Project description:Cuticular hydrocarbons (CHCs) have, in insects, important physiological and ecological functions, such as protection against desiccation and as semiochemicals in social taxa, including termites. CHCs are, in termites, known to vary qualitatively and/or quantitatively among species, populations, castes, or seasons. Changes to hydrocarbon profile composition have been linked to varying degrees of aggression between termite colonies, although the variability of results among studies suggests that additional factors might have been involved. One source of such variability may be colony age, as termite colony demographics significantly change over time, with different caste and instar compositions throughout the life of the colony. We here hypothesize that the intracolonial chemical profile heterogeneity would be high in incipient termite colonies but would homogenize over time as a colony ages and accumulates older workers in improved homeostatic conditions. We studied caste-specific patterns of CHC profiles in Coptotermes gestroi colonies of four different age classes (6, 18, 30, and 42 months). The CHC profiles were variable among castes in the youngest colonies, but progressively converged toward a colony-wide homogenized chemical profile. Young colonies had a less-defined CHC identity, which implies a potentially high acceptance threshold for non-nestmates conspecifics in young colonies. Our results also suggest that there was no selective pressure for an early-defined colony CHC profile to evolve in termites, potentially allowing an incipient colony to merge nonagonistically with another conspecific incipient colony, with both colonies indirectly and passively avoiding mutual destruction as a result.

Project description:BACKGROUND:Foraging in group living animals such as social insects, is collectively performed by individuals. However, our understanding on foraging behavior of subterranean termites is extremely limited, as the process of foraging in the field is mostly concealed. Because of this limitation, foraging behaviors of subterranean termites were indirectly investigated in the laboratory through tunnel geometry analysis and observations on tunneling behaviors. In this study, we tracked subsets of foraging workers from juvenile colonies of Coptotermes formosanus (2-yr-old) to describe general foraging behavioral sequences and to find how foraging workers allocate time between the foraging site (food acquisition or processing) and non-foraging site (food transportation). RESULTS:Once workers entered into the foraging site, they spent, on average, a significantly longer time at the foraging site than the non-foraging site. Our clustering analysis revealed two different types of foraging workers in the subterranean termite based on the duration of time they spent at the foraging site and their foraging frequency. After entering the foraging site, some workers (cluster 1) immediately initiated masticating wood fragments, which they transferred as food boluses to recipient workers at the foraging site. Conversely, the recipient workers (cluster 2) moved around after entering the foraging site and received food from donating workers. CONCLUSIONS:This study provides evidence of task specialization within foraging cohorts in subterranean termites.

Project description:In insect societies, the presence of reproductives or eggs has been shown to shape several biological traits in the colony members. Social interactions are one of these traits that involve modification of the communication system of the entire colony. Many studies described the role of chemical compounds and dominance behaviors in the presence of reproductive but vibratory behaviors received very few investigations. Yet, vibratory behaviors are ideal candidates, particularly for subterranean species like termites, as they could be quickly transmitted through the substrate and could be very diversified (origin, modulation). Here, we investigated whether the presence of reproductives/eggs affects the vibratory behavior (body-shaking) of workers in the subterranean termite Reticulitermes flavipes. Our results reveal that the presence of reproductives or eggs triggers an increase of workers' body-shaking, independent of their colony of origin after 24 hr. We hypothesize that vibratory communication could be used to transfer information about the presence of reproductives and eggs to the entire colony, suggesting that vibratory behaviors could serve as an important yet neglected mediator of social regulation.

Project description:In social insects, alerting nestmates to the presence of a pathogen should be critical for limiting its spread and initiating social mechanisms of defense. Here we show that subterranean termites use elevated vibratory alarm behavior to help prevent fatal fungal infections. The elevated alarm leads to elevated social hygiene. This requires that termites coalesce so that they can groom each other's cuticular surfaces of contaminating conidial spores. Groups of 12 Reticulitermes flavipes workers varied in their response when immersed in conidia solutions of nine different strains of Metarhizium. Pathogen alarm displays of short 2-7-second bursts of rapid longitudinal oscillatory movement (LOM), observed over 12 min following a fungal challenge, were positively correlated with the time that workers spent aggregated together grooming each other. The frequency of these LOMs was inversely correlated with fatal fungal infections. The variation in fatalities appeared to be largely attributable to a differential response to Metarhizium brunneum and Metarhizium robertsii in the time spent in aggregations and the frequency of allogrooming. Isolated workers challenged with conidia did not display LOMs, which suggests that the alarm is a conditional social response. LOMs appear to help signal the presence of fungal pathogens whose virulence depends on the level of this emergency alert.

Project description:Analyzing the information-rich content of RNA can help uncover genetic events associated with social insect castes or other social polymorphisms. Here, we exploit a series of cDNA libraries previously derived from whole-body tissue of different castes as well as from three behaviourally distinct populations of the Eastern subterranean termite Reticulitermes flavipes. We found that the number (~0.5 M) of single nucleotide variants (SNVs) was roughly equal between nymph, worker and soldier caste libraries, but dN/dS (ratio of nonsynonymous to synonymous substitutions) analysis suggested that some of these variants confer a caste-specific advantage. Specifically, the dN/dS ratio was high (~4.3) for genes expressed in the defensively specialized soldier caste, relative to genes expressed by other castes (~1.7−1.8) and regardless of the North American population (Toronto, Raleigh, Boston) from which the castes were sampled. The populations, meanwhile, did show a large difference in SNV count but not in the manner expected from known demographic and behavioural differences; the highly invasive unicolonial population from Toronto was not the least diverse and did not show any other unique substitution patterns, suggesting any past bottleneck associated with invasion or with current unicoloniality has become obscured at the RNA level. Our study raises two important hypotheses relevant to termite sociobiology. First, the positive selection (dN/dS > 1) inferred for soldier-biased genes is presumably indirect and of the type mediated through kin selection, and second, the behavioural changes that accompany some social insect urban invasions (i.e., ‘unicoloniality’) may be detached from the loss-of-diversity expected from invasion bottlenecks.

Project description:In monogamous species that exhibit extensive biparental investment, such as termites, both sexes are predicted to be selective when choosing a mate. Size-related traits are expected to be important in partner selection because the fat reserves of the colony founders sustain the incipient colony. Partner relatedness and heterozygosity determine the degree of inbreeding and genetic diversity within the colony and may thus also influence partner selection. To test these predictions, we investigated whether phenotypic and genetic traits influence mate choice and/or competitive advantage during pair formation of Formosan subterranean termites, Coptotermes formosanus (Isoptera: Rhinotermitidae). Pair formation in termites normally occurs within a short period after swarming when alates form tandem pairs on the ground. Alates were collected from 5 light trap samples in the French Quarter of New Orleans, LA. From each sample, both tandem pairs and single individuals were collected and their sex, body weights, and head widths were recorded. Pairwise relatedness and individual levels of heterozygosity were determined by microsatellite genotyping. Males in tandem pairs with females had a significantly larger head width than males that did not form tandem pairs. Weights as well as head widths of tandem running partners were positively correlated. For the majority of the samples, relatedness between tandem partners did not differ from the relatedness to members of the other tandem pairs. Thus, no kin discrimination occurred during tandem running. However, females engaged in tandem running had a higher degree of heterozygosity than females that remained single. These findings suggest partner selection and/or competitive advantage based on size-related phenotypic parameters and genetic diversity. The pairing advantage of heterozygous females might explain previous findings of sex-biased alate production depending on the degree of inbreeding in colonies of several species of the genus Coptotermes.

Project description:A long-standing goal of evolutionary biology is to understand how paleoclimatic and geological events shape the geographical distribution and genetic structure within and among species. Using a diverse set of markers (cuticular hydrocarbons, mitochondrial and nuclear gene sequences, microsatellite loci), we studied Reticulitermes grassei and R. banyulensis, two closely related termite species in southwestern Europe. We sought to clarify the current genetic structure of populations that formed following postglacial dispersal from refugia in southern Spain and characterize the gene flow between the two lineages over the last several million years. Each marker type separately provided a fragmented picture of the evolutionary history at different timescales. Chemical analyses of cuticular hydrocarbons and phylogenetic analyses of mitochondrial and nuclear genes showed clear separation between the species, suggesting they diverged following vicariance events in the Late Miocene. However, the presence of intermediate chemical profiles and mtDNA introgression in some Spanish colonies suggests ongoing gene flow. The current genetic structure of Iberian populations is consistent with alternating isolation and dispersal events during Quaternary glacial periods. Analyses of population genetic structure revealed postglacial colonization routes from southern Spain to France, where populations underwent strong genetic bottlenecks after traversing the Pyrenees resulting in parapatric speciation.

Project description:Termites are distributed throughout the world and often cause economic losses. This study aims to; (1) analyze the relationship between the distribution of termite species and the environmental conditions of Makassar city; (2) determine the level of attack in the Makassar City; and (3) map the risk of termite attack in the Makassar City. Samples of Pinus merkusii (2 × 2 × 20 cm) were placed at 13 observation stations and covered using polyvinyl chloride (PVC) tubes (4? in diameter, 25-cm in length). Samples remained in place for 6 months, at which point the presence of termite attacks as well as their intensity and frequency were analyzed. Three species of termites were found among the 13 stations: Schedorhinotermes sp., Coptotermes gestroi, and Microcerotermes serrula. Their presence was significantly influenced by environmental factors at each station. The study results showed a low rate of attack in several subdistricts of Bontoala, Biring Kanaya, Makassar, Mamajang, Mariso, Manggala, Panakukang, Rappocini, Tallo, Tamalanrea, Tamalate, Ujung Pandang, Ujung Tanah, and Wajo districts. A medium level of attacks was found in subdistricts within Biring Kanaya, Manggala, Mamajang, Panakukang, Rappocini, and Tallo Districts. Finally, a high risk of termite attacks was present in several subdistricts of Biringkanaya, Tamalantera, Rappocini, Manggala, and Tamalate districts.

Project description:In population genetics studies, detecting and quantifying the distribution of genetic variation can help elucidate ecological and evolutionary processes. In social insects, the distribution of population-level genetic variability is generally linked to colony-level genetic structure. It is thus especially crucial to conduct complementary analyses on such organisms to examine how spatial and social constraints interact to shape patterns of intraspecific diversity. In this study, we sequenced the mitochondrial COII gene for 52 colonies of the subterranean termite Reticulitermes grassei (Isoptera: Rhinotermitidae), sampled from a population in southwestern France. Three haplotypes were detected, one of which was found exclusively in the southern part of the study area (near the Pyrenees). After genotyping 6 microsatellite loci for 512 individual termites, we detected a significant degree of isolation by distance among individuals over the entire range; however, the cline of genetic differentiation was not continuous, suggesting the existence of differentiated populations. A spatial principal component analysis based on allele frequency data revealed significant spatial autocorrelation among genotypes: the northern and southern groups were strongly differentiated. This finding was corroborated by clustering analyses; depending on the randomized data set, two or three clusters, exhibiting significant degrees of differentiation, were identified. An examination of colony breeding systems showed that colonies containing related neotenic reproductives were prevalent, suggesting that inbreeding may contribute to the high level of homozygosity observed and thus enhance genetic contrasts among colonies. We discuss the effect of evolutionary and environmental factors as well as reproductive and dispersal modes on population genetic structure.

Project description:Social insects nesting in soil environments are in constant contact with entomopathogens but have evolved a range of defence mechanisms, resulting in both individual and social immunity that reduce the chance for epizootics in the colony, as in the case of subterranean termites. Coptotermes formosanus uses its faeces as building material for its nest structure that result into a 'carton material', and here, we report that the faecal nest supports the growth of Actinobacteria which provide another level of protection to the social group against entomopathogens. A Streptomyces species with in vivo antimicrobial activity against fungal entomopathogens was isolated from the nest material of multiple termite colonies. Termite groups were exposed to Metarhizium anisopliae, a fungal entomopathogen, during their foraging activity and the presence of Streptomyces within the nest structure provided a significant survival benefit to the termites. Therefore, this report describes a non-nutritional exosymbiosis in a termite, in the form of a defensive mutualism which has emerged from the use of faecal material in the nesting structure of Coptotermes. The association with an Actinobacteria community in the termite faecal material provides an extended disease resistance to the termite group as another level of defence, in addition to their individual and social immunity.
