ABSTRACT: Soy protein concentrate improves nutrient utilization and growth performance compared to soybean meal, and diets with a low crude protein (CP) level decreases diarrhoea. The objectives were to (1) test a low CP diet based on different soy products, and (2) to test a very-low CP diet (15.1%) with amino acids (AA) on diarrhoea and productivity. A total of 5,635 weaned pigs (~28 days), were assigned to five dietary treatments; PC (positive control): Standard CP levels (192, 189, 191 g/kg CP) with 2500 ppm ZnO; NC (negative control): Same as PC without ZnO; SP (Soy protein concentrate): Low CP levels (176, 174, 191 g/kg CP); SB (Soybean meal): Low CP levels (177, 176, 191 g/kg CP); and XLA (X-low CP + AA): Very low CP levels (154, 151, 191 g/kg CP) with AA. The PC and XLA diets reduced diarrhoea by 41 and 61%, respectively, compared to the NC group, while no difference between SB and SP were observed. The XLA diet reduced feed intake and daily gain compared with PC and NC, where SP, SB, and XLA had a poorer feed conversion compared with PC. Conclusively, the SP and SB low-protein diets did not reduce diarrhoea or growth performance, whereas the XLA diet decreased both diarrhoea and performance.

Project description:The present study investigated the growth performance, feed utilization, intestinal morphology, and microbiota communities of juvenile large yellow croaker (Larimichthys crocea) fed diets containing different proportions of soy protein concentrate (SPC) (0, 15%, 30%, and 45%, namely FM, SPC15, SPC30, and SPC45) as a substitute for fish meal (FM) for 8 weeks. The weight gain (WG) and specific growth rate (SGR) in fish fed SPC45 were significantly lower than those fed FM and SPC15 but not differ with these fed SPC30. The feed efficiency (FE) and protein efficiency ratio (PER) decreased sharply when the dietary SPC inclusion level was higher than 15%. The activity of alanine aminotransferase (ALT) and expression of alt and aspartate aminotransferase (ast) were significantly higher in fish fed SPC45 than those fed FM. The activity and mRNA expression of acid phosphatase were opposite. The villi height (VH) in distal intestine (DI) showed a significant quadratic response to increasing dietary SPC inclusion levels and was highest in SPC15. The VH in proximal intestine, middle intestine decreased significantly with increasing dietary SPC levels. The 16S rRNA sequences in intestine revealed that fish fed SPC15 had higher bacterial diversity and abundance of Phylum Firmicutes such as order Lactobacillales and order Rhizobiaceae than those fed other diets. Genus vibrio, family Vibrionaceae and order Vibrionales within phylum Proteobacteria were enriched in fish fed FM and SPC30 diets. Tyzzerella and Shewanella that belongs to phylum Firmicutes and Proteobacteria, respectively, were enriched in fish fed SPC45 diet. Our results indicated that SPC replacing more than 30% FM could lead to lower quality diet, retard growth performance, ill health, disordered intestine structure, and microbiota communities. Tyzzerella could be the bacteria indicator of intestinal in large yellow croaker fed low quality diet due to high SPC content. Based on the quadratic regression analysis of WG, the best growth performance could be observed when the replacement of FM with SPC was 9.75%.

Project description:Antibiotic use has been limited in U.S. swine production. Therefore, the objective was to determine whether supplementing l-glutamine at cost-effective levels can replace dietary antibiotics to improve piglet welfare and productivity following weaning and transport. Based on previous research, we hypothesized that withholding dietary antibiotics would negatively affect pigs while diet supplementation with 0.20% l-glutamine (GLN) would have similar effects on pig performance and health as antibiotics. Mixed sex piglets (N = 480; 5.62 ± 0.06 kg BW) were weaned (18.4 ± 0.2 d of age) and transported for 12 h in central Indiana, for 2 replicates, during the summer of 2016 and the spring of 2017. Pigs were blocked by BW and allotted to 1 of 3 dietary treatments (n = 10 pens/dietary treatment/replicate [8 pigs/pen]); antibiotics (A; chlortetracycline [441 ppm] + tiamulin [38.6 ppm]), no antibiotics (NA), or GLN fed for 14 d. On days 15 to 34, pigs were provided common antibiotic-free diets in 2 phases. Data were analyzed using PROC MIXED in SAS 9.4. Day 14 BW and days 0 to 14 ADG were greater (P = 0.01) for A (5.6% and 18.5%, respectively) and GLN pigs (3.8% and 11.4%, respectively) compared with NA pigs, with no differences between A and GLN pigs. Days 0 to 14 ADFI increased for A (P < 0.04; 9.3%) compared with NA pigs; however, no differences were detected when comparing GLN with A and NA pigs. Once dietary treatments ceased, no differences (P > 0.05) in productivity between dietary treatments were detected. On day 13, plasma tumor necrosis factor alpha (TNF-α) was reduced (P = 0.02) in A (36.7 ± 6.9 pg/mL) and GLN pigs (40.9 ± 6.9 pg/mL) vs. NA pigs (63.2 ± 6.9 pg/mL). Aggressive behavior tended to be reduced overall (P = 0.09; 26.4%) in GLN compared with A pigs, but no differences were observed between A and GLN vs. NA pigs. Huddling, active, and eating/drinking behaviors were increased overall (P < 0.02; 179%, 37%, and 29%, respectively) in the spring replicate compared with the summer replicate. When hot carcass weight (HCW) was used as a covariate, loin depth and lean percentage were increased (P = 0.01; 4.0% and 1.1%, respectively) during the spring replicate compared with the summer replicate. In conclusion, GLN supplementation improved pig performance and health after weaning and transport similarly to A across replicates; however, the positive effects of A and GLN were diminished when dietary treatments ceased.

Project description:The residues left over from cold-pressing oilseeds and fruits offer a promising source of plant proteins, serving as an alternative resource within the circular economy framework. This research aims to develop protein concentrates (PCs) from pistachio meal with enhanced functional properties using ultrasound -assisted extraction (UAE). A response surface methodology was employed to optimize UAE considering power (200-600 W), time (5-25 min), and pH (8-11). Through a Box-Behnken experimental design, the optimal extraction conditions were determined as 595 W, pH 11, and 19 min, targeting maximal protein extraction yield (R2: 0.9359, desirability of 0.993). Under these ideal conditions, UAE achieved higher protein extraction yield (51%), compared to the conventional alkaline extraction (AE) (39%). Protein recovery rate increased from 33.1% to 44% by UAE. PCs obtained through the AE (PC-AE) and optimal UAE (PC-UAE) were evaluated for their functional, structural, and morphological properties. UAE significantly improved the emulsion activity index of the PC-UAE by 34.9% (p < 0.05), whereas solubility and foaming activity showed slight increases compared to PC-AE (p > 0.05). US-induced size reduction as observed by smaller and irregular particles in morphological analysis. The Fourier-transform infrared spectroscopy spectra, analyzed using a curve-fitting approach on the second derivative, revealed hidden peaks that highlighted changes in the secondary structure of the PCs, including slight shifts in characteristic wavenumbers and variations in the proportional amounts of β-sheet, α-helix, and random coil structures. This study demonstrated that UAE achieved similar protein purity as AE while producing larger quantities through higher recovery of proteins with improved functional properties. PRACTICAL APPLICATION: Ultrasound-assisted alkaline extraction significantly boosts protein extraction yield and recovery from pistachio meal and enhances functional properties (especially emulsion activity) of the extracted proteins. These improvements make the UAE protein concentrates suitable for applications in food formulations, such as plant-based beverages and emulsified products. UAE offers a sustainable approach to valorize oilseed residues in the context of circular economy.

Project description:Post-weaning diarrhoea (PWD) is a frequent, multifactorial disease of piglets leading to increased mortality rates and high economic losses. Due to the emergence of multi-resistant Escherichia coli isolates and the ban of zinc oxide (ZnO) in the EU since June 2022, alternative measures to prevent PWD are urgently needed. While an abundance of feed supplements is described to prevent PWD, there are hardly any studies reflecting the current situation of PWD in the field. Thus, we aimed to identify differences in management practices, housing and feeding strategies between farms with PWD and farms without PWD. Data were personally collected using a semi-structured questionnaire in 257 Austrian piglet-producing farms. Farms with PWD in more than 10% of all weaned groups within twelve months prior to data collection were defined as case farms (n = 101), while the remaining 136 farms were defined as control farms. Data from 237 farms and 69 explanatory variables were analysed via penalized binary logistic regression using elastic-net in 100 different splits into randomly selected training and test datasets (80:20). Treatment with ZnO and/or colistin (136 farms) was negatively associated with PWD in all splits and had the biggest estimated absolute log odds ratio out of all tested variables. Implementation of an all-in/all-out system in the nursery units and administration of probiotics or horseradish also had preventive effects in most splits (≥ 97%). A higher number of feeding phases for piglets within the first seven weeks of life and housing on fully slatted floors was associated negatively with the occurrence of PWD as well in > 95% of all splits. PWD was more likely to occur on farms having problems with neonatal diarrhoea or postpartum dysgalactia syndrome. While our data demonstrate that treatment with ZnO or colistin had the biggest statistical effect on PWD, we were able to identify other preventive measures like supplementation with probiotics or horseradish. Since implementation of all-in/all-out measures and fully slatted floors were also negatively associated with the occurrence of PWD on visited farms, we assume that reduction of bacterial load by the implementation of simple hygiene measures are still crucial to prevent PWD.
