ABSTRACT: We report the discovery of a dead Magellanic penguin (Spheniscus magellanicus) found on Juquehy Beach (23°46'S 45°44'W), municipality of São Sebastião, Brazil, on September 9th 2020. Following necropsy, we noted the presence of an adult size PFF-2 protective mask within the stomach of the penguin which we inferred as the cause of death. As far as we are aware, this is the first recorded instance of marine animal mortality by protective face mask ingestion. Whilst concerns have been raised relating microplastic contamination in marine environments from Covid-19 related waste, there has been relatively less attention paid to the potential risk of macro-scale contaminants, such as protective face coverings. We suggest that Covid-19 related macro contaminants be considered in coastal marine risk assessments and urge further research on this topic.

Project description:Enterococci are commensals that proliferated as animals crawled ashore hundreds of millions of years ago. They are also leading causes of multidrug-resistant hospital-acquired infections. While most studies are driven by clinical interest, comparatively little is known about enterococci in the wild or the effect of human activity on them. Pharmaceutical pollution and runoff from other human activities are encroaching widely into natural habitats. To assess their reach into remote habitats, we investigated the identity, genetic relatedness, and presence of specific traits among 172 enterococcal isolates from wild Magellanic penguins. Four enterococcal species, 18 lineage groups, and different colonization patterns were identified. One Enterococcus faecalis lineage, sequence type 475 (ST475), was isolated from three different penguins, making it of special interest. Its genome was compared to those of other E. faecalis sequence types (ST116 and ST242) recovered from Magellanic penguins, as well as to an existing phylogeny of E. faecalis isolated from diverse origins over the past 100 years. No penguin-derived E. faecalis strains were closely related to dominant clinical lineages. Most possessed intact CRISPR defenses, few mobile elements, and antibiotic resistances limited to those intrinsic to the species and lacked pathogenic features conveyed by mobile elements. Interestingly, plasmids were identified in penguin isolates that also had been reported for other marine mammals. Enterococci isolated from penguins showed limited anthropogenic impact, indicating that they are likely representative of those naturally circulating in the ecosystem inhabited by the penguins. These findings establish an important baseline for detecting the encroachment of human activity into remote planetary environments.IMPORTANCE Enterococci are host-associated microbes that have an unusually broad range, from the built hospital environment to the guts of insects and other animals in remote locations. Despite their occurrence in the guts of animals for hundreds of millions of years, we know little about the properties that confer this range or how anthropogenic activities may be introducing new selective forces. Magellanic penguins live at the periphery of human habitation. It was of interest to examine enterococci from these animals for the presence of antibiotic resistance and other markers reflective of anthropogenic selection. Diverse enterococcal lineages found discount the existence of a single well-adapted intrinsic penguin-specific species. Instead, they appear to be influenced by a carnivorous lifestyle and enterococci present in the coastal sea life consumed. These results indicate that currently, the penguin habitat remains relatively free of pollutants that select for adaptation to human-derived stressors.

Project description:The main motivation for this study was to determine the occurrence of Toxoplasma gondii, a cosmopolitan widespread zoonotic parasite distribution that can infect a wide variety of mammals and birds, in Magellanic penguins (Spheniscus magellanicus) in Brazil. In recent decades there has been a significant increase in the number of penguins originating from Argentinian and Chilean Patagonia, where these birds are born, that arrive on the Brazilian coast, where many of them are stranded and rescued. Tissue samples were collected from 330 individuals surveyed from 2012-2015 at the Institute for Marine Animal Research and Rehabilitation (IPRAM) located in Cariacica, state of Espirito Santo, Brazil. Serum were collected from 145 animals surveyed in 2015 for the detection of anti-T. gondii antibodies using the Modified Agglutination Test (MAT ≥20) and 18 birds were positive, with titers of 20 (7 birds), 40 (9 birds) and 80 (2 birds). Mouse bioassay for the isolation of T. gondii was performed using tissues from 54 penguins that were also surveyed in 2015, but no isolates were obtained. DNA from tissue samples of 330 individuals was PCR amplified and sequenced to detect tissue cyst forming coccidians by using pan sarcocystids-directed primers (based on 18S rDNA). These samples were from animals surveyed in 2015 and from frozen stocked tissues from animals surveyed in the years 2012 and 2013. The positives were PCR amplified and sequenced with genus Sarcocystis-specific primers (based on internal transcribed spacer 1, RNA polymerase beta subunit coding gene, and cytochrome B coding gene) and with Sarcocystis falcatula/Sarcocystis neurona- specific primers (based on surface antigens SAG2, SAG3 and SAG4). Sixteen (3.0%) of pectoral muscle samples were positive by all the seven molecular markers and all the samples were identical to each other. Organisms close related to Sarcocystis falcatula were confirmed in all cases. This is the first report on molecular detection of infection by S. falcatula-related organisms and the first report of seropositivity for T. gondii in free-living Magellanic penguins in Brazil. Felids and didephid opossums are definitive hosts of T. gondii and S. falcatula, respectively. Where the penguins acquire the infective forms of the parasites shed by the terrestrial mammals remains to be elucidated.

Project description:Growing animals should allocate their limited resources in ways that maximize survival. Seabird chicks must balance the growth of features and fat reserves needed to survive on land with those needed to successfully fledge and survive at sea. We used a large, 34-year dataset to examine energy allocation in Magellanic penguin chicks. Based on the temporal trends in the selective pressures that chicks faced, we developed predictions relating to the timing of skeletal feature growth (Prediction 1), variation in skeletal feature size and shape (Prediction 2), and responses to periods of high energetic constraint (Prediction 3). We tested our predictions using descriptive statistics, generalized additive models, and principal component analysis. Nearly all of our predictions were supported. Chicks grew their feet first, then their flippers. They continued to grow their bill after fledging (Prediction 1). Variance in feature size increased in young chicks but declined before fledging; this variance was largely driven by overall size rather than by shape (Prediction 2). Chicks that died grew slower and varied more in feature size than those that fledged (Prediction 2). Skeletal features grew rapidly prior to thermoregulation and feet and flippers were 90% grown prior to juvenile feather growth; both thermoregulation and feather growth are energetically expensive (Prediction 3). To avoid starvation, chicks prioritized storing mass during the first 10 days after hatching; then, the body condition of chicks began to decline (Prediction 3). In contrast to our prediction of mass prioritization in young chicks, chicks that were relatively light for their age had high skeletal size to mass ratios. Chicks did not show evidence of reaching physiological growth limits (Prediction 3). By examining energy allocation patterns at fine temporal scales and in the context of detailed natural history data, we provide insight into the trade-offs faced by growing animals.
