Effects of L-Glutamine Supplementation during the Gestation of Gilts and Sows on the Offspring Development in a Traditional Swine Breed.

ABSTRACT: The use of amino acids during pregnancy, such as glutamine (Gln), seems to be a promising strategy in selected swine breeds to improve the offspring prenatal development. The main goal of the current study was to assess the development of the offspring from parity 1-3 sows of a traditional breed, which were supplemented with 1% glutamine after Day 35 of gestation, under farm conditions. A total of 486 (288 treated) piglets from 78 (46 treated) Iberian sows were used. At birth and slaughterhouse, fatty acid composition, metabolism, and mTOR pathway gene expression were analyzed. At birth, treated newborns showed greater amounts of specific amino acids in plasma, such as glutamine, asparagine, or alanine, and Σn-3 fatty acids in cellular membranes than control newborns. The expression of genes belonging to mTOR Complex 1 was also higher in treated piglets with normal birth-weight. However, these findings did not improve productive traits at birth or following periods in litters from supplemented gilts (parity 1) or sows (parities 2-3). Thus, further research is needed to properly understand the effects of prenatal glutamine supplementation, particularly in traditional swine breeds.

Project description:This study investigated the effects of supplementing sow diets with lysozyme during the late gestation to lactation stage on the performance of sows and their offspring. Sixty sows (Yorkshire × Landrace, 3 to 6 of parity) at day 85 of gestation were allocated to the following 3 dietary treatments: 1) sows fed a basal diet from late gestation to lactation (control, n = 20), 2) sows fed a basal diet with lysozyme 150 g/t (LZM 150, n = 20), and 3) sows fed a basal diet with lysozyme 300 g/t (LZM 300, n = 20). During the lactation period, sows fed diets containing lysozyme had increased average daily feed intake (ADFI) (P < 0.01) and decreased weaning-to-estrus interval (WEI, P < 0.05), but there were no significant effects on backfat during the trial among treatments. Sows fed lysozyme diets had increased (P < 0.05) serum concentration of total protein (TP) compared with those fed the control diets. Serum immunoglobulin M (IgM) of the sows increased (P < 0.05) on day 1 of lactation, immunoglobulin A (IgA) and interleukin-10 (IL-10) increased (P < 0.05) on day 7 of lactation, and immunoglobulin G (IgG) had a tendency to increase (P = 0.05) during the lactation. Milk concentration of IgA increased (P < 0.05) on day 1 and 7 of lactation and tended to be greater (P = 0.06) on day 21 of lactation. No significant differences among the dietary treatments were observed in placental tissue mRNA expression of interleukin-6 (IL-6), IL-10, tumor necrosis factor-α (TNF-α), polymeric immunoglobulin receptor (pIgR), or the concentrations of IL-6, IL-10, TNF-α, or secretory immunoglobulin A (sIgA). Moreover, there was a decrease (P < 0.05) in stillborn in sows fed lysozyme diets. The diarrhea rate decreased (P < 0.05) and serum concentrations of IgA, IgG, IgM, and IL-10 increased (P < 0.05) in piglets from sows fed the diets containing lysozyme compared with piglets from sows fed the control diet. The serum concentrations of TP increased (P < 0.05), and albumin (ALB) and globulin (GLB) had a tendency to increase (P = 0.08, P = 0.06) in piglets from sows fed the diets containing lysozyme compared with piglets from sows fed the control diet. In conclusion, this study indicates that feeding sows diets supplemented with lysozyme from the late gestation through lactation stage increased sow ADFI during the lactation, shortened the WEI, and improved the maternal and offspring health status as indicated by immunological characteristics and a reduced incidence of diarrhea in piglets.

Project description:Background:Mammary gland development during late gestation in gilts is a major factor that alters the composition of colostrum and growth performance of piglets. Plasma valine is taken up and metabolized extensively by the mammary gland; however, the effects of valine on mammary gland development during late gestation are still unclear. Thirty primiparous gilts were divided into three treatment groups (n =?10) and received one of the three diets starting on day 75 of gestation until the day of farrowing. The total dietary valine to lysine ratio of the three diets was 0.63 (LV), 0.73 (MV), and 0.93 (HV), respectively. Results:Dietary valine supplementation during late gestation did not affect (P >?0.05) the litter size and weight at farrowing; however, the piglet weight and average daily gain at weaning were linearly increased (P <?0.05) as the dietary valine increased. The highest piglet weight at weaning was observed when the gilts were provided the HV diet. Dietary valine supplementation linearly elevated (P <?0.05) protein, fat and solids-not-fat and some free amino acids content in colostrum. The concentration of prolactin in plasma of gilts was linearly increased in response to valine supplementation at days 1 and 10 of lactation (P <?0.05). Furthermore, with increasing dietary valine allowance, a linear increase (P <?0.05) was observed in the area of the lumen of alveolus and the content of DNA, RNA, and total protein in the mammary tissues at day 1 of lactation. Moreover, the protein expression of cyclin D1, p-mTOR, p-S6, and p-4EBP1 was also linearly increased (P <?0.05) in the mammary tissue at day 1 of lactation. However, no difference (P >?0.05) was observed in the indices related to mammary development and the mTOR signaling pathway at day 21 of lactation. Conclusion:The results revealed that increasing the total dietary valine to lysine ratio to 0.93 during late gestation significantly enhances the piglet weight and average daily gain at weaning probably due to improved development of mammary gland.

Project description:BackgroundThe creatine-creatine kinase-phosphocreatine (Cr-CK-PCr) system maintains intracellular ratios of ATP/ADP for support of cellular functions and has been characterized at the placental-uterine interface of rodents, primates, swine and sheep, and thus may support fetal development. This study determined effects of dietary supplementation of creatine (Cr) to gestating gilts on fetal development, the number and ratio of primary and secondary muscle fibers, and on protein expression in endometrium and fetal biceps-femoris muscle, respectively in fetal pigs on d 60 and d 90 of gestation.MethodsReproductively mature gilts were synchronized to estrus using Matrix, observed for estrus (d 0), and artificially inseminated 12 h and 36 h later. Gilts were individually housed and fed 0.86 kg of 14% crude protein diet twice daily that meets nutritional requirements for pregnant gilts. Gilts were assigned to either basal diet control (CON) group, or Cr supplemented group (provided 30 g Cr monohydrate daily) from d 10 to either d 60 or d 90 of gestation. Gilts were euthanized and hysterectomized on either d 60 or d 90 of gestation. These protocols were completed in two replicates, as gilts were bred in spring and euthanized in summer or bred in fall and euthanized in winter (n = 20 gilts/replicate). Litter size, crown-rump length, sex, and fetal weight was recorded. Three female and male fetuses closest to mean litter weight were selected to assess effects of treatment on weight of fetal brain, kidney, liver, spleen, and biceps-femoris muscle. Data were analyzed to determine effects of treatment, days of gestation, replicate, and sex on litter size, fetal measurements, and incidence of intrauterine growth restriction.ResultsDietary Cr supplementation increased fetal brain weight to body weight ratios on d 90 of gestation (P < 0.05) and fetal kidney weight to body weight ratios on d 60 of gestation (P < 0.01), while days of gestation had significant effect on expression of mitochondrial CK isoform in gilt endometria (P < 0.05).ConclusionsResults suggest that dietary supplementation of Cr in gestating gilts enhanced development of select fetal organs and contribute to understanding roles of the Cr-CK-PCr system in pregnancy.

Project description:Increased metabolic burdens in breeding sows, which are induced by elevated systemic oxidative stress, could increase the need for nucleotides to repair lymphocyte DNA damage; however, de novo synthesis of nucleotides may be insufficient to cover this increased need. This study investigated the effects of dietary nucleotides on milk composition, oxidative stress status, and the reproductive and lactational performance of sows. Forty multiparous sows were assigned to 2 dietary treatments (Control group, and 1 g/kg Nucleotides group) based on a randomized complete block design using their BW at 85 d of gestation as a block. Sows from 2 groups were fed a restricted diet during gestation and ad libitum during lactation. The experiment lasted from 85 d of gestation to 21 d of lactation. The reproductive performance of sows and the growth performance of suckling piglets were measured. Oxidative stress parameters and milk components were also analysed. Data were analyzed using contrasts in the MIXED procedure of SAS. Sows in the Nucleotides group consumed more feed during the first week (P < 0.01) and from 1 to 21 d (P < 0.05) of lactation than those in Control group. Correspondingly, the litter weight gain of piglets showed a tendency to increase from cross-fostering to 9 d (P = 0.09) and from cross-fostering to 20 d (P = 0.10) in the Nucleotides group relative to the Control group. Additionally, the Nucleotides group was higher (P < 0.01) than the Control group in the concentrations of uridine 5'monophosphate, guanosine 5'monophosphate, inosine 5'monophosphate, adenosine 5'monophosphate and total nucleotides in milk. Furthermore, the Nucleotides group was higher (P < 0.01) than the Control group in the serum levels of total antioxidant capacity (P < 0.01) for sows at 109 d of gestation and glutathione peroxidase for weaning piglets, but lower at the levels of thiobarbituric acid-reactive substances (P < 0.05) in serum of weaning piglets. This study indicated that maternal dietary nucleotides could promote piglet growth, probably due to the higher lactational feed intake and higher concentration of nucleotides in the milk of sows, and lower oxidative stress for both sows and piglets.

Project description:IntroductionThe effects of maternal addition of yeast cultures on offspring gut development and intestinal microorganisms are not yet known, so the aim of this study was to investigate the effects of maternal addition of yeast cultures to the diet of sows during late gestation and lactation on growth performance, antioxidant properties and intestinal microorganisms of offspring weaned piglets.Methods40 Landrace × Yorkshire sows (3-7 of parity) with similar backfat were randomly divided into two treatment groups: control diet (CON) and control diet +2.0 g/kg yeast culture (XPC), and the trial started on day 90 of gestation and ended on day 21 of lactation.ResultsThe results showed that maternal addition of yeast culture significantly increased weaned piglet weight and mean daily gain (p < 0.05), with a tendency to increase litter weight gain (p = 0.083) and liver weight (p = 0.076) compared to the control group. The content of thymus malondialdehyde (MDA) was significantly higher (p < 0.05) and the content of colon total antioxidant capacity (T-AOC) was significantly lower (p < 0.05) in the offspring weaned piglets of the XPC group compared to the control group. The expression of thymus SOD1 and SOD2, spleen SOD1, jejunum SOD2, and colon GPX1, SOD1, and SOD2 were significantly downregulated in the XPC group of offspring weaned piglets compared with the control group (p < 0.05). The intestinal morphology and the content of short-chain fatty acids in colonic chyme did not differ between the two groups (p > 0.05). Compared with the control group, the XPC group significantly increased the relative abundance of colonic chyme Bacteroidetes (p < 0.05), tended to decrease the relative abundance of Lactobacillus (p = 0.078), and tended to increase the relative abundance of Alloprevotella (p = 0.055). The XPC group significantly upregulated Blautia and Fournierella (p < 0.05) and significantly downregulated Candidatus_Competibacter, Nitrospira, Dechloromonas, Haliangium, and Oscillospira (p < 0.05).DiscussionIn conclusion, maternal addition of yeast cultures improved the growth performance of offspring weaned piglets and changed the intestinal microbial community, but did not improve their antioxidant performance.
