The complete mitochondrial genome of Aulacochilus grouvellei Achard, 1923 (Coleoptera: Erotylidae: Erotylinae) with a phylogenetic analysis of Cucujoidea.

ABSTRACT: The complete mitochrondiral genome of Aulacochilus grouvellei Achard, 1923 was sequenced using an Illumina HiSeq platform. It represents the first mitochondrial genome of the subfamily Erotylinae. The mitogenome is a double-stranded circular molecule 15,607 bp in length with 22 transfer RNA genes, 13 protein-coding genes, 2 ribosomal RNA genes, and a control region. A Bayesian inference phylogenetic analysis including the new mitochondrial genome and a broad selection of other Cucujoidea recovered four major clades, including a 'Cryptophagidae-Laemophloeidae-Cucujidae' clade, a 'Monotomidae-Nitidulidae' clade, an Erotylidae clade, and a 'Coccinellidae-Silvanidae' clade. The family Erotylidae was recovered closely related to the 'Cryptophagidae-Laemophloeidae-Cucujidae' - 'Monotomidae -Nitidulidae' clade.

Three Complete Mitochondrial Genomes of Erotylidae (Coleoptera: Cucujoidea) with Higher Phylogenetic Analysis.

Project description:The family Erotylidae belongs to the superfamily Cucujoidea, which are phytophagous and mycophagous. So far, only two representative complete mitochondrial (mt) genomes of Erotylidae have been sequenced. Mitochondrial genomes of Tritoma metasobrina, Neotriplax arisana, and Episcapha opaca, which all belong to the subfamily Erotylinae, were sequenced using next-generation sequencing technology to better understand the diversity of mt genomes of Erotylidae. A comparative mt genomic analysis was conducted on the three sequenced representatives of Erotylinae and Languriinae sp. (Languriinae). The size of the complete mt genome of the 4 species ranged from 15,581 bp to 16,502 bp in length, including 37 genes (13 protein-coding genes, 22 transfer RNAs, and 2 ribosomal RNAs) and the control region. The arrangements of their mt genomes are highly consistent with other Coleoptera species. The start codons of two PCGs (ND1 and ND5) and the stop codons of one PCG (ATP8) were illustrated differences between Languriinae sp. and the other three species. All tRNAs of these 4 species exhibited cloverleaf secondary structures except that the dihydorouridine (DHU) arm of tRNASer(AGN) was absent. The phylogenetic analyses using both Bayesian inference (BI) and maximum likelihood (ML) methods all supported that Erotylidae as monophyletic. Erotylinae was monophyletic being the sister group to Xenocelinae. Languriinae was closely related to 'Erotylinae-Xenocelinae'. Our results recovered Languriinae nested within Erotylidae.

The complete mitochondrial genome of Triplax ainonia Lewis, 1877 (Coleoptera: Erotylidae).

Project description:The beetle Triplax ainonia Lewis, 1877 is a serious pest of cultivated the mushroom Pleurotus ostreatus in China. The complete mitochondrial genome of this species was reported for the first time in this study. The mitogenome was 17,555 bp in length and had a base composition of 39.4% A, 36.1% T, 8.7% G and 15.3% C, which indicated that the base composition was AT-biased. Similar to other species of Coleoptera, the mitogenome of T. ainonia contained 13 protein-coding genes, 22 transfer RNA genes, two ribosomal RNA unit genes, and a large noncoding region. Phylogenetic analysis based on mitogenomes suggested that the family Erotylidae was a monophyletic group.

The complete mitochondrial genome of Tenomerga trabecula (Coleoptera: Archostemata: Cupedidae) and phylogenetic analysis among Coleoptera.

Project description:Tenomerga trabecula belongs to the Cupedidae family of Coleoptera. The first complete mitogenome of Cupedidae is reported in this paper. The genome is 16,741 bp in length and contains the typical 37 genes with 22 transfer RNA genes, 13 protein-coding genes, and 2 ribosomal RNA genes, which are arranged in the same order as the putative ancestor of beetles. The average base composition of the mitogenome is 42.9% for A, 14.5% for C, 8.7% for G, and 33.9% for T. The percentage of A + T is 76.8%. The genome organization, nucleotide composition, and codon usage are similar to other beetles. Phylogenetic analysis shows that Archostemata is monophyletic. Myxophaga, Adephaga, and Polyphaga are also monophyletic.

The complete mitochondrial genome of Melanotus cribricollis (Coleoptera: Elateridae) and phylogenetic analysis.

Project description:We determined the complete mitogenome sequence of Melanotus cribricollis (Faldermann) (GenBank accession no. MK792748). The mitogenome is 15,908 bp in length, consisting of 13 protein-coding genes, 22 transfer RNA genes, two ribosomal RNA genes, and one non-coding control region. The overall nucleotide composition was 40.8% A, 31.5% T, 17.4% C, and 10.3% G, with 72.3% of AT, respectively. Phylogenetic analysis based on the 13 coding protein genes nucleotide sequences revealed that M. cribricollis clustered with the same genus species M. villosus, and the three genus Melanotus Eschscholtz, Agriotes Eschscholtz and Adrastus Eschscholtz had a close relationship.

First Complete Mitochondrial Genome of Melyridae (Coleoptera, Cleroidea): Genome Description and Phylogenetic Implications.

Project description:To explore the characteristics of the mitogenome of Melyridae and reveal phylogenetic relationships, the mitogenome of Cordylepherus sp. was sequenced and annotated. This is the first time a complete mitochondrial genome has beengenerated in this family. Consistent with previous observations of Cleroidea species, the mitogenome of Cordylepherus sp. is highly conserved in gene size, organization and codon usage, and secondary structures oftRNAs. All protein-coding genes(PCGs) initiate withthe standard start codon ATN,except ND1, which starts with TTG, and terminate with the complete stop codons of TAA and TAG, or incomplete forms, TA- and T-. MosttRNAs have the typical clover-leaf structure, except trnS1 (Ser, AGN), whose dihydrouridine (DHU) armis reduced. In the A+T-rich region, three types of tandem repeat sequence units are found, including a 115 bp sequence tandemly repeated twice, a 16 bp sequence tandemly repeated three timeswith a partial third repeat and a 10 bp sequence tandemly repeated seven times. Phylogenetic analyses based on 13 protein-coding genes by both Bayesian inference (BI) and maximum likelihood (ML) methods suggest that Melyridae sensu lato is polyphyletic, and Dasytinae and Malchiinae are supported as independent families.

Complete mitochondrial genome of Pseudoechthistatus hei (Coleoptera: Cerambycidae: Lamiinae) and its phylogenetic analysis.

Project description:The little-known genus Pseudoechthistatus Pic, 1917 belongs to the subfamily Lamiinae of the family Cerambycidae. Adult Pseudoechthistatus hei Xie and W. Wang, 2019 specimens were collected from Bāijì Hill, Xīntángfáng Village, Wéixī County, Yúnnán Province, China. The complete mitochondrial genome (GenBank accession number: ON641973.1) of P. hei was sequenced, annotated, and characterized; it is a circular DNA molecule of 16,103 bp with a 75.71% AT content, and it comprised 13 protein-coding genes (PCG), 22 tRNA genes, two rRNA genes, and 1 control region. The PCGs initiated with the typical ATN (Met) start codons, and were terminated by typical TAN stop codons. The Bayesian Inference phylogenetic tree was first constructed using JTT + F + I + G4 model for P. hei, which showed that P. hei was closely related to Monochamus alternatus alternatus.

The phylogenetic tree analysis using 16 species of Elateriformia shows that L. asperipennis is closest to Chauliognathus opacus, which belongs to the subfamily Chauliognathinae of Cantharidae.

Complete Mitochondrial Genome of Scolytoplatypodini Species (Coleoptera: Curculionidae: Scolytinae) and Phylogenetic Implications.

Project description:The complete mitochondrial genomes (mitogenomes) of beetles in the tribe Scolytoplatypodini (genus Scolytoplatypus) were sequenced and annotated. These included Scolytoplatypus raja (15,324 bp), Scolytoplatypus sinensis (15,394 bp), Scolytoplatypus skyliuae (15,167 bp), and Scolytoplatypus wugongshanensis (15,267 bp). The four mitogenomes contained 37 typical genes, including 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), and 2 ribosomal RNA genes (rRNAs). The gene orientation and arrangement of the four mitogenomes were similar to other Coleoptera mitogenomes. PCGs mostly started with ATN and terminated with TAA. The Ka/Ks ratio of 13 PCGs in the four species revealed that cox1 had the slowest evolutionary rate and atp8 and nad6 had a higher evolutionary rate. All tRNAs had typical cloverleaf secondary structures, but trnS1 lacked dihydrouridine arm. Partial tRNAs lost the discriminator nucleotide. The trnY did not possess the discriminator nucleotide and also lost three bases, showing a special amino-acyl arm. Bayesian inference (BI) and maximum likelihood (ML) methods were conducted for phylogenetic analyses using 13 PCGs. Scolytoplatypodini was clustered with Hylurgini and Hylastini, and the monophyly of Scolytoplatypodini was supported. The four newly sequenced mitogenomes increase understanding of the evolutionary relationships of Scolytoplatypodini and other Scolytinae species.

Complete mitochondrial genome and phylogenetic analysis of Mastax latefasciata Liebke 1931 (Insecta: Coleoptera: Carabidae).

Project description:The genus Mastax Fischer von Waldheim 1827 belongs to the family Carabidae. Specimens of adult Mastax latefasciata Liebke, 1931 were collected from Yājì Hill, Huáihuà City, Húnán Province, China. The complete mitochondrial genome (GenBank accession number ON674050.1) of M. latefasciata was sequenced, annotated, and characterized. The results showed that it was a circular DNA molecule of 16,735 bp with 81.07% AT content and comprised 13 protein-coding genes (PCG), 22 tRNA genes, 2 rRNA genes, and 1 control region. The PCGs were initiated using typical ATN (Met) and TTG (Met) start codons and terminated using typical TAN stop codons. The phylogenetic position of Mastax within the Carabidae was first evaluated using complete mitogenomes, and the results showed that it was close to Cicindela anchoralis and Manticora tibialis.
