Gradual onset of the Maunder Minimum revealed by high-precision carbon-14 analyses.

ABSTRACT: The Sun exhibits centennial-scale activity variations and sometimes encounters grand solar minimum when solar activity becomes extremely weak and sunspots disappear for several decades. Such an extreme weakening of solar activity could cause severe climate, causing massive reductions in crop yields in some regions. During the past decade, the Sun's activity has tended to decline, raising concerns that the Sun might be heading for the next grand minimum. However, we still have an underdeveloped understanding of solar dynamo mechanisms and hence precise prediction of near-future solar activity is not attained. Here we show that the 11-year solar cycles were significantly lengthened before the onset of the Maunder Minimum (1645-1715 CE) based on unprecedentedly high-precision data of carbon-14 content in tree rings. It implies that flow speed in the convection zone is an essential parameter to determine long-term solar activity variations. We find that a 16 year-long cycle had occurred three solar cycles before the onset of prolonged sunspot disappearance, suggesting a longer-than-expected preparatory period for the grand minimum. As the Sun has shown a tendency of cycle lengthening since Solar Cycle 23 (1996-2008 CE), the behavior of Solar Cycle 25 can be critically important to the later solar activity.

Project description:The classic phenotype of Fabry disease, X-linked alpha -galactosidase A (alpha -Gal A) deficiency, has an estimated incidence of approximately 1 in 50,000 males. The recent recognition of later-onset variants suggested that this treatable lysosomal disease is more frequent. To determine the disease incidence, we undertook newborn screening by assaying the alpha-Gal A activity in blood spots from 37,104 consecutive Italian male neonates. Enzyme-deficient infants were retested, and "doubly screened-positive" infants and their relatives were diagnostically confirmed by enzyme and mutation analyses. Twelve (0.03%) neonates had deficient alpha-Gal A activities and specific mutations, including four novel missense mutations (M51I, E66G, A73V, and R118C), three missense mutations (F113L, A143T, and N215S) identified previously in later-onset patients, and one splicing defect (IVS5(+1G-->T)) reported in a patient with the classic phenotype. Molecular modeling and in vitro overexpression of the missense mutations demonstrated structures and residual activities, which were rescued/enhanced by an alpha-Gal A-specific pharmacologic chaperone, consistent with mutations that cause the later-onset phenotype. Family studies revealed undiagnosed Fabry disease in affected individuals. In this population, the incidence of alpha-Gal A deficiency was 1 in approximately 3,100, with an 11 : 1 ratio of patients with the later-onset : classic phenotypes. If only known disease-causing mutations were included, the incidence would be 1 in approximately 4,600, with a 7 : 1 ratio of patients with the later-onset : classic phenotypes. These results suggest that the later-onset phenotype of Fabry disease is underdiagnosed among males with cardiac, cerebrovascular, and/or renal disease. Recognition of these patients would permit family screening and earlier therapeutic intervention. However, the higher incidence of the later-onset phenotype in patients raises ethical issues related to when screening should be performed--in the neonatal period or at early maturity, perhaps in conjunction with screening for other treatable adult-onset disorders.

Project description:BackgroundThe transition from a high forage to a highly fermentable diet can induce digestive disorders in the rumen. To date, the host mechanisms that regulate the adaption to such dietary transition are largely unknown. To understand the molecular mechanisms involved in such phenomena, RNA-sequencing was performed to identify the changes in the transcriptome of ruminal epithelia during gradual transition from a diet containing 0% to 89% grain.ResultsIn total, the expression of 11,044, 11,322 and 11,282 genes were detected in ruminal epithelia of beef heifers (n = 15) fed 0%, 72% and 89% barley grain diet, respectively. The transcriptome profiles of rumen epithelia differed between low grain diet (LGD) (0% grain) and high grain diet (HGD) (72% and 89%), and HGD tended to reduce the expression of genes involved in epithelial catalytic and binding activities. When diet was changed from 72% to 89% grain, the mean ruminal pH change was significantly different among individual heifers with five of them decreased (down group (DG); from 6.30±0.09 to 5.87±0.15, P < 0.01) and five of them increased (up group (UG); from 5.84±0.42 to 6.35±0.37, P < 0.05). The functional analysis of differentially expressed (DE) genes revealed inhibited "Immune response of leukocytes", "Attraction of phagocytes", and "Cell movement of leukocytes" (P < 0.05) functions (Z-score = -2.2, -2.2 and -2.0, respectively) in DG, and inhibited "Concentration of lipid" and "Proliferation of epithelial cells" functions in UG (Z-score = -2.0, and -1.8, respectively). In addition, the expression of genes involved in ketogenesis (HMGCL) and lipid synthesis (SREBF2, FABP4) was increased in DG, while the expression of ketogenesis (ACAT2, HMGCS) and cholesterol synthesis related genes (HMGC and FDPS) were deceased in UG. Furthermore, the upstream regulators were found to be involved in the regulation of immune response and cell cycle progress, and SNP (g.46834311A > G) in FABP4 was identified between two groups of animals (P < 0.1).ConclusionThe identified genes, upstream regulators, and SNP could be potential genetic markers that may account for the varied individual ruminal pH responses to the dietary transition stress.
