Age-Related EEG Power Reductions Cannot Be Explained by Changes of the Conductivity Distribution in the Head Due to Brain Atrophy.

ABSTRACT: Electroencephalogram (EEG) power reductions in the aging brain have been described by numerous previous studies. However, the underlying mechanism for the observed brain signal power reduction remains unclear. One possible cause for reduced EEG signals in elderly subjects might be the increased distance from the primary neural electrical currents on the cortex to the scalp electrodes as the result of cortical atrophies. While brain shrinkage itself reflects age-related neurological changes, the effects of changes in the distribution of electrical conductivity are often not distinguished from altered neural activity when interpreting EEG power reductions. To address this ambiguity, we employed EEG forward models to investigate whether brain shrinkage is a major factor for the signal attenuation in the aging brain. We simulated brain shrinkage in spherical and realistic brain models and found that changes in the conductor geometry cannot fully account for the EEG power reductions even when the brain was shrunk to unrealistic sizes. Our results quantify the extent of power reductions from brain shrinkage and pave the way for more accurate inferences about deficient neural activity and circuit integrity based on EEG power reductions in the aging population.

Project description:ObjectivesA core assumption of meditation training is that cognitive capacities developed during formal practice will transfer to other contexts or activities as expertise develops over time. This implies that meditation training might influence domain-general neurocognitive systems, the spontaneous activity of which should be reflected in the dynamics of the resting brain. Previous research has demonstrated that 3 months of meditation training led to reductions in EEG beta band power during mindfulness of breathing practice. The current study extends these findings to ask whether concomitant shifts in power are observed during 2 min of eyes closed rest, when participants are not explicitly engaged in formal meditation.MethodsExperienced meditation practitioners were randomly assigned to practice 3 months of focused attention meditation in a residential retreat, or to serve as waitlist controls. The waitlist controls later completed their own 3-month retreat. Permutation-based cluster analysis of 88-channel resting EEG data was used to test for spectral changes in spontaneous brain activity over the course of the retreats.ResultsLongitudinal reductions in EEG power in the beta frequency range were identified and replicated across the two independent training periods. Less robust reductions were also observed in the high alpha frequency range, and in individual peak alpha frequency. These changes closely mirror those previously observed during formal mindfulness of breathing meditation practice.ConclusionsThese findings suggest that the neurocognitive effects of meditation training can extend beyond the bounds of formal practice, influencing the spontaneous activity of the resting brain. Project description:There has been a growing interest in computational electroencephalogram (EEG) signal processing in a diverse set of domains, such as cortical excitability analysis, event-related synchronization, or desynchronization analysis. In recent years, several inconsistencies were found across different EEG studies, which authors often attributed to methodological differences. However, the assessment of such discrepancies is deeply underexplored. It is currently unknown if methodological differences can fully explain emerging differences and the nature of these differences. This study aims to contrast widely used methodological approaches in EEG processing and compare their effects on the outcome variables. To this end, two publicly available datasets were collected, each having unique traits so as to validate the results in two different EEG territories. The first dataset included signals with event-related potentials (visual stimulation) from 45 subjects. The second dataset included resting state EEG signals from 16 subjects. Five EEG processing steps, involved in the computation of power and phase quantities of EEG frequency bands, were explored in this study: artifact removal choices (with and without artifact removal), EEG signal transformation choices (raw EEG channels, Hjorth transformed channels, and averaged channels across primary motor cortex), filtering algorithms (Butterworth filter and Blackman-Harris window), EEG time window choices (-750 ms to 0 ms and -250 ms to 0 ms), and power spectral density (PSD) estimation algorithms (Welch's method, Fast Fourier Transform, and Burg's method). Powers and phases estimated by carrying out variations of these five methods were analyzed statistically for all subjects. The results indicated that the choices in EEG transformation and time-window can strongly affect the PSD quantities in a variety of ways. Additionally, EEG transformation and filter choices can influence phase quantities significantly. These results raise the need for a consistent and standard EEG processing pipeline for computational EEG studies. Consistency of signal processing methods cannot only help produce comparable results and reproducible research, but also pave the way for federated machine learning methods, e.g., where model parameters rather than data are shared.

Project description:IntroductionElectroencephalographic (EEG) data quality is severely compromised when recorded inside the magnetic resonance (MR) environment. Here we characterized the impact of the ballistocardiographic (BCG) artifact on resting-state EEG spectral properties and compared the effectiveness of seven common BCG correction methods to preserve EEG spectral features. We also assessed if these methods retained posterior alpha power reactivity to an eyes closure-opening (EC-EO) task and compared the results from EEG-informed fMRI analysis using different BCG correction approaches.MethodElectroencephalographic data from 20 healthy young adults were recorded outside the MR environment and during simultaneous fMRI acquisition. The gradient artifact was effectively removed from EEG-fMRI acquisitions using Average Artifact Subtraction (AAS). The BCG artifact was corrected with seven methods: AAS, Optimal Basis Set (OBS), Independent Component Analysis (ICA), OBS followed by ICA, AAS followed by ICA, PROJIC-AAS and PROJIC-OBS. EEG signal preservation was assessed by comparing the spectral power of traditional frequency bands from the corrected rs-EEG-fMRI data with the data recorded outside the scanner. We then assessed the preservation of posterior alpha functional reactivity by computing the ratio between the EC and EO conditions during the EC-EO task. EEG-informed fMRI analysis of the EC-EO task was performed using alpha power-derived BOLD signal predictors obtained from the EEG signals corrected with different methods.ResultsThe BCG artifact caused significant distortions (increased absolute power, altered relative power) across all frequency bands. Artifact residuals/signal losses were present after applying all correction methods. The EEG reactivity to the EC-EO task was better preserved with ICA-based correction approaches, particularly when using ICA feature extraction to isolate alpha power fluctuations, which allowed to accurately predict hemodynamic signal fluctuations during the EEG-informed fMRI analysis.DiscussionCurrent software solutions for the BCG artifact problem offer limited efficiency to preserve the EEG spectral power properties using this particular EEG setup. The state-of-the-art approaches tested here can be further refined and should be combined with hardware implementations to better preserve EEG signal properties during simultaneous EEG-fMRI. Existing and novel BCG artifact correction methods should be validated by evaluating signal preservation of both ERPs and spontaneous EEG spectral power.

Project description:Buprenorphine is a μ-opioid receptor (MOR) partial agonist used to manage pain and addiction. QTC prolongation that crosses the 10 msec threshold of regulatory concern was observed at a supratherapeutic dose in two thorough QT studies for the transdermal buprenorphine product BUTRANS®. Because QTC prolongation can be associated with Torsades de Pointes (TdP), a rare but potentially fatal ventricular arrhythmia, these results have led to further investigation of the electrophysiological effects of buprenorphine. Drug-induced QTC prolongation and TdP are most commonly caused by acute inhibition of hERG current (IhERG) that contribute to the repolarizing phase of the ventricular action potentials (APs). Concomitant inhibition of inward late Na+ (INaL) and/or L-type Ca2+ (ICaL) current can offer some protection against proarrhythmia. Therefore, we characterized the effects of buprenorphine and its major metabolite norbuprenorphine on cardiac hERG, Ca2+, and Na+ ion channels, as well as cardiac APs. For comparison, methadone, a MOR agonist associated with QTC prolongation and high TdP risk, and naltrexone and naloxone, two opioid receptor antagonists, were also studied. Whole cell recordings were performed at 37°C on cells stably expressing hERG, CaV1.2, and NaV1.5 proteins. Microelectrode array (MEA) recordings were made on human induced pluripotent stem cell-derived cardiomyocytes (iPSC-CMs). The results showed that buprenorphine, norbuprenorphine, naltrexone, and naloxone had no effect on IhERG, ICaL, INaL, and peak Na+ current (INaP) at clinically relevant concentrations. In contrast, methadone inhibited IhERG, ICaL, and INaL. Experiments on iPSC-CMs showed a lack of effect for buprenorphine, norbuprenorphine, naltrexone, and naloxone, and delayed repolarization for methadone at clinically relevant concentrations. The mechanism of QTC prolongation is opioid moiety-specific. This remains undefined for buprenorphine, while for methadone it involves direct hERG channel block. There is no evidence that buprenorphine use is associated with TdP. Whether this lack of TdP risk can be generalized to other drugs with QTC prolongation not mediated by acute hERG channel block warrants further study.
