Bioprospecting Fluorescent Plant Growth Regulators from Arabidopsis to Vegetable Crops.

ABSTRACT: The phytohormone auxin is involved in almost every process of a plant's life, from germination to plant development. Nowadays, auxin research connects synthetic chemistry, plant biology and computational chemistry in order to develop innovative and safe compounds to be used in sustainable agricultural practice. In this framework, we developed new fluorescent compounds, ethanolammonium p-aminobenzoate (HEA-pABA) and p-nitrobenzoate (HEA-pNBA), and investigated their auxin-like behavior on two main commercial vegetables cultivated in Europe, cucumber (Cucumis sativus) and tomato (Solanumlycopersicum), in comparison to the model plant Arabidopsis (Arabidopsis thaliana). Moreover, the binding modes and affinities of two organic salts in relation to the natural auxin indole-3-acetic acid (IAA) into TIR1 auxin receptor were investigated by computational approaches (homology modeling and molecular docking). Both experimental and theoretical results highlight HEA-pABA as a fluorescent compound with auxin-like activity both in Arabidopsis and the commercial cucumber and tomato. Therefore, alkanolammonium benzoates have a great potential as promising sustainable plant growth stimulators to be efficiently used in vegetable crops.

Project description:Evaluation of crop N status will assist optimal N management of intensive vegetable production. Simple procedures for monitoring crop N status such as petiole sap [NO 3(-)-N], leaf N content and soil solution [NO 3(-)] were evaluated with indeterminate tomato and muskmelon. Their sensitivity to assess crop N status throughout each crop was evaluated using linear regression analysis against nitrogen nutrition index (NNI) and crop N content. NNI is the ratio between the actual and the critical crop N contents (critical N content is the minimum N content necessary to achieve maximum growth), and is an established indicator of crop N status. Nutrient solutions with four different N concentrations (treatments N1-N4) were applied throughout each crop. Average applied N concentrations were 1, 5, 13 and 22 mmol L(-1) in tomato, and 2, 7, 13 and 21 mmol L(-1) in muskmelon. Respective rates of N were 23, 147, 421 and 672 kg N ha(-1) in tomato, and 28, 124, 245 and 380 kg N ha(-1) in muskmelon. For each N treatment in each crop, petiole sap [NO 3(-)-N] was relatively constant throughout the crop. During both crops, there were very significant (P < 0.001) linear relationships between both petiole sap [NO 3(-)-N] and leaf N content with NNI and with crop N content. In indeterminate tomato, petiole sap [NO 3(-)-N] was very strongly linearly related to NNI (R(2) = 0.88-0.95, P < 0.001) with very similar slope and intercept values on all dates. Very similar relationships were obtained from published data of processing tomato. A single linear regression (R(2) = 0.77, P < 0.001) described the relationship between sap [NO 3(-)-N] and NNI for both indeterminate and processing tomato, each grown under very different conditions. A single sap [NO 3(-)-N] sufficiency value of 1050 mg N L(-1) was subsequently derived for optimal crop N nutrition (at NNI = 1) of tomato grown under different conditions. In muskmelon, petiole sap [NO 3(-)-N] was strongly linearly related to NNI (R(2) = 0.75 - 0.88, P < 0.001) with very similar slope and intercept values for much of the crop (44-72 DAT, days after transplanting). A single linear relationship between sap [NO 3(-)-N] and NNI (R(2) = 0.77, P < 0.001) was derived for this period, but sap sufficiency values could not be derived for muskmelon as NNI values were >1. Relationships between petiole sap [NO 3(-)-N] with crop N content, and leaf N content with both NNI and crop N content had variable slopes and intercept values during the indeterminate tomato and the muskmelon crops. Soil solution [NO 3(-)] in the root zone was not a sensitive indicator of crop N status. Of the three systems examined for monitoring crop/soil N status, petiole sap [NO 3(-)-N] is suggested to be the most useful because of its sensitivity to crop N status and because it can be rapidly analysed on the farm.

Project description:Demand for agricultural crop continues to escalate in response to increasing population and damage of prime cropland for cultivation. Research interest is diverted to utilize soils with marginal plant production. Moisture stress has negative impact on crop growth and productivity. The plant growth promoting rhizobacteria (PGPR) and plant growth regulators (PGR) are vital for plant developmental process under moisture stress. The current study was carried out to investigate the effect of PGPR and PGRs (Salicylic acid and Putrescine) on the physiological activities of chickpea grown in sandy soil. The bacterial isolates were characterized based on biochemical characters including Gram-staining, P-solubilisation, antibacterial and antifungal activities and catalases and oxidases activities and were also screened for the production of indole-3-acetic acid (IAA), hydrogen cyanide (HCN) and ammonia (NH3). The bacterial strains were identified as Bacillus subtilis, Bacillus thuringiensis and Bacillus megaterium based on the results of 16S-rRNA gene sequencing. Chickpea seeds of two varieties (Punjab Noor-2009 and 93127) differing in sensitivity to drought were soaked for 3 h before sowing in fresh grown cultures of isolates. Both the PGRs were applied (150 mg/L), as foliar spray on 20 days old seedlings of chickpea. Moisture stress significantly reduced the physiological parameters but the inoculation of PGPR and PGR treatment effectively ameliorated the adverse effects of moisture stress. The result showed that chickpea plants treated with PGPR and PGR significantly enhanced the chlorophyll, protein and sugar contents. Shoot and root fresh (81%) and dry weights (77%) were also enhanced significantly in the treated plants. Leaf proline content, lipid peroxidation and antioxidant enzymes (CAT, APOX, POD and SOD) were increased in reaction to drought stress but decreased due to PGPR. The plant height (61%), grain weight (41%), number of nodules (78%) and pod (88%), plant yield (76%), pod weight (53%) and total biomass (54%) were higher in PGPR and PGR treated chickpea plants grown in sandy soil. It is concluded from the present study that the integrative use of PGPR and PGRs is a promising method and eco-friendly strategy for increasing drought tolerance in crop plants.

Project description:Domesticated grain crops evolved from wild plants under human cultivation, losing natural dispersal mechanisms to become dependent upon humans, and showing changes in a suite of other traits, including increasing seed size. There is tendency for seed enlargement during domestication to be viewed as the result of deliberate selection for large seeds by early farmers. However, like some other domestication traits, large seeds may have evolved through natural selection from the activities of people as they gathered plants from the wild, or brought them into cultivation in anthropogenic settings. Alternatively, larger seeds could have arisen via pleiotropic effects or genetic linkage, without foresight from early farmers, and driven by selection that acted on other organs or favored larger plants. We have separated these unconscious selection effects on seed enlargement from those of deliberate selection, by comparing the wild and domesticated forms of vegetable crops. Vegetables are propagated by planting seeds, cuttings, or tubers, but harvested for their edible leaves, stems, or roots, so that seed size is not a direct determinant of yield. We find that landrace varieties of seven vegetable crops have seeds that are 20% to 2.5-times larger than those of their closest wild relatives. These domestication effect sizes fall completely within the equivalent range of 14% to 15.2-times for grain crops, although domestication had a significantly larger overall effect in grain than vegetable crops. Seed enlargement in vegetable crops that are propagated vegetatively must arise from natural selection for larger seeds on the occasions when plants recruit from seed and are integrated into the crop gene pool, or via a genetic link to selection for larger plants or organs. If similar mechanisms operate across all species, then unconscious selection during domestication could have exerted stronger effects on the seed size of our staple crops than previously realized.
