High-intensity leg cycling alters the molecular response to resistance exercise in the arm muscles.

ABSTRACT: This study examined acute molecular responses to concurrent exercise involving different muscles. Eight men participated in a randomized crossover-trial with two sessions, one where they performed interval cycling followed by upper body resistance exercise (ER-Arm), and one with upper body resistance exercise only (R-Arm). Biopsies were taken from the triceps prior to and immediately, 90- and 180-min following exercise. Immediately after resistance exercise, the elevation in S6K1 activity was smaller and the 4E-BP1:eIF4E interaction greater in ER-Arm, but this acute attenuation disappeared during recovery. The protein synthetic rate in triceps was greater following exercise than at rest, with no difference between trials. The level of PGC-1α1 mRNA increased to greater extent in ER-Arm than R-Arm after 90 min of recovery, as was PGC-1α4 mRNA after both 90 and 180 min. Levels of MuRF-1 mRNA was unchanged in R-Arm, but elevated during recovery in ER-Arm, whereas MAFbx mRNA levels increased slightly in both trials. RNA sequencing in a subgroup of subjects revealed 862 differently expressed genes with ER-Arm versus R-Arm during recovery. These findings suggest that leg cycling prior to arm resistance exercise causes systemic changes that potentiate induction of specific genes in the triceps, without compromising the anabolic response.

Project description:Increases in power output and maximal oxygen consumption ( V˙O2 max) occur in response to sprint interval exercise (SIE), but common use of "all-out" intensities presents a barrier for many adults. Furthermore, lower-body SIE is not feasible for all adults. We compared physiological and perceptual responses to supramaximal, but "non-all-out" SIE between leg and arm cycling exercise. Twenty-four active adults (mean ± SD age: [25 ± 7] y; cycling V˙O2 max: [39 ± 7] mL·kg-1·min-1) performed incremental exercise using leg (LCE) and arm cycle ergometry (ACE) to determine V˙O2 max and maximal work capacity (Wmax). Subsequently, they performed four 20 s bouts of SIE at 130% Wmax on the LCE or ACE at cadence = 120-130 rev/min, with 2 min recovery between intervals. Gas exchange data, heart rate (HR), blood lactate concentration (BLa), rating of perceived exertion (RPE), and affective valence were acquired. Data showed significantly lower (p < 0.001) absolute mean ([1.24 ± 0.31] L·min-1 vs. [1.59 ± 0.34] L·min-1; d = 1.08) and peak V˙O2 ([1.79 ± 0.48] L·min-1 vs. [2.10 ± 0.44] L·min-1; d = 0.70) with ACE versus LCE. However, ACE elicited significantly higher (p < 0.001) relative mean ([62% ± 9%] V˙O2 max vs. [57% ± 7%] V˙O2 max, d = 0.63) and peak V˙O2 ([88% ± 10%] V˙O2 max vs. [75% ± 10%] V˙O2 max, d = 1.33). Post-exercise BLa was significantly higher ([7.0 ± 1.7] mM vs. [5.7 ± 1.5] mM, p = 0.024, d = 0.83) for LCE versus ACE. There was no significant effect of modality on RPE or affective valence (p > 0.42), and lowest affective valence recorded (2.0 ± 1.8) was considered "good to fairly good". Data show that non "all-out" ACE elicits lower absolute but higher relative HR and V˙O2 compared to LCE. Less aversive perceptual responses could make this non-all-out modality feasible for inactive adults.

Project description:AimThe aim was to assess safety and feasibility of Hybrid High-Intensity Interval Training (HIIT) using Functional Electrical Stimulation (FES) leg cycling and arm ski ergometer in people with Spinal Cord Injuries (SCI).MethodEight outpatients (mean age 42.8 years; 7 men) with stable SCI paraplegia (mean 14.5 years since injury) participated in hybrid HIIT (90% peak watts; 4 × 4-min intervals), three times a week (over 8 weeks). Primary outcomes were Adverse Events (AE), participant acceptability, shoulder pain, training intensity (% peak watts), and attendance. Secondary outcomes were effect on peak oxygen uptake (VO2peak) during FES hybrid poling, mean watts, self-reported leisure time physical activity, quality of life, and fatigue.ResultsNo serious AE occurred; acceptability with the training modality was high, while shoulder pain increased by 9% (SD 95.2). During training, 50% of the participants reached > 90% peak watts during the intervals, three with the legs (FES cycle) and one with the arms (Ski-Erg). Overall, mean training intensity (% peak watts) was 92% (SD 18.9) for legs and 82% (SD 10.3) for arms. Proportion of fulfilled training minutes was 82% (range 36-100%); one participant dropped out after 6 weeks due to back pain. Mean VO2peak increased by 17% (SD 17.5). Participants reported increased leisure time physical activity and health-related quality of life, besides reduced fatigue.ConclusionHybrid HIIT was safe for people with SCI paraplegia. The majority of the criteria for feasibility were met with acceptable attendance rate, limited drop out, participants enjoyed training, and increased VO2peak and mean watts. However, the intensity of 90% peak watts was reached by < 60% of the participants despite high RPE ratings during training. The method of measuring and calculating intensity needs to be studied further before a study using this HIIT protocol is undertaken.Trial registrationClinicaltrials.gov , NCT04211311 , registered 12 December 2019 retrospectively registered.

Project description:BACKGROUND:Hepcidin is an iron regulating hormone, and exercise-induced hepcidin elevation is suggested to increase the risk of iron deficiency among athletes. OBJECTIVE:We compared serum hepcidin responses to resistance exercise and endurance (cycling) exercise. METHODS:Ten males [mean ± standard error: 172 ± 2 cm, body weight: 70 ± 2 kg] performed three trials: a resistance exercise trial (RE), an endurance exercise trial (END), and a rest trial (REST). The RE consisted of 60 min of resistance exercise (3-5 sets × 12 repetitions, 8 exercises) at 65% of one repetition maximum, while 60 min of cycling exercise at 65% of [Formula: see text] was performed in the END. Blood samples were collected before exercise and during a 6-h post-exercise (0h, 1h, 2h, 3h, 6h after exercise). RESULTS:Both RE and END significantly increased blood lactate levels, with significantly higher in the RE (P < 0.001). Serum iron levels were significantly elevated immediately after exercise (P < 0.001), with no significant difference between RE and END. Both the RE and END significantly increased serum growth hormone (GH), cortisol, and myoglobin levels (P < 0.01). However, exercise-induced elevations of GH and cortisol were significantly greater in the RE (trial × time: P < 0.001). Plasma interleukin-6 (IL-6) levels were significantly elevated after exercise (P = 0.003), with no significant difference between the trials. Plasma hepcidin levels were elevated after exercise (P < 0.001), with significantly greater in the RE (463 ± 125%) than in the END (137 ± 27%, P = 0.03). During the REST, serum hepcidin and plasma IL-6 levels did not change significantly. CONCLUSION:Resistance exercise caused a greater exercise-induced elevation in hepcidin than did endurance (cycling) exercise. The present findings indicate that caution will be required to avoid iron deficiency even among athletes in strength (power) types of events who are regularly involved in resistance exercise.

Project description:IntroductionThere is a well-established relationship between the respiratory compensation point (RCP) and local muscular breakpoints determined from near-infrared spectroscopy (NIRS) and electromyography (EMG). However, these breakpoints have not yet been compared both in locomotor and non-locomotor muscles simultaneously in single-leg cycling exercise. Therefore, the aim of the study was to investigate the relationship and agreement between systemic and local breakpoints in locomotor and non-locomotor muscles.MethodData from twelve physically-active participants (25.5 ± 3.9 years, 176.1 ± 11.6 cm, 71.2 ± 9.4 kg, 4 females) who completed a continuous single-leg step incremental cycling test (10 W min-1) with their right leg were included in the analysis. Ventilation and gas exchange were recorded to determine RCP. Surface EMG (sEMG) and NIRS signals were measured from both vasti lateralis muscles and breakpoints were determined from root mean Q square sEMG and deoxygenated hemo- and myoglobin signal m[HHb].ResultsThere was no significant difference in the power output at RCP (127.3 ± 21.8 W) and local muscular breakpoints both from the locomotor (m[HHb]: 119.7 ± 23.6 W, sEMG: 126.6 ± 26.0 W) and non-locomotor (m[HHb]: 117.5 ± 17.9 W, sEMG: 126.1 ± 28.4 W) muscles. Breakpoints also showed significant (p < 0.01) correlations (r = 0.67-0.90, ICC = 0.80-0.94) to each other with weaker correlations in the non-locomotor muscle (r = 0.66-0.86, ICC = 0.74-0.90). Despite the strong correlations, high individual variability and weak limits of agreement (up to -32.5-46.5 W) and substantial absolute differences (10.2-16.7 W) were observed which indicates that these breakpoints cannot be used interchangeably.DiscussionThese findings offer further insights into the mechanistic relationship between local and systemic physiological response to exercise with increasing workload. We conclude that, despite strong correlations, local muscular breakpoints do not have to coincide with systemic boundaries of physiological domains.
