Project description:Decades of hippocampal neurophysiology research have linked the hippocampal theta rhythm to voluntary movement. A consistent observation has been a robust correlation between the amplitude (or power) and frequency of hippocampal theta and running speed. Recently, however, it has been suggested that acceleration, not running speed, is the dominating influence on theta frequency. There is an inherent interdependence among these two variables, as acceleration is the rate of change in velocity. Therefore, we investigated theta frequency and amplitude of the local-field potential recorded from the stratum pyramidale, stratum radiatum, and stratum lacunosum moleculare of the CA1 subregion, considering both speed and acceleration in tandem as animals traversed a circular task or performed continuous alternation. In male and female rats volitionally controlling their own running characteristics, we found that running speed carries nearly all of the variability in theta frequency and power, with a minute contribution from acceleration. These results contradicted a recent publication using a speed-clamping task, where acceleration and movement are compelled through the use of a bottomless car (Kropff et al., 2021a). Therefore, we reanalyzed the speed-clamping data replicating a transient increase in theta frequency during acceleration. Compared with track running rats, the speed-clamped animals exhibited lower velocities and acceleration values but still showed a stronger influence of speed on theta frequency relative to acceleration. As navigation is the integration of many sensory inputs that are not necessarily linearly related, we offer caution in making absolute claims regarding hippocampal physiology from correlates garnered from a single behavioral repertoire.SIGNIFICANCE STATEMENT A long-standing, replicable observation has been the increase of hippocampal theta power and frequency with increasing running speed. Recently, however, an experimental approach that clamps the running speed of an animal has suggested that acceleration is the dominant influence. Therefore, we analyzed data from freely behaving rats as well as data from the speed-clamping experiment. In unrestrained behavior, speed remains the dominant behavioral correlate to theta amplitude and frequency. Positive acceleration in the speed-clamp experiment induced a transient increase in theta frequency and power. However, speed retained the dominant influence over theta frequency, changing with velocity in both acceleration and deceleration conditions.

Project description:Theta (6-12 Hz) rhythmicity in the local field potential (LFP) reflects a clocking mechanism that brings physically isolated neurons together in time, allowing for the integration and segregation of distributed cell assemblies. Variation in the theta signal has been linked to locomotor speed, sensorimotor integration as well as cognitive processing. Previously, we have characterized the relationship between locomotor speed and theta power and how that relationship varies across the septotemporal (long) axis of the hippocampus (HPC). The current study investigated the relationship between whole body acceleration, deceleration and theta indices at CA1 and dentate gyrus (DG) sites along the septotemporal axis of the HPC in rats. Results indicate that whole body acceleration and deceleration predicts a significant amount of variability in the theta signal beyond variation in locomotor speed. Furthermore, deceleration was more predictive of variation in theta amplitude as compared to acceleration as rats traversed a linear track. Such findings highlight key variables that systematically predict the variability in the theta signal across the long axis of the HPC. A better understanding of the relative contribution of these quantifiable variables and their variation as a function of experience and environmental conditions should facilitate our understanding of the relationship between theta and sensorimotor/cognitive functions.

Project description:Computational models of neural circuits with varying levels of biophysical detail have been generated in pursuit of an underlying mechanism explaining the ubiquitous hippocampal theta rhythm. However, within the theta rhythm are at least two types with distinct frequencies associated with different behavioral states, an aspect that must be considered in pursuit of these mechanistic explanations. Here, using our previously developed excitatory-inhibitory network models that generate theta rhythms, we investigate the robustness of theta generation to intrinsic neuronal variability by building a database of heterogeneous excitatory cells and implementing them in our microcircuit model. We specifically investigate the impact of three key "building block" features of the excitatory cell model that underlie our model design: these cells' rheobase, their capacity for post-inhibitory rebound, and their spike-frequency adaptation. We show that theta rhythms at various frequencies can arise dependent upon the combination of these building block features, and we find that the speed of these oscillations are dependent upon the excitatory cells' response to inhibitory drive, as encapsulated by their phase response curves. Taken together, these findings support a hypothesis for theta frequency control that includes two aspects: (i) an internal mechanism that stems from the building block features of excitatory cell dynamics; (ii) an external mechanism that we describe as "inhibition-based tuning" of excitatory cell firing. We propose that these mechanisms control theta rhythm frequencies and underlie their robustness.

Project description:In hippocampal area CA1 of rats, the frequency of gamma activity has been shown to increase with running speed (Ahmed and Mehta, 2012). This finding suggests that different gamma frequencies simply allow for different timings of transitions across cell assemblies at varying running speeds, rather than serving unique functions. However, accumulating evidence supports the conclusion that slow (∼25-55 Hz) and fast (∼60-100 Hz) gamma are distinct network states with different functions. If slow and fast gamma constitute distinct network states, then it is possible that slow and fast gamma frequencies are differentially affected by running speed. In this study, we tested this hypothesis and found that slow and fast gamma frequencies change differently as a function of running speed in hippocampal areas CA1 and CA3, and in the superficial layers of the medial entorhinal cortex (MEC). Fast gamma frequencies increased with increasing running speed in all three areas. Slow gamma frequencies changed significantly less across different speeds. Furthermore, at high running speeds, CA3 firing rates were low, and MEC firing rates were high, suggesting that CA1 transitions from CA3 inputs to MEC inputs as running speed increases. These results support the hypothesis that slow and fast gamma reflect functionally distinct states in the hippocampal network, with fast gamma driven by MEC at high running speeds and slow gamma driven by CA3 at low running speeds.

Project description:Hippocampal theta rhythm is believed to play a critical role in learning and memory. In animal models of temporal lobe epilepsy (TLE), there is evidence that alterations of hippocampal theta oscillations are involved in the cognitive impairments observed in this model. However, hippocampal theta frequency and amplitude at both the local field potential (LFP) and single unit level are strongly modulated by running speed, suggesting that the integration of locomotor information into memory processes may also be critical for hippocampal processing. Here, we investigate whether hippocampal speed-theta integration influences spatial memory and whether it could account for the memory deficits observed in TLE rats. LFPs were recorded in both Control (CTR) and TLE rats as they were trained in a spatial alternation task. TLE rats required more training sessions to perform the task at CTR levels. Both theta frequency and power were significantly lower in the TLE group. In addition, speed/theta frequency correlation coefficients and regression slopes varied from session to session and were worse in TLE. Importantly, there was a strong relationship between speed/theta frequency parameters and performance. Our analyses reveal that speed/theta frequency correlation with performance cannot merely be explained by the direct influence of speed on behavior. Therefore, variations in the coordination of theta frequency with speed may participate in learning and memory processes. Impairments of this function could explain at least partially memory deficits in epilepsy.

Project description:The speed of theta brain oscillatory activity is thought to play a key role in determining working memory (WM) capacity. Individual differences in the length of a theta cycle (ranging between 4 and 7 Hz) might determine how many gamma cycles (>30 Hz) can be nested into a theta wave. Gamma cycles are thought to represent single memory items; therefore, this interplay could determine individual memory capacity. We directly tested this hypothesis by means of parietal transcranial alternating current stimulation (tACS) set at slower (4 Hz) and faster (7 Hz) theta frequencies during a visuospatial WM paradigm. Accordingly, we found that 4-Hz tACS enhanced WM capacity, while 7-Hz tACS reduced WM capacity. Notably, these effects were found only for items presented to the hemifield contralateral to the stimulation site. This provides causal evidence for a frequency-dependent and spatially specific organization of WM storage, supporting the theta-gamma phase coupling theory of WM capacity.

Project description:Measuring impact-related quantities in running is of interest to improve the running technique. Many quantities are typically measured in a controlled laboratory setting, even though most runners run in uncontrolled outdoor environments. While monitoring running mechanics in an uncontrolled environment, a decrease in speed or stride frequency can mask fatigue-related changes in running mechanics. Hence, this study aimed to quantify and correct the subject-specific effects of running speed and stride frequency on changes in impact-related running mechanics during a fatiguing outdoor run. Seven runners ran a competitive marathon while peak tibial acceleration and knee angles were measured with inertial measurement units. Running speed was measured through sports watches. Median values over segments of 25 strides throughout the marathon were computed and used to create subject-specific multiple linear regression models. These models predicted peak tibial acceleration, knee angles at initial contact, and maximum stance phase knee flexion based on running speed and stride frequency. Data were corrected for individual speed and stride frequency effects during the marathon. The speed and stride frequency corrected and uncorrected data were divided into ten stages to investigate the effect of marathon stage on mechanical quantities. This study showed that running speed and stride frequency explained, on average, 20%-30% of the variance in peak tibial acceleration, knee angles at initial contact, and maximum stance phase knee angles while running in an uncontrolled setting. Regression coefficients for speed and stride frequency varied strongly between subjects. Speed and stride frequency corrected peak tibial acceleration, and maximum stance phase knee flexion increased throughout the marathon. At the same time, uncorrected maximum stance phase knee angles showed no significant differences between marathon stages due to a decrease in running speed. Hence, subject-specific effects of changes in speed and stride frequency influence the interpretation of running mechanics and are relevant when monitoring, or comparing the gait pattern between runs in uncontrolled environments.

Project description:Individuals regulate the transmission of shock to the head during running at different stride frequencies although the consequences of this on head-gaze stability remain unclear. The purpose of this study was to examine if providing individuals with visual feedback of their head-gaze orientation impacts tibial and head accelerations, shock attenuation and head-gaze motion during preferred speed running at different stride frequencies. Fifteen strides from twelve recreational runners running on a treadmill at their preferred speed were collected during five stride frequencies (preferred, ±10% and ±20% of preferred) in two visual task conditions (with and without real-time visual feedback of head-gaze orientation). The main outcome measures were tibial and head peak accelerations assessed in the time and frequency domains, shock attenuation from tibia to head, and the magnitude and velocity of head-gaze motion. Decreasing stride frequency resulted in greater vertical accelerations of the tibia (p<0.01) during early stance and at the head (p<0.01) during early and late stance; however, for the impact portion the increase in head acceleration was only observed for the slowest stride frequency condition. Visual feedback resulted in reduced head acceleration magnitude (p<0.01) and integrated power spectral density in the frequency domain (p<0.01) in late stance, as well as overall of head-gaze motion (p<0.01). When running at preferred speed individuals were able to stabilize head acceleration within a wide range of stride frequencies; only at a stride frequency 20% below preferred did head acceleration increase. Furthermore, impact accelerations of the head and tibia appear to be solely a function of stride frequency as no differences were observed between feedback conditions. Increased visual task demands through head gaze feedback resulted in reductions in head accelerations in the active portion of stance and increased head-gaze stability.

Project description:Cross-frequency coupling (CFC) between theta and high-frequency oscillations (HFOs) is predominant during active wakefulness, REM sleep and behavioral and learning tasks in rodent hippocampus. Evidence suggests that these state-dependent CFCs are linked to spatial navigation and memory consolidation processes. CFC studies currently include only the cortical and subcortical structures. To our knowledge, the study of nucleus tractus solitarius (NTS)-cortical structure CFC is still lacking. Here we investigate CFC in simultaneous local field potential recordings from hippocampal CA1 and the NTS during behavioral states in freely moving rats. We found a significant increase in theta (6-8 Hz)-HFO (120-160 Hz) coupling both within the hippocampus and between NTS theta and hippocampal HFOs during REM sleep. Also, the hippocampal HFOs were modulated by different but consistent phases of hippocampal and NTS theta oscillations. These findings support the idea that phase-amplitude coupling is both state- and frequency-specific and CFC analysis may serve as a tool to help understand the selective functions of neuronal network interactions in state-dependent information processing. Importantly, the increased NTS theta-hippocampal HFO coupling during REM sleep may represent the functional connectivity between these two structures which reflects the function of the hippocampus in visceral learning with the sensory information provided by the NTS. This gives a possible insight into an association between the sensory activity and REM-sleep dependent memory consolidation.

Project description:The anterior thalamic nuclei are assumed to support episodic memory with anterior thalamic dysfunction a core feature of diencephalic amnesia. To date, the electrophysiological characterization of this region in behaving rodents has been restricted to the anterodorsal nucleus. Here we compared single-unit spikes with population activity in the anteroventral nucleus (AV) of freely moving rats during foraging and during naturally occurring sleep. We identified AV units that synchronize their bursting activity in the 6-11 Hz range. We show for the first time in freely moving rats that a subgroup of AV neurons is strongly entrained by theta oscillations. This feature together with their firing properties and spike shape suggests they be classified as "theta" units. To prove the selectivity of AV theta cells for theta rhythm, we compared the relation of spiking rhythmicity to local field potentials during theta and non-theta periods. The most distinguishable non-theta oscillations in rodent anterior thalamus are sleep spindles. We therefore compared the firing properties of AV units during theta and spindle periods. We found that theta and spindle oscillations differ in their spatial distribution within AV, suggesting separate cellular sources for these oscillations. While theta-bursting neurons were related to the distribution of local field theta power, spindle amplitude was independent of the theta units' position. Slow- and fast-spiking bursting units that are selectively entrained to theta rhythm comprise 23.7% of AV neurons. Our results provide a framework for electrophysiological classification of AV neurons as part of theta limbic circuitry.
