Emerging anthropogenic circularity science: principles, practices, and challenges.

ABSTRACT: Material depletion over reliance of linear economies and environmental pollution may be resolved by applying the principles and practices of anthropogenic circularity science. Here we systematically review the emergence of anthropogenic circularity science in the interdisciplinary development of green chemistry, supply chain, and industrial ecology at different scales. The first, second, and third laws of circularity chemistry are proposed as forming the basic principles of circularity science. To close the loop on critical materials, these three basic principles have been exemplified in the anthropogenic circularity practices. We highlight the spatial distribution of critical metal, waste generation, and recycling rate. Future opportunities and challenges for a circular economy and urban mining will predominate in anthropogenic circularity. Therefore, anthropogenic circularity science will play an increasing role in enabling a smooth transition to a circular society.

Do biodiversity monitoring citizen science surveys meet the core principles of open science practices?

Project description:Citizen science (CS), as an enabler of open science (OS) practices, is a low-cost and accessible method for data collection in biodiversity monitoring, which can empower and educate the public both on scientific research priorities and on environmental change. Where OS increases research transparency and scientific democratisation; if properly implemented, CS should do the same. Here, we present the findings of a systematic review exploring "openness" of CS in biodiversity monitoring. CS projects were scored between - 1 (closed) and 1 (open) on their adherence to defined OS principles: accessible data, code, software, publication, data management plans, and preregistrations. Openness scores per principle were compared to see where OS is more frequently utilised across the research process. The relationship between interest in CS and openness within the practice was also tested. Overall, CS projects had an average open score of 0.14. There was a significant difference in open scores between OS principles (p = < 0.0001), where "open data" was the most adhered to practice compared to the lowest scores found in relation to preregistrations. The apparent level of interest in CS was not shown to correspond to a significant increase in openness within CS (p = 0.8464). These results reveal CS is not generally "open" despite being an OS approach, with implications for how the public can interact with the research that they play an active role in contributing to. The development of systematic recommendations on where and how OS can be implemented across the research process in citizen science projects is encouraged.

Pulmonary Arterial Hypertension: Emerging Principles of Precision Medicine across Basic Science to Clinical Practice.

Project description:Pulmonary arterial hypertension (PAH) is an enigmatic and deadly vascular disease with no known cure. Recent years have seen rapid advances in our understanding of the molecular underpinnings of PAH, with an expanding knowledge of the molecular, cellular, and systems-level drivers of disease that are being translated into novel therapeutic modalities. Simultaneous advances in clinical technology have led to a growing list of tools with potential application to diagnosis and phenotyping. Guided by fundamental biology, these developments hold the potential to usher in a new era of personalized medicine in PAH with broad implications for patient management and great promise for improved outcomes.

Emerging challenges and opportunities in innovating food science technology and engineering education.

Project description:Progress in science, technology, innovation, and digital capabilities call for reassessing food science, technology, and engineering (FST&E) education and research programs. This survey targeted global professionals and students across food disciplines and nutrition. Its main objectives included assessing the status of FST&E higher education, identifying challenges and opportunities, and furnishing recommendations. Seven topics affecting the future of the FST&E curricula were evaluated by the panel as 'High' to 'Very high', namely: 'Critical thinking', followed by 'Problem-solving projects', 'Teamwork/collaboration', 'Innovation/Open innovation' and 'Multidisciplinary'. The importance of academic partnership/collaboration with the Food Industry and Nutrition Sciences was demonstrated. Significant positive roles of the food industry in collaboration and partnerships were found. Other essential food industry attributes were related to internships, education, strategy, and vision. Collaboration between FST&E and nutrition sciences indicated the high standing of this direction. The need to integrate or converge nutrition sciences and FST&E is emphasized, especially with the growing consumer awareness of health and wellness. The study provides insights into new education and learning opportunities and new topics for future curricula.

FRET-based dynamic structural biology: Challenges, perspectives and an appeal for open-science practices.

Project description:Single-molecule FRET (smFRET) has become a mainstream technique for studying biomolecular structural dynamics. The rapid and wide adoption of smFRET experiments by an ever-increasing number of groups has generated significant progress in sample preparation, measurement procedures, data analysis, algorithms and documentation. Several labs that employ smFRET approaches have joined forces to inform the smFRET community about streamlining how to perform experiments and analyze results for obtaining quantitative information on biomolecular structure and dynamics. The recent efforts include blind tests to assess the accuracy and the precision of smFRET experiments among different labs using various procedures. These multi-lab studies have led to the development of smFRET procedures and documentation, which are important when submitting entries into the archiving system for integrative structure models, PDB-Dev. This position paper describes the current 'state of the art' from different perspectives, points to unresolved methodological issues for quantitative structural studies, provides a set of 'soft recommendations' about which an emerging consensus exists, and lists openly available resources for newcomers and seasoned practitioners. To make further progress, we strongly encourage 'open science' practices.

Bridging the Science Practices Gap: Analyzing Laboratory Materials for Their Opportunities for Engagement in Science Practices.

Project description:Laboratory courses provide a unique environment for students to engage in the practices of scientists and develop the skills necessary for a career in science. Research has indicated that inquiry-based laboratory activities, which allow students to make key decisions throughout the laboratory experiment, provide opportunities to develop these necessary skills and practices. This study focuses on characterizing laboratory experiments used in analytical chemistry courses for their level of inquiry and opportunities for engagement in scientific practices. Data for this analysis was collected from participants of the MICRO project, a project designed to assist instructors in adopting inquiry-based laboratory experiments. Laboratory experiments used by instructors prior to their involvement in the project and then during the semester of implementation of the inquiry-based laboratory experiments were analyzed for their level of inquiry and opportunities for engagement in scientific practices. We found that, despite an increase in the level of inquiry of laboratory materials, there was not an increase in prompted opportunities to engage students in all of the science practices. We also found that certain science practices were more likely to be prompted in certain sections of the laboratory materials, such as through the pre-laboratory and post-laboratory questions.

Genetically engineered bacterium: Principles, practices, and prospects.

Project description:Advances in synthetic biology and the clinical application of bacteriotherapy enable the use of genetically engineered bacteria (GEB) to combat various diseases. GEB act as a small 'machine factory' in the intestine or other tissues to continuously produce heterologous proteins or molecular compounds and, thus, diagnose or cure disease or work as an adjuvant reagent for disease treatment by regulating the immune system. Although the achievements of GEBs in the treatment or adjuvant therapy of diseases are promising, the practical implementation of this new therapeutic modality remains a grand challenge, especially at the initial stage. In this review, we introduce the development of GEBs and their advantages in disease management, summarize the latest research advances in microbial genetic techniques, and discuss their administration routes, performance indicators and the limitations of GEBs used as platforms for disease management. We also present several examples of GEB applications in the treatment of cancers and metabolic diseases and further highlight their great potential for clinical application in the near future.

Project description:Whole-cell computational models aim to predict cellular phenotypes from genotype by representing the entire genome, the structure and concentration of each molecular species, each molecular interaction, and the extracellular environment. Whole-cell models have great potential to transform bioscience, bioengineering, and medicine. However, numerous challenges remain to achieve whole-cell models. Nevertheless, researchers are beginning to leverage recent progress in measurement technology, bioinformatics, data sharing, rule-based modeling, and multi-algorithmic simulation to build the first whole-cell models. We anticipate that ongoing efforts to develop scalable whole-cell modeling tools will enable dramatically more comprehensive and more accurate models, including models of human cells.

Emerging principles in protease-based drug discovery.

Project description:Proteases have an important role in many signalling pathways, and represent potential drug targets for diseases ranging from cardiovascular disorders to cancer, as well as for combating many parasites and viruses. Although inhibitors of well-established protease targets such as angiotensin-converting enzyme and HIV protease have shown substantial therapeutic success, developing drugs for new protease targets has proved challenging in recent years. This in part could be due to issues such as the difficulty of achieving selectivity when targeting protease active sites. This Perspective discusses the general principles in protease-based drug discovery, highlighting the lessons learned and the emerging strategies, such as targeting allosteric sites, which could help harness the therapeutic potential of new protease targets.

Looking lively: emerging principles of pseudokinase signaling.

Project description:Progress towards understanding catalytically 'dead' protein kinases - pseudokinases - in biology and disease has hastened over the past decade. An especially lively area for structural biology, pseudokinases appear to be strikingly similar to their kinase relatives, despite lacking key catalytic residues. Distinct active- and inactive-like conformation states, which are crucial for regulating bona fide protein kinases, are conserved in pseudokinases and appear to be essential for function. We discuss recent structural data on conformational transitions and nucleotide binding by pseudokinases, from which some common principles emerge. In both pseudokinases and bona fide kinases, a conformational toggle appears to control the ability to interact with signaling effectors. We also discuss how biasing this conformational toggle may provide opportunities to target pseudokinases pharmacologically in disease.
