The complete mitochondrial genomes of Diplonevra funebris and Diplonevra peregrina (Diptera: Phoridae).

ABSTRACT: Diplonevra is one of the most important genera in the family Phoridae. This genus is mainly distributed in Palearctic region, and its species can be used to estimate the postmortem interval. In this study, we first present two mitochondrial genomes of common necrophagous species of this genus, Diplonevra funebris (Meigen, 1830) and Diplonevra peregrina (Wiedemann, 1830). Maximum-likelihood phylogenetic tree revealed that the genus Diplonevra is closely related to the genus Dohrniphora within the family Phoridae. This work expands the knowledge about the Phoridae genomes, and contributes to the further study of species identification and phylogenetics of this family.

Project description:Megaselia spiracularis and Dohrniphora cornuta were two forensically important flies in relatively sealed environments. Their mitochondrial genomes were first sequenced, annotated, and phylogenetic analyses were performed with other 8 species of the Asehiza in this study. Maximum-likelihood phylogenetic tree revealed that Phoridae is closer to Platypezidae and Lonchopteridae within Diptera. This work increases the databases of Phoridae species, and contributes to the further study of species identification and phylogenetics of this family.

Project description:BackgroundFlies of the family Phoridae (Insecta: Diptera) are amongst the most diverse insects in the world, with an incredible array of species, structures and life histories. Wiithin their structural diversity is the world's smallest fly, Euryplatea nanaknihali Brown, 2012.New informationA second minute, limuloid female phorid parasitoid fly (Diptera: Phoridae) is described. Known from a single specimen from a site near Manaus, Brazil, Megapropodiphora arnoldigen. n., sp. n. is only 0.395 mm in body length, slightly smaller than the currently recognised smallest fly, Euryplatea nanaknihali from Thailand. The distinctive body shape of M. arnoldi, particularly the relatively enormous head, mesothorax and scutellum, the latter of which covers most of the abdomen, easily separates it from other described phorids. Most remarkably, the forelegs are extremely enlarged, whereas mid- and hind legs are reduced to small, possibly vestigial remnants. A possible male specimen, unfortunately destroyed during processing, is briefly described.

The Application of Mitochondrial COI Gene-Based Molecular Identification of Forensically Important Scuttle Flies (Diptera: Phoridae) in Korea.

Project description:Phoridae are a family of necrophagous flies commonly found in indoor death scene. They account for approximately 19.7% of the entomofauna in human cadavers in Korea. Additionally, this taxon is an indicator of indoor hygiene, and these flies appear in environments where access by other necrophagous insects is difficult, such as enclosed rooms. Thus, they are likely to be used as forensic evidence. Despite their importance in forensic investigations and environmental hygiene, detailed studies on the taxonomy and molecular barcoding for this family are scarce, including in Korea. Because accurate taxonomic information regarding necrophagous insects collected from a death-related scene is essential during medicolegal investigations, molecular barcoding data could be useful as well as reliable. In this paper, full-length nucleotide sequences of genes coding for the cytochrome c oxidase subunit I (COI) in 79 Phoridae larvae collected from 20 medicolegal autopsy cases in Korea were phylogenetically analyzed by comparing their sequences to the foreign barcoding data of Phoridae. Six mitochondrial haplogroups were identified, which two of them matched to foreign Phoridae fly species haplotypes, Megaselia scalaris (Loew, 1866) and M. spiracularis Schmitz 1938. Taxonomies of five other haplogroups, with nucleotide distances ranging from 1.68% to 2.26% from the M. scalaris group, could not be confirmed solely based on the molecular barcoding data. Further research should be performed to determine whether these five haplogroups are diverged conspecifics of M. scalaris or a closely related sister cryptic species of M. scalaris.

Project description:BackgroundPhorid flies are amongst the most biologically diverse and species-rich groups of insects. Ways of life range from parasitism, herbivory, fungivory, to scavenging. Although the lifestyles of most species are unknown, many are parasitoids, especially of social insects. Some species of ant-parasitoids are attracted to injured hosts for feeding purposes to develop eggs, as well as for oviposition, requiring each female to find two injured hosts.New informationFemales of the phorid fly Megaselia steptoeae Hartop et al. (Diptera: Phoridae) were found to be quickly attracted to crushed glass snails of the species Oxychilus draparnaudi (Beck) (Gastropoda: Oxychilidae). Most females were without mature eggs and apparently were attracted for feeding purposes only; other injured molluscs offered at the same time were not attractive. One female laid eggs in captivity and offspring were reared to the pupal stage. The lifestyle of this species is similar to that of parasitoids of injured ants, which also require separate hosts of the same species for feeding and oviposition. We conclude that injured hosts must be common in the environment to attract these host-specific scavengers.

A new type of ant-decapitation in the Phoridae (Insecta: Diptera).

Project description:The genus Dohrniphora is a hyperdiverse group of phorid flies, a family whose species are commonly characterized as generalized scavengers. The lifestyle of most species of Dohrniphora is unknown, although one cosmopolitan, synanthropic species, D.cornuta (Bigot) fits the general scavenger mold. Here we show that flies of the D.longirostrata species group exhibit highly specific "headhunting" behavior in which injured Odontomachus ants are decapitated, the heads dragged away, and females either feed on their contents or lay an egg nearby. Since most females studied lacked eggs in their ovaries, we conclude that this bizarrely specialized feeding is necessary to provide nutrients for reproduction in these flies. Our study provides further evidence that injured ants are a common, stable resource in tropical ecosystems that support a wide array of phorid flies. Such narrowly constrained lifestyles, as exemplified by exclusively feeding on and breeding in the head contents of certain ponerine worker ants, could allow the co-existence of a huge community of saprophagous flies.

Complete mitochondrial genomes of two scaphopod molluscs.

Project description:Complete mitochondrial genomes were determined for two scaphopod molluscs: the dentaliid Antalis entalis and an unidentified Antarctic gadilid. Both genomes are complete except, in Gadilida sp. indet., a short stretch of nad5 was undetermined and trnR could not be annotated. Organization of the Gadilida sp. genome is nearly identical to that previously reported for the gadilid Siphonodentalium whereas trnK, nad5, trnD, nad4, and nad4l are transposed to the opposite strand in the previously published Graptacme genome relative to that of Antalis. Phylogenetic analysis of the 13 protein-coding and 2 rRNA genes recovered Scaphopoda, Gadilida, and Dentaliida monophyletic with maximal support.
