The complete mitochondrial genome sequence of Dodona eugenes (Lepidoptera: Riodinidae).

ABSTRACT: The complete mitochondrial genome (mitogenome) sequence of Dodona eugenes (Lepidoptera: Riodinidae) was determined and analyzed. The mitogenome is 15,680 bp in length with consisting of 13 protein-coding genes (PCGs), 22 transfer RNA (tRNA) genes, two ribosomal RNA genes (rrnL and rrnS), and one AT-rich region. The gene content, orientation, and order are identical to that of the majority of other lepidopteran insects. The D. eugenes mitogenome includes a cox1 gene with an atypical CGA(R) start codon and three genes (cox1, nad5, and nad4) exhibiting incomplete stop codons. All tRNAs have a typical secondary cloverleaf structure, except for trnS1 which lacks the dihydrouridine (DHU) arm. The 825-bp long AT-rich region is the longest among sequenced riodinids, which range from 349 to 423 bp. The conclusion of phylogenetic analysis highly supported the monophyly of Riodinidae, which is standing as the sister of the family Lycaenidae.

Project description:Taxonomic status and phylogenetic position of some skippers within Hesperiidae remains a controversial issue, here, we sequenced and analyzed the complete mitochondrial genome (mitogenome) of Abraximorpha davidii, one of species in Hesperiidae. This mitogenome is 15,469 bp long and encodes 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), and two ribosomal RNA unit genes (rRNAs). The overall base composition of the mitogenome is A 40.2%, T 41.4%, C 11.2%, and G 7.2%, with a high A + T content of 81.6%. Except for cox1 starting with CGA, all other PCGs start with the standard ATN codons (seven ATG and five ATT). Most of the PCGs terminate with the stop codon TAA, whereas cox1, cox2, nad5, and nad4 end with the incomplete codon T-. Phylogenetic analysis showed that A. davidii is closely related to Daimio tethys and Tagiades vajuna, then this clade clusters Ctenoptilum vasava and Celaenorrhinus maculosa.

Characterization of the complete mitochondrial genome sequence of Smerinthus caecus (Lepidoptera: Sphingidae).

Project description:In this study, we sequenced and analyzed the complete mitogenome of Smerinthus caecus Ménétriés, 1857. The mitogenome of S. caecus is a circular structure, and 15,363 bp long in size and encodes 13 protein-coding genes (PCGs), two ribosomal RNA genes (rRNAs), 22 transfer RNA genes (tRNAs), and a control region (CR). An extremely high AT bias of 79.2% was found in the nucleotide composition of mitogenome. Most of the PCGs used ATN as the start codon and TAA or TAG as the stop codon, which is similar to most other insect mitogenomes, except cox1, which starts with CGA. The phylogeny of Smerinthinae was reconstructed using a maximum-likelihood method, a total of 33 mitogenomes were sampled for phylogenetic analyses. The subfamily Langiinae was selected as outgroup. The results confirmed the position of S. caecus in the Smerinthinae, in which Smerinthus caecus was placed as the sister taxon to Smerinthus planus, then to Laothoe amurensis.

Project description:The complete mitochondrial genome (mitogenome) of Aglossa dimidiata (Lepidoptera: Pyralidae) was sequenced using high-throughput sequencing. The mitogenome of A. dimidiata was 15,225 bp in length. It comprised 37 typical genes and one control region. All protein-coding genes (PCGs) were initiated with ATN, except for COX1 (TTG). All PCGs used TAN as stop codon, except for ND5 and ND4 terminated with incomplete T. Twenty-two tRNA genes ranged from 61 to 71 bp in size. The monophyly of family Pyralidae and the sister relationship between A. dimidiata and Orthopygia glanucinalis are both supported by maximum likelihood method using the nucleotide sequences of 13 PCGs.

The complete mitogenome sequence of T. oldenlandiae was observed to be a circular molecule 15,312 bp long and consisting of 13 protein-coding genes (PCG), 2 ribosomal RNA (rRNA) genes, and 22 transfer RNA (tRNA) genes (GenBank accession number MN885801). The nucleotide composition is biased toward adenine and thymine (80.0% A + T). The A + T-rich region was found between rrnS and trnM, and this entire region was 423 bp long.

Project description:The complete mitochondrial genome of the fruit piercing moth Eudocima phalonia (Linnaeus, 1763) (Lepidoptera: Noctuoidea) was sequenced and characterized (Genbank Accession No: KY196412). The complete mitogenome is a circular molecule of 15,575 bp length, consisting of 13 protein-coding genes (PCGs), two ribosomal RNA genes (rrnS and rrnL), 22 transfer RNA (tRNA) genes and an A + T-rich region (D-loop). The nucleotide composition of the genome is highly A + T biased, accounting for 80.67% of nucleotides. All tRNAs have putative secondary structures that are characteristic of mitochondrial tRNA. Most of the PCGs were initiated by typical ATN codons. Five genes were initiated by unusual codons. Cox1 gene was initiated by an unusual CGA codon and terminated by the typical stop codon GAA. Six genes ended with a single T. The A + T-rich region of 336 bp consisted of repetitive sequences, including two ATAGA motifs, a 19 bp poly-T stretch and three microsatellite-like regions ((TA)4, (TA)6 and two (TA)7). Moreover, three large tandem (one 40 bp and two 25 bp) repeated elements were identified in A + T-rich region. Phylogenetic analysis using PCGs revealed that Superfamily Noctuoidea is a monophyletic group.

The mitogenome is a DNA molecule of 15125 bp, and consists of 13 protein-coding genes (PCGs), 22 transfer RNAs, 2 rRNAs, and 1 A-T rich region. The nucleotide composition biases toward A and T is 79.4% of the entirety, which is a typical structure of Lepidopterans. All PCGs started with ATN, except cox1, which started with CGA, 8 PCGs stop with TAN and 5 genes exhibited incomplete stop codon. Phylogenetic analysis revealed that Satyrinae is a monophyletic group and Coenonympha (C. amaryllis) as sister of the genus Triphysa (T. phryne).

The complete mitochondrial genome of Rondotia menciana (Lepidoptera: Bombycidae).

Project description:The mulberry white caterpillar, Rondotia menciana Moore (Lepidoptera: Bombycidae) is a species with closest relationship with Bombyx mori and Bombyx mandarina, and the genetic information of R. menciana is important for understanding the diversity of the Bombycidae. In this study, the mitochondrial genome (mitogenome) of R. menciana was amplified by polymerase chain reaction and sequenced. The mitogenome of R. menciana was determined to be 15,301 bp, including 13 protein-coding genes (PCGs), 2 ribosomal RNA genes, 22 transfer RNA genes, and an AT-rich region. The A+T content (78.87%) was lower than that observed for other Bombycidae insects. All PCGs were initiated by ATN codons and terminated with the canonical stop codons, except for coxII, which was terminated by a single T. All the tRNA genes displayed a typical clover-leaf structure of mitochondrial tRNA. The length of AT-rich region (360 bp) of R. menciana mitogenome is shorter than that of other Bombycidae species. Phylogenetic analysis showed that the R. menciana was clustered on one branch with B. mori and B. mandarina from Bombycidae.
