Linear and nonlinear chromatic integration in the mouse retina.

ABSTRACT: The computations performed by a neural circuit depend on how it integrates its input signals into an output of its own. In the retina, ganglion cells integrate visual information over time, space, and chromatic channels. Unlike the former two, chromatic integration is largely unexplored. Analogous to classical studies of spatial integration, we here study chromatic integration in mouse retina by identifying chromatic stimuli for which activation from the green or UV color channel is maximally balanced by deactivation through the other color channel. This reveals nonlinear chromatic integration in subsets of On, Off, and On-Off ganglion cells. Unlike the latter two, nonlinear On cells display response suppression rather than activation under balanced chromatic stimulation. Furthermore, nonlinear chromatic integration occurs independently of nonlinear spatial integration, depends on contributions from the rod pathway and on surround inhibition, and may provide information about chromatic boundaries, such as the skyline in natural scenes.

Chromatic bipolar cell pathways in the mouse retina.

Project description:Like most mammals, mice feature dichromatic color vision based on short (S) and middle (M) wavelength-sensitive cone types. It is thought that mammals share a retinal circuit that in dichromats compares S- and M-cone output to generate blue/green opponent signals, with bipolar cells (BCs) providing separate chromatic channels. Although S-cone-selective ON-BCs (type 9 in mouse) have been anatomically identified, little is known about their counterparts, the M-cone-selective OFF-BCs. Here, we characterized cone connectivity and light responses of selected mouse BC types using immunohistochemistry and electrophysiology. Our anatomical data indicate that four (types 2, 3a/b, and 4) of the five mouse OFF-BCs indiscriminately contact both cone types, whereas type 1 BCs avoid S-cones. Light responses showed that the chromatic tuning of the BCs strongly depended on their position along the dorsoventral axis because of the coexpression gradient of M- and S-opsin found in mice. In dorsal retina, where coexpression is low, most type 2 cells were green biased, with a fraction of cells (? 14%) displaying strongly blue-biased responses, likely reflecting S-cone input. Type 1 cells were also green biased but did not comprise blue-biased "outliers," consistent with type 1 BCs avoiding S-cones. We therefore suggest that type 1 represents the green OFF pathway in mouse. In addition, we confirmed that type 9 BCs display blue-ON responses. In ventral retina, all BC types studied here displayed similar blue-biased responses, suggesting that color vision is hampered in ventral retina. In conclusion, our data support an antagonistically organized blue/green circuit as the common basis for mammalian dichromatic color vision.

Nonlinear Spatiotemporal Integration by Electrical and Chemical Synapses in the Retina.

Project description:Electrical and chemical synapses coexist in circuits throughout the CNS. Yet, it is not well understood how electrical and chemical synaptic transmission interact to determine the functional output of networks endowed with both types of synapse. We found that release of glutamate from bipolar cells onto retinal ganglion cells (RGCs) was strongly shaped by gap-junction-mediated electrical coupling within the bipolar cell network of the mouse retina. Specifically, electrical synapses spread signals laterally between bipolar cells, and this lateral spread contributed to a nonlinear enhancement of bipolar cell output to visual stimuli presented closely in space and time. Our findings thus (1) highlight how electrical and chemical transmission can work in concert to influence network output and (2) reveal a previously unappreciated circuit mechanism that increases RGC sensitivity to spatiotemporally correlated input, such as that produced by motion.

Encoding of luminance and contrast by linear and nonlinear synapses in the retina.

Project description:Understanding how neural circuits transmit information is technically challenging because the neural code is contained in the activity of large numbers of neurons and synapses. Here, we use genetically encoded reporters to image synaptic transmission across a population of sensory neurons-bipolar cells in the retina of live zebrafish. We demonstrate that the luminance sensitivities of these synapses varies over 10(4) with a log-normal distribution. About half the synapses made by ON and OFF cells alter their polarity of transmission as a function of luminance to generate a triphasic tuning curve with distinct maxima and minima. These nonlinear synapses signal temporal contrast with greater sensitivity than linear ones. Triphasic tuning curves increase the dynamic range over which bipolar cells signal light and improve the efficiency with which luminance information is transmitted. The most efficient synapses signaled luminance using just 1 synaptic vesicle per second per distinguishable gray level.

Nonlinear visuoauditory integration in the mouse superior colliculus.

Project description:Sensory information from different modalities is processed in parallel, and then integrated in associative brain areas to improve object identification and the interpretation of sensory experiences. The Superior Colliculus (SC) is a midbrain structure that plays a critical role in integrating visual, auditory, and somatosensory input to assess saliency and promote action. Although the response properties of the individual SC neurons to visuoauditory stimuli have been characterized, little is known about the spatial and temporal dynamics of the integration at the population level. Here we recorded the response properties of SC neurons to spatially restricted visual and auditory stimuli using large-scale electrophysiology. We then created a general, population-level model that explains the spatial, temporal, and intensity requirements of stimuli needed for sensory integration. We found that the mouse SC contains topographically organized visual and auditory neurons that exhibit nonlinear multisensory integration. We show that nonlinear integration depends on properties of auditory but not visual stimuli. We also find that a heuristically derived nonlinear modulation function reveals conditions required for sensory integration that are consistent with previously proposed models of sensory integration such as spatial matching and the principle of inverse effectiveness.

A chromatic feature detector in the retina signals visual context changes.

Project description:The retina transforms patterns of light into visual feature representations supporting behaviour. These representations are distributed across various types of retinal ganglion cells (RGCs), whose spatial and temporal tuning properties have been studied extensively in many model organisms, including the mouse. However, it has been difficult to link the potentially nonlinear retinal transformations of natural visual inputs to specific ethological purposes. Here, we discover a nonlinear selectivity to chromatic contrast in an RGC type that allows the detection of changes in visual context. We trained a convolutional neural network (CNN) model on large-scale functional recordings of RGC responses to natural mouse movies, and then used this model to search in silico for stimuli that maximally excite distinct types of RGCs. This procedure predicted centre colour opponency in transient suppressed-by-contrast (tSbC) RGCs, a cell type whose function is being debated. We confirmed experimentally that these cells indeed responded very selectively to Green-OFF, UV-ON contrasts. This type of chromatic contrast was characteristic of transitions from ground to sky in the visual scene, as might be elicited by head or eye movements across the horizon. Because tSbC cells performed best among all RGC types at reliably detecting these transitions, we suggest a role for this RGC type in providing contextual information (i.e. sky or ground) necessary for the selection of appropriate behavioural responses to other stimuli, such as looming objects. Our work showcases how a combination of experiments with natural stimuli and computational modelling allows discovering novel types of stimulus selectivity and identifying their potential ethological relevance.

Chromatic cues for the sign of defocus in the peripheral retina.

Project description:Detecting optical defocus at the retina is crucial for accurate accommodation and emmetropization. However, the optical characteristics of ocular defocus are not fully understood. To bridge this knowledge gap, we simulated polychromatic retinal image quality by considering both the monochromatic wavefront aberrations and chromatic aberrations of the eye, both in the fovea and the periphery (nasal visual field). Our study revealed two main findings: (1) chromatic and monochromatic aberrations interact to provide a signal to the retina (chromatic optical anisotropy) to discern positive from negative defocus and (2) that chromatic optical anisotropy exhibited notable differences among refractive error groups (myopes, emmetropes and hyperopes). These findings could enhance our understanding of the underlying mechanisms of defocus detection and their subsequent implications for myopia control therapies. Further research is needed to explore the retinal architecture's ability to utilize the optical signals identified in this study.

Data-Driven Models of Efficient Chromatic Coding in the Outer Retina.

Project description:Recent experimental work on zebrafish has shown the in vivo activity of photoreceptors and horizontal cells (HCs) as a function of the stimulus spectrum, highlighting the appearance of chromatic-opponent signals at their first synaptic connection. Altogether with the observed lack of excitatory intercone connections, these findings suggest that the mechanism yielding early color opponency in zebrafish is dominated by inhibitory feedback. We propose a neuronal population model based on zebrafish retinal circuitry to investigate whether networks with predominantly inhibitory feedback are more advantageous in encoding chromatic information than networks with mixed excitatory and inhibitory mechanisms. We show that networks with dominant inhibitory feedback exhibit a unique and reliable encoding of chromatic information. In contrast, this property is not guaranteed in networks with strong excitatory intercone connections, exhibiting bistability. These findings provide a theoretical explanation for the absence of excitatory intercone couplings in zebrafish color circuits. In addition, our study shows that these networks, with only one type of horizontal cell, are suitable to encode most of the variance from the zebrafish environment. However, at least two successive layers of inhibitory neurons are needed to reach the optimum. Finally, we contrast the encoding performance of networks with different opsin sensitivities, showing an improvement of only 13% compared with zebrafish, suggesting that the zebrafish retina is adapted to encode color information from its habitat efficiently.

Nonlinear processing with linear optics.

Project description:Deep neural networks have achieved remarkable breakthroughs by leveraging multiple layers of data processing to extract hidden representations, albeit at the cost of large electronic computing power. To enhance energy efficiency and speed, the optical implementation of neural networks aims to harness the advantages of optical bandwidth and the energy efficiency of optical interconnections. In the absence of low-power optical nonlinearities, the challenge in the implementation of multilayer optical networks lies in realizing multiple optical layers without resorting to electronic components. Here we present a novel framework that uses multiple scattering, and which is capable of synthesizing programmable linear and nonlinear transformations concurrently at low optical power by leveraging the nonlinear relationship between the scattering potential, represented by data, and the scattered field. Theoretical and experimental investigations show that repeating the data by multiple scattering enables nonlinear optical computing with low-power continuous-wave light. Moreover, we empirically find that scaling of this optical framework follows a power law.

Ex Vivo Integration of Human Stem Retinal Ganglion Cells into the Mouse Retina.

Project description:Cell replacement therapies may be key in achieving functional recovery in neurodegenerative optic neuropathies diseases such as glaucoma. One strategy that holds promise in this regard is the use of human embryonic stem cell and induced pluripotent stem-derived retinal ganglion cells (hRGCs). Previous hRGC transplantation studies have shown modest success. This is in part due to the low survival and integration of the transplanted cells in the host retina. The field is further challenged by mixed assays and outcome measurements that probe and determine transplantation success. Thefore, we have devised a transplantation assay involving hRGCs and mouse retina explants that bypasses physical barriers imposed by retinal membranes. We show that hRGC neurites and somas are capable of invading mouse explants with a subset of hRGC neurites being guided by mouse RGC axons. Neonatal mouse retina explants, and to a lesser extent, adult explants, promote hRGC integrity and neurite outgrowth. Using this assay, we tested whether suppmenting cultures with brain derived neurotrophic factor (BDNF) and the adenylate cyclase activator, forskolin, enhances hRGC neurite integration, neurite outgrowth, and integrity. We show that supplementing cultures with a combination BDNF and forskolin strongly favors hRGC integrity, increasing neurite outgrowth and complexity as well as the invasion of mouse explants. The transplantation assay presented here is a practical tool for investigating strategies for testing and optimizing the integration of donor cells into host tissues.

Migration, integration and maturation of photoreceptor precursors following transplantation in the mouse retina.

Project description:Retinal degeneration leading to loss of photoreceptors is a major cause of untreatable blindness. Recent research has yielded definitive evidence for restoration of vision following the transplantation of rod photoreceptors in murine models of blindness, while advances in stem cell biology have enabled the generation of transplantable photoreceptors from embryonic stem cells. Importantly, the amount of visual function restored is dependent upon the number of photoreceptors that migrate correctly into the recipient retina. The developmental stage of the donor cells is important for their ability to migrate; they must be immature photoreceptor precursors. Little is known about how and when donor cell migration, integration, and maturation occurs. Here, we have performed a comprehensive histological analysis of the 6-week period following rod transplantation in mice. Donor cells migrate predominately as single entities during the first week undergoing a stereotyped sequence of morphological changes in their translocation from the site of transplantation, through the interphotoreceptor matrix and into the recipient retina. This includes initial polarization toward the outer nuclear layer (ONL), followed by formation of an apical attachment and rudimentary segment during migration into the ONL. Strikingly, acquisition of a nuclear architecture typical of mature rods was accelerated compared with normal development and a feature of migrating cells. Once within the ONL, precursors formed synaptic-like structures and outer segments in accordance with normal maturation. The restoration of visual function mediated by transplanted photoreceptors correlated with the later expression of rod ?-transducin, achieving maximal function by 5 weeks.
