Benthic Crustacean Digestion Can Modulate the Environmental Fate of Microplastics in the Deep Sea.

ABSTRACT: Microplastics (MPs) are ubiquitous contaminants of the marine environment, and the deep seafloor is their ultimate sink compartment. Manipulative and field experiments provided evidence of the ingestion of MPs by deep-sea fauna, but knowledge of MPs' fate once ingested still remains scant. We provide evidence of MP partial retention and fragmentation mediated by digestion activity of a Norwegian langoustine, a good bioindicator for MP contamination of the deep sea. We report here that MPs in the intestines were more abundant and significantly smaller (up to 1 order of magnitude in surface) than those in the stomachs. Our results show that the stomach can act as a size-bottleneck for ingested MPs, enhancing the retention of larger particles within the stomach and promoting fragmentation into smaller plastic debris, which is then released in the intestine. Our results provide evidence that the langoustine is responsible for the fragmentation of MPs already accumulated in sediments through its scavenging activity and digestion. These findings highlight the existence of a new peculiar kind of "secondary" MPs, introduced in the environment by biological activities, which could represent a significant pathway of plastic degradation in a secluded and stable environment such as the deep sea.

Project description:The waters around Iceland, bounding the Northern North Atlantic and the Nordic seas, are a region characterized by complex hydrography and seabed topography. This and the presence of the Greenland-Iceland-Faroe-Scotland ridge (GIFR) are likely to have a major impact on the diversity and distribution of the benthic fauna there. Biodiversity in this region is also under increasing threat from climate-induced changes, ocean warming and acidification in particular, affecting the marine realm. The aim of the present study was to investigate the biodiversity and distributional patterns of amphipod crustaceans in Icelandic waters and how it relates to environmental variables and depth. A comprehensive data set from the literature and recent expeditions was compiled constituting distributional records for 355 amphipod species across a major depth gradient (18-3,700 m). Using a 1° hexagonal grid to map amphipod distributions and a set of environmental factors (depth, pH, phytobiomass, velocity, dissolved oxygen, dissolved iron, salinity and temperature) we could identify four distinct amphipod assemblages: A Deep-North, Deep-South, and a Coastal cluster as well as one restricted to the GIFR. In addition to depth, salinity and temperature were the main parameters that determined the distribution of amphipods. Diversity differed greatly between the depth clusters and was significantly higher in coastal and GIFR assemblages compared to the deep-sea clusters north and south of the GIFR. A variety of factors and processes are likely to be responsible for the perceived biodiversity patterns, which, however, appear to vary according to region and depth. Low diversity of amphipod communities in the Nordic basins can be interpreted as a reflection of the prevailing harsh environmental conditions in combination with a barrier effect of the GIFR. By contrast, low diversity of the deep North Atlantic assemblages might be linked to the variable nature of the oceanographic environment in the region over multiple spatio-temporal scales. Overall, our study highlights the importance of amphipods as a constituent part of Icelandic benthos. The strong responses of amphipod communities to certain water mass variables raise the question of whether and how their distribution will change due to climate alteration, which should be a focus of future studies.

Project description:Archaea are widespread in marine sediments and play important roles in the cycling of sedimentary organic carbon. However, factors controlling the distribution of archaea in marine sediments are not well understood. Here we investigated benthic archaeal communities over glacial-interglacial cycles in the northern South China Sea and evaluated their responses to sediment organic matter sources and inter-species interactions. Archaea in sediments deposited during the interglacial period Marine Isotope Stage (MIS) 1 (Holocene) were significantly different from those in sediments deposited in MIS 2 and MIS 3 of the Last Glacial Period when terrestrial input to the South China Sea was enhanced based on analysis of the long-chain n-alkane C31. The absolute archaeal 16S rRNA gene abundance in subsurface sediments was highest in MIS 2, coincident with high sedimentation rates and high concentrations of total organic carbon. Soil Crenarchaeotic Group (SCG; Nitrososphaerales) species, the most abundant ammonia-oxidizing archaea in soils, increased dramatically during MIS 2, likely reflecting transport of terrestrial archaea during glacial periods with high sedimentation rates. Co-occurrence network analyses indicated significant association of SCG archaea with benthic deep-sea microbes such as Bathyarchaeota and Thermoprofundales in MIS 2 and MIS 3, suggesting potential interactions among these archaeal groups. Meanwhile, Thermoprofundales abundance was positively correlated with total organic carbon (TOC), along with n-alkane C31 and sedimentation rate, indicating that Thermoprofundales may be particularly important in processing of organic carbon in deep-sea sediments. Collectively, these results demonstrate that the composition of heterotrophic benthic archaea in the South China Sea may be influenced by terrestrial organic input in tune with glacial-interglacial cycles, suggesting a plausible link between global climate change and microbial population dynamics in deep-sea marine sediments.

Project description:Microplastics in aquatic environments provide novel habitats for surface-colonizing microorganisms. Given the continuing debate on whether substrate-specific properties or environmental factors prevail in shaping biofilm assemblages on microplastics, we examined the influence of substrate vs. spatial factors in the development of bacterial assemblages on polyethylene (PE), polystyrene (PS), wood, and seston and in the free-living fraction. Further, the selective colonization of microplastics by potential pathogens was investigated because among the bacterial species found in microplastic-associated biofilms are potentially pathogenic Vibrio spp. Due to their persistence and great dispersal potential, microplastics could act as vectors for these potential pathogens and for biofilm assemblages in general. Incubation experiments with these substrates were conducted for 7 days during a summer cruise along the eastern Baltic Sea coastline in waters covering a salinity gradient of 4.5-9 PSU. Bacterial assemblages were analyzed using 16S rRNA-gene amplicon sequencing, distance-based redundancy analyses, and the linear discriminant analysis effect size method to identify taxa that were significantly more abundant on the plastics. The results showed that the sample type was the most important factor structuring bacterial assemblages overall. Surface properties were less significant in differentiating attached biofilms on PE, PS, and wood; instead, environmental factors, mainly salinity, prevailed. A potential role for inorganic-nutrient limitations in surface-specific attachment was identified as well. Alphaproteobacteria (Sphingomonadaceae, Devosiaceae, and Rhodobacteraceae) and Gammaproteobacteria (Alteromonadaceae and Pseudomonas) were distinctive for the PE- and PS-associated biofilms. Vibrio was more abundant on the PE and PS biofilms than on seston, but its abundances were highest on wood and positively correlated with salinity. These results corroborate earlier findings that microplastics constitute a habitat for biofilm-forming microorganisms distinct from seston, but less from wood. In contrast to earlier reports of low Vibrio numbers on microplastics, these results also suggest that vibrios are early colonizers of surfaces in general. Spatial as well as temporal dynamics should therefore be considered when assessing the potential of microplastics to serve as vectors for bacterial assemblages and putative pathogens, as these parameters are major drivers of biofilm diversity.
