Project description:BackgroundNitrate is a common water contaminant that has been associated with birth defects, although the evidence is limited. The purpose of this study was to examine whether maternal consumption of nitrate through drinking water is associated with an increased risk of congenital anomalies.MethodsThe study included a total of 348,250 singletons births from the state of Missouri between January 1, 2004 and December 31, 2008. Individual-level birth defect data and maternal and child characteristics were obtained from the Missouri birth defects registry and state vital statistics records. Outcomes were linked with county-specific monthly estimates of the nitrate concentration in finished water, based on data collected for compliance with the Safe Drinking Water Standard. Poisson models were fit to examine the association between nitrate exposure and birth defects. Average nitrate exposure during the first trimester and over 12 months prior to birth were modeled as continuous variables. Sensitivity analyses included restriction of the sample to counties with <20% and <10% private well usage to reduce exposure misclassification as well as limiting the analyses to residents of rural counties only to account for potential confounding by urbanicity.ResultsEstimated water concentrations of nitrate were generally low and below the Environmental Protection Agency's maximum contaminant level of 10 mg/L. Nitrate exposure was associated with a significantly increased risk of limb deficiencies (RR for 1 mg/L (RR1) = 1.26, 95% CI = 1.05, 1.51) in models without well restriction. Nitrate was also weakly associated with an increased risk of congenital heart defects (RR1 = 1.13, 95%CI = 0.93, 1.51) and neural tube defects (RR1 = 1.18, 95%CI = 0.93, 1.51) in models with well restriction (<10%).ConclusionThe positive associations found between nitrate exposure via drinking water and congenital abnormalities are largely consistent with some previous epidemiologic studies. The results of this study should be interpreted with caution given limitations in our ability to estimate exposures and the lack information on some risk factors for congenital abnormalities. Our findings may have serious policy implications given that exposure levels in our study were well below current EPA standards for nitrate in drinking water.

Project description:The main objective of this data was determination of the nitrate concentration and its health risk assessment in the drinking water resources. In the aim of this data article a number of 30 samples of nitrate concentration in drinking water resources were collected from villages of the Khash city, Sistan and Baluchistan province, Iran. The nitrate concentration was measured using a Spectrophotometer accordance standard methods for examination of water and wastewater. Data indicated that nitrate concentration in drinking water ranged from 6 to 35 mg/L (average 16.083 mg/L). The mean EDI values for nitrate in different groups of infants, children, teenagers and adults were 0.1287, 0.9114, 0.6433 and 0.5155 mg/Kg, respectively. The findings of data showed that HQ value was less than 1 in 96% of samples in age groups of infants, children, teenagers and adults.

Project description:This study aims to develop a software framework for predicting aridity using vegetation indices (VI) from LANDSAT 8 OLI images. VI data are predicted using machine learning (ml): Random Forest (RF) and Correlation and Regression Trees (CART). Comparison of prediction using Artificial Neural Network (ANN), Support Vector Machine (SVM), k-nearest neighbors (k-nn) and Multivariate Adaptive Regression Spline (MARS). Prediction results are interpolated using Inverse Distance Weight (IDW). This study was conducted in stages: (1) Image preprocessing; (2) calculating numerical data extracted from the LANDSAT band imagery using vegetation indices; (3) analyzing correlation coefficients between VI; (4) prediction using RF and CART; (5) comparing performances between RF and CART using ANN, SVM, k-nn, and MARS; (6) testing the accuracy of prediction using Mean Square Error (MSE) and Mean Absolute Percentage Error (MAPE); (7) interpolating with IDW. Correlation coefficient of VI data shows a positive correlation, the lowest r (0.07) and the highest r (0.98). The experiments show that the RF and CART algorithms have efficiency and effectivity in determining the aridity areas better than the ANN, SVM, k-nn, and MARS algorithm. RF has a difference between the predicted results and 1.04% survey data MAPE and the smallest value close to zero is 0.05 MSE. CART has a difference between the predicted results and 1.05% survey data MAPE and the smallest value approaching to zero which is 0.05 MSE. The prediction results of VI show that in 2020 most of the study areas were low vegetation areas with the Normalized Difference Vegetation Index (NDVI) < 0.21, had an indication of drought with the Vegetation Health Index (VHI) < 31.10, had a Vegetation Condition Index (VCI) in some areas between 35%-50% (moderate drought) and < 35% (high drought). The Burn Area Index (dBAI) values are between -3, 971 and -2,376 that show the areas have a low fire risk, and index values are between -0, 208 and -0,412 that show the areas are starting vegetation growth. The result of this study shows that the machine learning algorithms is an accurate and stable algorithm in predicting the risks of drought and land fire based on the VI data extracted from the LANDSAT 8 OLL imagery. The VI data contain the record of vegetation condition and its environment, including humidity, temperatures, and the environmental vegetation health.

Project description:Evidence is emerging that preterm birth (PTB, birth before 37 completed weeks of gestation), a risk factor for neonatal mortality and future morbidity, may be induced by maternal nitrate ( NO3- ) exposure from drinking water. The objective of this study is to assess the association between maternal exposure to nitrate and the risk of PTB in a nationwide study of liveborn singletons.MethodsWe estimated maternal nitrate exposure from household tap water for 1,055,584 births in Denmark to Danish-born parents during 1991-2015 by linkage of individual home address(es) with nitrate concentrations from a national monitoring database. Nitrate exposure during pregnancy was modeled using four categories and continuously. Logistic models adjusted for sex, birth year, birth order, urbanicity, and maternal age, smoking, education, income, and employment, with generalized estimating equations were used to account for sibling clusters.ResultsA total of 1,009,189 births were included, comprising 51,747 PTB. An increase in the risk of PTB was seen across categories of exposure (P < 0.001) with an odds ratio (OR) in the uppermost category (>25 mg/L nitrate) of 1.05 (95% confidence interval [CI] = 1.00, 1.10). Evidence of an exposure-response relationship was observed in models using continuous nitrate (OR = 1.01 [95% CI = 1.00, 1.03] per 10 mg/L nitrate). In sensitivity analyses, results were robust to the addition of variables for short inter-pregnancy interval (<1 year between births), maternal pre-pregnancy body mass index, paternal socioeconomic status and age, season of birth, and inclusion of post-term births. Results were virtually unchanged when the analysis was restricted to women exposed to less than the current European Union standard of 50 mg/L.ConclusionWe observed an increasing risk of PTB with increases in nitrate in household tap water. These findings add to a growing body of evidence of adverse effects from nitrate in drinking water at levels below current regulatory levels.

Project description:Soil-transmitted helminth (STH) infections are endemic in Indonesia. However, prevalence data for many parts of the country are incomplete. The aim of this study was to determine human STH prevalence and knowledge and practices relating to STH risk behaviour, to provide a current view of the status of STH infection in rural communities in Central Java. A cross-sectional survey of 16 villages was conducted in Semarang, Central Java in 2015. Demographic and household data together with information about knowledge and practices relating to STH and hygiene were elicited through face-to-face interviews. Stool samples were collected and examined using the flotation method. Children (aged 2-12 years) also had their haemoglobin (Hb) levels, height and weight data collected, and BMI estimated. Data were analysed using univariate logistic regression analysis. A total of 6,466 individuals with a mean age of 33.5 years (range: 2-93) from 2,195 households were interviewed. The overall prevalence of STH was 33.8% with Ascaris lumbricoides (roundworm) the predominant nematode identified (prevalence = 26.0%). Hookworm and Trichuris trichiura (whipworm) were found in 7.9% and 1.8% of participants, respectively. Females were at increased odds of infection with A. lumbricoides (adjusted OR 1.14, 95% CI [1.02-1.29], p = 0.02). Adults in age groups 51-60 and over 60 years had the highest odds of being infected with hookworm (adjusted OR 3.01, 95% CI [1.84-4.91], p<0.001 and adjusted OR 3.79, 95% CI [2.30-6.26], p<0.001, respectively) compared to 6-12 year olds. Farmers also had higher odds of being infected with hookworm (adjusted OR 2.36, 95% CI [1.17-4.76], p = 0.02) compared to other occupation categories. Poverty (OR 2.14, 95% CI [1.77-2.58], p<0.001), overcrowding (OR 1.35, 95% CI [1.27-1.44], p<0.001), goat ownership (OR 1.61, 95% CI [1.10-2.41], p = 0.02) and the presence of dry floor space in the home (OR 0.73, 95% CI [0.58-0.91], p = 0.01) were all household factors significantly associated with an increased odds of infection. Infection with STH was not significantly associated with the gastrointestinal illness (p>0.05), BMI or Hb levels; however, one third of all 2-12 year olds surveyed were found to be anaemic (i.e. Hb concentrations below 110g/l or 115g/l for children under 5 and 5 years or older, respectively), with a greater proportion of school-age children at risk. Knowledge and behaviour related to hygiene and gastrointestinal diseases varied widely and were generally not associated with STH infection. The study revealed that STH infection remains endemic in Central Java despite ongoing deworming programs. Current control efforts would benefit from being re-evaluated to determine a more effective way forward.

Project description:Background and aimActinomycetes are a group of Gram-positive bacteria with a fungus-like morphology. Their natural habitat encompasses terrestrial and water areas, including mangrove ecosystems. This study aimed to assess the PKS and NRPS genes as the producers of secondary metabolites and to determine the target bacterial species using molecular DNA tests.Materials and methodsIn this study, we isolated bacteria from sediment samples from mangrove forests located on Karimunjawa Islands and in Semarang city, purified bacteria, screened for antibacterial activity, extracted bacterial DNA, amplified the NRPS gene, detected and amplified the PKS-I and PKS-II genes, amplified and sequenced the 16S rRNA, processed molecular data, and simulated a map of secondary metabolite producing genes.ResultsSamples from the Karimunjawa Islands yielded 19 bacterial isolates, whereas samples from Semarang yielded 11 bacterial isolates after culture in different media. Further experiments identified three active isolates, which were termed PN.SB.6.2, S.SK.6.3, and S.SK.7.1, against pathogenic species of Escherichia coli, Staphylococcus aureus, and Listeria monocytogenes. Isolate PN.SB.6.2 was determined to possess three biosynthetic gene clusters (BGCs), whereas the remaining two isolates, S.SK.6.3 and S.SK.7.1, only possessed two BGCs, namely, NRPS and PKS II.ConclusionProducts were estimated to be in the NRPS, thiopeptide, RiPP-like, siderophore, betalactone, terpene, Type III PKS, CDPS, and lassopeptide groups. DNA identification of the isolates found three species of actinomycetes with antibacterial potential, namely, Virgibacillus salaries, Bacillus licheniformis, and Priestia flexa.

Project description:BackgroundNitrate contamination is seen in drinking water worldwide. Nitrate may pass the placental barrier. Despite suggestive evidence of fetal harm, the potential association between nitrate exposure from drinking water and pregnancy loss remains to be studied. We aimed to investigate if nitrate in drinking water was associated with the risk of pregnancy loss.MethodsWe conducted a nationwide cohort study of 100,410 pregnancies (enrolled around gestational week 11) in the Danish National Birth Cohort (DNBC) during 1996-2002. Spontaneous pregnancy losses before gestational week 22 were ascertained from the Danish National Patient Registry and DNBC pregnancy interviews. Using the national drinking water quality-monitoring database Jupiter, we estimated the individual and time-specific nitrate exposure by linking geocoded maternal residential addresses with water supply areas. The nitrate exposure was analyzed in spline models using a log-transformed continuous level or classified into five categories. We used Cox proportional hazards models to estimate associations between nitrate and pregnancy loss and used gestational age (days) as the time scale, adjusting for demographic, health, and lifestyle variables.ResultsNo consistent associations were found when investigating the exposure as a categorical variable and null findings were also found in trimester specific analyses. In the spline model using the continuous exposure variable, a modestly increased hazard of pregnancy loss was observed for the first trimester at nitrate exposures between 1 and 10 mg/L, with the highest. adjusted hazard ratio at 5 mg/L of nitrate of 1.16 (95% CI: 1.01, 1.34). This trend was attenuated in the higher exposure ranges.ConclusionNo association was seen between drinking water nitrate and the risk of pregnancy loss when investigating the exposure as a categorical variable. When we modelled the exposure as a continuous variable, a dose-dependent association was found between drinking water nitrate exposure in the first trimester and the risk of pregnancy loss. Very early pregnancy losses were not considered in this study, and whether survival bias influenced the results should be further explored.

Project description:In 2011, the United Nations Environment Programme discovered high levels of hydrocarbon pollution in drinking water wells in Nsisioken Agbi Ogale Rivers State, Nigeria. However, the level of polycyclic aromatic hydrocarbons (PAHs) in the same community's water supply was unknown. A comprehensive study of PAHs in three household dug wells and three boreholes was conducted using Agilent 7890B gas chromatography and 5975A mass spectrometry. The detected PAHs were mainly 4 - 5 ringed PAHs, such as Chrysene, Fluoranthene, Pyrene, Benzo[a]anthracene, and Benzo[b]fluoranthene. The total mean concentration was 5.8 ± 2.3 μg/L, with values ranging from not detected in borehole 3 to 8.0 μg/L at well 2. Source identification analysis suggested that the PAHs originated from fuel and biomass combustion. The incremental lifetime cancer risk for children and adults due to groundwater ingestion and skin contact ranged from ND to 7.448 × 10-3 and ND to 1.83 × 10-3 respectively. The Risk index (RI) values from ingestion and dermal routes were 1.5 ×10-2 and 2.4 × 10-2, indicating high risk of cancer. The hazard quotient for the two non-carcinogenic PAHs was greater than 1, indicating high toxicity. The PAH concentrations exceeded the maximum contamination limits set by the World Health Organization and the U.S. environmental Protection Agency, highlighting potential health risks associated with water use in the community. Authorities should provide a safe alternative water source for the community.

Project description:Due to the large amounts of pharmaceuticals and personal care products (PPCPs) currently being consumed and released into the environment, this study provides a comprehensive analysis of pharmaceutical pollution in both raw and treated water from full-scale drinking water treatment plants nationwide. Our investigation revealed that 30 out of 37 PPCPs were present in raw water with mean concentrations ranging from 0.01-131 ng/L. The raw water sources, surface water (ND - 147 ng/L), subsurface water (ND - 123 ng/L) and reservoir sources (ND - 135 ng/L) exhibited higher mean concentration levels of pharmaceutical residues compared to groundwater sources (ND - 1.89 ng/L). Meanwhile, in treated water, 17 of the 37 analyzed PPCPs were present with carbamazepine, clarithromycin, fluconazole, telmisartan, valsartan, and cotinine being the most common (detection frequency > 40 %), and having mean concentrations of 1.22, 0.12, 3.48, 40.1, 6.36, and 3.73 ng/L, respectively. These findings highlight that, while water treatment processes are effective, there are some persistent compounds that prove challenging to fully eliminate. Using Monte Carlo simulations, risk assessment indicated that most of these compounds are likely to have negligible impact on human health, except for the antihypertensives. Telmisartan was identified as posing the highest ecological risk (RQ > 1), warranting further investigation, and monitoring. The study concludes by prioritizing specific 14 pharmaceuticals, including telmisartan, clarithromycin, lamotrigine, cotinine, lidocaine, tramadol, and others, for future monitoring to safeguard both ecological and human health.

Project description:BackgroundHealth issues have been associated with the consumption of high levels of nitrates in drinking water. Rural agricultural communities in Belize play a large role in the economic growth of the country. These communities obtain drinking water directly from the ground and may be susceptible to nitrate consumption and at risk of developing diseases associated with nitrates.ObjectivesThe present study examined nitrate levels in Belize's rural water supply with the aim of assessing its suitability for human and livestock consumption. The study also provides baseline data for monitoring the concentration of nitrates to prevent public health hazards in Belize.MethodsForty-three (43) water samples from reservoirs, wells, vats, and standpipes were collected from 40 villages in Belize and analyzed for nitrates using the cadmium reduction method. Nitrates were detected with an Orion® AquaMate® 8000 UV-Vis spectrophotometer at 520 nm. The Belize Coastal Zone Management Authority and Institute laboratory analyzed all water samples for nitrates.ResultsExcept for four water samples from four different sites, all analyzed water samples were found to contain nitrate levels below 10 mg/L. Nitrate levels above 10 mg/L were seen in a few samples in the northern part of the country, probably due to agricultural activities in these areas.ConclusionsBelize's rural drinking water contains low levels of nitrates, except for a few villages where the levels exceeded the acceptable limit of 10 mg/L. Higher levels of nitrates detected in a few villages need regular evaluation and monitoring to avoid public health issues as well as prevent harm to livestock.Competing interestsThe authors declare no competing financial interests.
