after exposure was determined by attached conidia to each insect after 12 h of exposure. The results showed that females' mortality ranged between 3.4% and 50.3% in a concentration-response manner. Furthermore, we did not observe statistical differences among strains at the highest concentration. CHE-CNRCB 44 showed the highest mortality at the lowest concentration and reduced larvae and laid eggs at the highest concentration (p < 0.01). Strains CHE-CNRCB 44, 431, and 485 significantly decreased larvae, as compared with the untreated control. After 12 h, up to 70% of conidia was removed by the effect of the artificial diet. In conclusion, B. bassiana has the potential to control X. affinis adult females and progeny.

The complete mitochondrial genome of the important mycoparasite Clonostachys rosea (Hypocreales, Ascomycota).

Project description:The complete nucleotide sequence of mitochondrial genome of the important mycoparasitic fungus Clonostachys rosea was determined using the next-generation sequencing technology. The circular molecule is 40,921 bp long with a GC content of 27.90%. Gene prediction revealed 42 genes encoding 15 conserved proteins, 25 tRNAs, the large and small ribosomal RNAs. All genes are located on the same strand. It is found to be similar to the previously sequenced mitochondrial genomes of Acremonium chrysogenum and Nectria cinnabarina. The differences lie in the copy number of trnG-UCC and locations of trnN-GUU and cox2. The phylogenetic analysis confirmed C. rosea as a sister taxon of A. chrysogenumin (Bionectriaceae). The mitochondrial genome of C. rosea will contribute to the understanding of phylogeny and evolution of Hypocreales.

An efficient Agrobacterium tumefaciens-mediated transformation method for Simplicillium subtropicum (Hypocreales: Cordycipitaceae).

Project description:Filamentous fungi are the organisms of choice for most industrial biotechnology. Some species can produce a variety of secondary metabolites and enzymes of commercial interest, and the production of valuable molecules has been enhanced through different molecular tools. Methods for genetic manipulation and transformation have been essential for the optimization of these organisms. The genus Simplicillium has attracted increased attention given several potential biotechnological applications. The Simplicillium genus harbors several entomopathogenic species and some isolates have been explored for bioremediation of heavy metal contaminants. Furthermore, the myriad of secondary metabolites isolated from Simplicillium spp. render these organisms as ideal targets for deep exploration and further biotechnological mining possibilities. However, the lack of molecular tools hampered the exploration of this genus. Thus, an Agrobacterium tumefaciens-mediated transformation method was established for Simplicillium subtropicum, employing the far-red fluorescent protein TURBOFP635/Katushka, as a visual marker, and the selection marker SUR gene, that confers resistance to chlorimuron ethyl. Notably, one round of transformation using the established method yielded almost 400 chlorimuron resistant isolates. Furthermore, these transformants displayed mitotic stability for, at least, five generations. We anticipate that this method can be useful for deep molecular exploration and improvement of strains in the Simplicillium genus.

The complete mitochondrial genome of Trichoderma simmonsii (Hypocreales: Hypocreaceae) from the Southern Coast of Korea.

Project description:Fungal species in the genus Trichoderma are widely used for industrial enzyme production and as biocontrol agents. In this study, we report the complete mitochondrial genome of a marine-derived Trichoderma simmonsii strain GH-Sj1, which belongs to the Harzianum clade of Trichoderma. GH-Sj1 was isolated from an edible sea alga Saccharina japonica collected from the southern coast of Korea. This newly assembled circular molecule is 28,668 bp in length and consists of 15 protein-coding genes, 26 transfer RNA genes, and two ribosomal RNA genes. Phylogenetic analysis using the maximum likelihood method shows that T. simmonsii GH-Sj1 is closely related to Trichoderma harzianum and Trichoderma lixii. To the best of our knowledge, this is the first characterization of a marine-derived mitogenome within the genus Trichoderma.
