Evaluation of Coated Biochar as an Intestinal Binding Agent for Skatole and Indole in Male Intact Finishing Pigs.

ABSTRACT: The ban on piglet castration without anaesthesia poses a challenge for the meat industry since alternatives ensuring the production of flawless pork have to be established. The aim of this study was to evaluate the effects of biochar on skatole and indole concentration in faeces and plasma on a small scale in finishing boars to prove whether biochar was suitable for use in commercial pork production. Moreover, it was investigated whether biochar affects faecal properties or the performance. For a four-week trial period, 54 boars (bodyweight 97.2 ± 6.88 kg) were divided into three groups. The control (BC0) received no dietary biochar, one group received a diet containing 4% coated biochar (corresponding to 2% pure biochar) for the final two experimental weeks (BC2), and another group for the entire four weeks (BC4), respectively, prior to slaughter. Skatole and indole concentrations were measured in faeces and plasma at the beginning, in the middle and at the end of the trial. Mean skatole concentrations did not differ between groups, but in BC2 faecal skatole was significantly decreased at day 26, whereas in BC4 initial and final faecal skatole levels did not differ. At day 15 and 26, the faecal dry matter content was significantly higher in pigs fed the biochar diet (p < 0.05).

Project description:Skatole, a strong fecal odor substance, is generated through microbial degradation of tryptophan in the animal hindgut. It easily accumulates in adipose tissue and affects meat quality. In this study, the effect of mulberry leaf supplementation on skatole in finishing pigs was studied. In a 35-day trial, 20 finishing pigs (barrows and gilts) were fed with a basal diet or basal diet with 6% mulberry leaves. Growth performance of the pigs (n = 10) was automatically recorded by a performance-testing feeder system and 8 pigs in each treatment were slaughtered and sampled for the remaining tests. Skatole and short-chain fatty acids were detected using HPLC and gas chromatography, respectively. Fecal microbiota were analyzed using 16S rRNA gene sequencing. The metabolomics analysis of feces and serum was performed with UHPLC-MS/MS. The major cytochrome P450 (CYP) enzymes that catalyze skatole degradation in the liver were tested by using RT-PCR and Western blot. Effects of major bioactive compounds in mulberry leaves on the CYP genes were verified in the hepatic cell line HepG2 in an in vitro test (n = 3). In finishing pigs, mulberry leaf supplementation had no significant effect on the average daily gain, average daily feed intake, and feed conversion ratio (P > 0.05), but reduced skatole levels in feces, serum, and backfat (P < 0.05), and increased acetic acid levels in feces (P = 0.027). Mulberry leaf supplementation decreased the relative abundance of the skatole-producing bacteria Megasphaera and Olsenella (P < 0.05). Indole-3-acetic acid, the intermediate that is essential for skatole production, was significantly reduced in feces by mulberry leaf supplementation (P < 0.05) and was positively correlated with skatole content in feces (P = 0.004). In pigs treated with mulberry leaves, liver CYP1A1 expression was increased (P < 0.05) and was negatively correlated with skatole content in backfat (P = 0.045). The in vitro test demonstrated that mulberry leaf polyphenols and polysaccharides could directly stimulate CYP1A1 expression in hepatic cells. These findings suggest that mulberry leaf supplementation reduces skatole production and deposition in finishing pigs by regulating the gut microbiota and promoting skatole degradation in liver.

Project description:Tail biting lesions are a potential measure of on-farm animal welfare, as a large range of stressors increase the risk for tail biting outbreaks. Further, tail biting is a major challenge, as lesions due to tail biting decrease animal welfare and health, as well as production efficiency and carcass quality. The aim of this study was to suggest a tail scoring system for use at slaughterhouses processing undocked pigs, and to link tail lesion scores to meat inspection data. A further aim was to suggest a definition for an intact enough tail. To validate the suggested scoring system we assessed tails before and after scalding and compared results to pathological examinations. In total, 14,433 tails were scored, and 117 tails were collected for pathological examination. After scalding, 49.2% of all tails were scored as fully intact. Of tails with lesions 2.5% were scored as having major acute wounds (>2 cm), while 11.6% had minor acute wounds (<2 cm), and 36.7% healed lesions. Intact tails were on average 31.5 cm (SD 2.5 cm) long. Lesion scored at the slaughter-line agreed well with the pathological assessment. Tail lesions were associated with several meat inspection findings: tails with more severe lesions and of shorter length increased the risk for meat inspection findings to a higher degree. A detailed lesion scoring method helps to identify carcasses at risk for condemnations, as well as being a potential method for on-farm welfare estimation. We suggest that a system for scoring tail lesions in undocked pigs should utilize a combination of scoring of the lesion and measuring the tail length. As bite marks or bruises on an otherwise intact tail were not a concern for meat hygiene, we suggest the definition of an intact enough tail could allow the inclusion of tails with these mild changes. Meat inspection findings in carcasses with tails scored as healed, but with no fresh lesions, and with more than 75% of the average intact length remaining were rather similar to those of fully intact tails. Based on these findings we suggest that a tail of this length, and with no visible fresh lesions could also be considered intact enough.

Project description:The effect of high levels of dietary chicory roots (25%) and intracecal exogenous butyrate infusion on skatole formation and gut microbiota was investigated in order to clarify the mechanisms underlying the known reducing effect of chicory roots on skatole production in entire male pigs. A Latin square design with 3 treatments (control, chicory, and butyrate), 3 periods, and 6 animals was carried out. Chicory roots showed the lowest numerical levels of skatole in both feces and plasma and butyrate infusion the highest. In the chicory group, an increased abundance of the skatole-producing bacterium Olsenella scatoligenes compared to the control group (P = 0.06), and a numerically higher relative abundance of Olsenella than for the control and butyrate groups, was observed. Regarding butyrate-producing bacteria, the chicory group had lower abundance of Roseburia but a numerically higher abundance of Megasphaera than the control group. Lower species richness was found in the chicory group than in the butyrate group. Moreover, beta diversity revealed that the chicory group formed a distinct cluster, whereas the control and butyrate groups clustered more closely to each other. The current data indicated that the skatole-reducing effect of chicory roots is neither via inhibition of cell apoptosis by butyrate nor via suppression of skatole-producing bacteria in the pig hindgut. Thus, the mode of action is most likely through increased microbial activity with a corresponding high incorporation of amino acids into bacterial biomass, and thereby suppressed conversion of tryptophan into skatole, as indicated in the literature.IMPORTANCE Castration is practiced to avoid the development of boar taint, which negatively affects the taste and odor of pork, and undesirable aggressive behavior. Due to animal welfare issues, alternatives to surgical castration are sought, though. Boar taint is a result of high concentrations of skatole and androstenone in back fat. Skatole is produced by microbial fermentation in the large intestine, and therefore, its production can be influenced by manipulation of the microbiota. Highly fermentable dietary fiber reduces skatole production. However, various theories have been proposed to explain the mode of action. In order to search for other alternatives, more efficient or less expensive, to reduce skatole via feeding, it is important to elucidate the mechanism behind the observed effect of highly fermentable dietary fiber on skatole. Our results indicate that highly fermentable dietary fiber does not affect skatole production by reducing the number of skatole-producing bacteria or stimulating butyrate production in the large intestine.

Project description:This study investigated the different addition levels of iron (Fe) in growing-finishing pigs and the effect of different Fe levels on growth performance, hematological status, intestinal barrier function, and intestinal digestion. A total of 1,200 barrows and gilts ([Large White × Landrace] × Duroc) with average initial body weight (BW; 27.74 ± 0.28 kg) were housed in 40 pens of 30 pigs per pen (gilts and barrows in half), blocked by BW and gender, and fed five experimental diets (eight replicate pens per diet). The five experimental diets were control diet (basal diet with no FeSO4 supplementation), and the basal diet being supplemented with 150, 300, 450, or 600 mg/kg Fe as FeSO4 diets. The trial lasted for 100 d and was divided into the growing phase (27 to 60 kg of BW) for the first 50 d and the finishing phase (61 to 100 kg of BW) for the last 50 d. The basal diet was formulated with an Fe-free trace mineral premix and contained 203.36 mg/kg total dietary Fe in the growing phase and 216.71 mg/kg in the finishing phase based on ingredient contributions. And at the end of the experiment, eight pigs (four barrows and four gilts) were randomly selected from each treatment (selected one pig per pen) for digesta, blood, and intestinal samples collection. The results showed that the average daily feed intake (P = 0.025), average daily gain (P = 0.020), and BW (P = 0.019) increased linearly in the finishing phase of pigs fed with the diets containing Fe. On the other hand, supplementation with different Fe levels in the diet significantly increased serum iron and transferrin saturation concentrations (P < 0.05), goblet cell numbers of duodenal villous (P < 0.001), and MUC4 mRNA expression (P < 0.05). The apparent ileal digestibility (AID) of amino acids (AA) for pigs in the 450 and 600 mg/kg Fe groups was greater (P < 0.05) than for pigs in the control group. In conclusion, dietary supplementation with 450 to 600 mg/kg Fe improved the growth performance of pigs by changing hematological status and by enhancing intestinal goblet cell differentiation and AID of AA.
