The complete mitochondrial genome of the New Zealand parasitic blowfly Lucilia sericata (Insecta: Diptera: Calliphoridae).

ABSTRACT: In this study, the complete mitochondrial (mt) genome of the New Zealand parasitic blowfly Lucilia sericata (green bottle blowfly) field strain NZ_LucSer_NP was generated using next-generation sequencing technology. The length of complete the mt genome is 15,938 bp, with 39.4% A, 13.0% C, 9.3% G, and 38.2% T nucleotide distribution. The complete mt genome consists of 13 protein-coding genes (PCGs), two ribosomal RNAs, 22 transfer RNAs, and a 1124 bp non-coding region, similar to most metazoan mt genomes. Phylogenetic analysis showed that L. sericata NZ_LucSer_NP forms a monophyletic cluster with the remaining six Lucilia species and the Calliphoridae are polyphyletic. This study provides the first complete mt genome sequence for a L. sericata blowfly species derived from New Zealand to facilitate species identification and phylogenetic analysis.

Project description:In this study, the complete mitochondrial (mt) genome of the New Zealand parasitic blowfly Calliphora vicina (blue bottle blowfly) field strain NZ_CalVic_NP was generated using next-generation sequencing technology and annotated. The 16,518 bp mt genome consists of 13 protein-coding genes, two ribosomal RNAs, 22 transfer RNAs, and a 1689 bp non-coding region, similar to the two other available C. vicina and most metazoan mt genomes. Phylogenetic analysis showed that C. vicina NZ_CalVic_NP forms a monophyletic cluster with the remaining three Calliphorinae species. The complete mt genome sequence of C. vicina NZ_CalVic_NP is a resource to facilitate future species- and strain-level identification research and investigations into the evolutionary provenance within the Calliphoridae.

Development modeling of Lucilia sericata (Diptera: Calliphoridae).

Project description:The relationship between insect development and temperature has been well established and has a wide range of uses, including the use of blow flies for postmortem (PMI) interval estimations in death investigations. To use insects in estimating PMI, we must be able to determine the insect age at the time of discovery and backtrack to time of oviposition. Unfortunately, existing development models of forensically important insects are only linear approximations and do not take into account the curvilinear properties experienced at extreme temperatures. A series of experiments were conducted with Lucilia sericata, a forensically important blow fly species, that met the requirements needed to create statistically valid development models. Experiments were conducted over 11 temperatures (7.5 to 32.5 °C, at 2.5 °C) with a 16:8 L:D cycle. Experimental units contained 20 eggs, 10 g beef liver, and 2.5 cm of pine shavings. Each life stage (egg to adult) had five sampling times. Each sampling time was replicated four times, for a total of 20 measurements per life stage. For each sampling time, the cups were pulled from the chambers and the stage of each maggot was documented morphologically through posterior spiracle slits and cephalopharyngeal skeletal development. Data were normally distributed with the later larval stages (L3f, L3m) having the most variation within and transitioning between stages. The biological minimum was between 7.5 °C and 10 °C, with little egg development and no egg emergence at 7.5 °C. Temperature-induced mortality was highest from 10.0 to 17.5 °C and 32.5 °C. The development data generated illustrates the advantages of large datasets in modeling Lucilia sericata development and the need for curvilinear models in describing development at environmental temperatures near the biological minima and maxima.

Morphological identification of Lucilia sericata, Lucilia cuprina and their hybrids (Diptera, Calliphoridae).

Project description:Hybrids of Lucilia sericata and Lucilia cuprina have been shown to exist in previous studies using molecular methods, but no study has shown explicitly that these hybrids can be identified morphologically. Published morphological characters used to identify L. sericata and L. cuprina were reviewed, and then scored and tested using specimens of both species and known hybrids. Ordination by multi-dimensional scaling indicated that the species were separable, and that hybrids resembled L. cuprina, whatever their origin. Discriminant function analysis of the characters successfully separated the specimens into three unambiguous groups - L. sericata, L. cuprina and hybrids. The hybrids were morphologically similar irrespective of whether they were from an ancient introgressed lineage or more modern. This is the first evidence that hybrids of these two species can be identified from their morphology. The usefulness of the morphological characters is also discussed and photographs of several characters are included to facilitate their assessment.

The length of complete the mitochondrial genome is 16,279 bp, with 39.50% A, 13.20% C, 9.30% G, and 38.0% T nucleotide distribution. The complete mitochondrial genome consists of 13 protein-coding genes, 22 transfer RNAs, and 2 ribosomal RNAs likewise the most metazoan mitochondrial genomes. Furthermore, phylogenetic relationships of C. nigribarbis in the subfamily Calliphorinae investigated. The results suggested that C. vomitoria is the most related species to C. nigribarbis and the genus Calliphora is not monophyletic. This study provides the first complete mitochondrial genome sequence for the species.

Project description:Here, we present a 16S rRNA gene amplicon sequence data set and profiles demonstrating the bacterial diversity of larval and adult Lucilia sericata, collected from Ashhurst, New Zealand (May 2020). The two dominant genera among adult male and female L. sericata were Serratia and Morganella (phylum Proteobacteria), while the larvae were also dominated by the genera Lactobacillus, Carnobacterium, and Lactococcus (phylum Firmicutes).

The complete mitochondrial genome of Lucilia shenyangensis (Diptera: Calliphoridae).

Project description:Lucilia shenyangensis Fan, 1965 (Diptera: Calliphoridae) is of potential importance in epidemiology, veterinary medicine, and forensic entomology due to their necrophilous habit and behaviors associated with mammals. In this study, we report the complete mitochondrial genome (mitogenome) of L. shenyangensis. The mitogenome is 14,989 bp in length, comprising 13 protein-coding genes (PCGs), two ribosomal RNAs (rRNAs), 22 transfer RNAs (tRNAs), and a non-coding control region. The arrangement of genes is identical to that of the ancestral metazoan. Nucleotide composition revealed a high A/T bias, accounting for 76.50% total mitogenome nucleotides (A 39.2%, G 9.6%, C 14.0%, T 37.3%). Phylogenetic analysis indicated that L. shenyangensis was clear separated from other blow flies and emerged as the sister lineage to the rest species from genus Lucilia (L. illustris, L. sericata, L. coeruleiviridis, and L. porphyrina). The mitogenome data of L. shenyangensis could facilitate further evolutionary genetic researches on blow flies.

Canine Wound Myiasis Caused by Lucilia sericata (Diptera: Calliphoridae) in Korea.

Project description:Myiasis is a relatively common infection of animals kept as pets, although only 1 case of canine myiasis has been described so far in the Republic of Korea. In the present study, we report an additional case of canine wound myiasis with identification of its causative agent, Lucilia sericata. An 8-year-old male Siberian husky dog was referred with anorexia, vomiting, and diarrhea to the Chungbuk National University Veterinary Medical Center, Cheongju-si (city), Chungcheongbuk-do (province), Korea in July 2013. Physical examination indicated the patient had a deep wound filled with a maggot swarm as a left gluteal lesion. A total of 216 maggots were removed by forceps, and the wounded area was sponged with gauzes and disinfected with 70% alcohol and a povidone-iodine solution. After daily care and suturing the wound, the patient was discharged at day 19 after admission. Recovered worms possessed morphological characteristics similar to those of L. sericata, namely, a sub-cylindrical body with 6-8 lobed anterior spiracles, round shaped with a button surrounded by a peritremal ring with no gaps, and similar distances between dorsal, median, and outer papillae of the 12th segment. Additionally, cox1 partial sequences (528 bp) obtained in the present study showed 100% identity with those of L. sericata (GenBank no. KT272854.1). L. sericata is indicated as a pathogen of myiasis infection not only in humans, but also in animals kept as pets in Korea.

The Growth Model of Forensically Important Lucilia sericata (Meigen) (Diptera: Calliphoridae) in South Korea.

Project description:Development of forensically important Lucilia sericata (Meigen) was analyzed in South Korea. Rearing was replicated five times at seven constant temperatures between 20-35 °C to elucidate changes in accumulated degree hours, based on developmental stage and body length, and 2673 individuals were statistically analyzed. The results indicated that the optimum temperature, the base temperature, and the overall thermal constant were 22.31 °C (±1.21 °C, 95% CI), 9.07 °C, and 232.81 ± 23 (mean ± SD) accumulated degree days, respectively. In the minimum ADH models of each development stage, nonlinear regression graphs were parallel at the immature stages. Based on the scatter plot (n = 973) of immature stages using ADH values and body length, the logarithmic model using Log10ADH as the dependent variable was identified as the best fitting regression model. Additionally, the adjusted R2 value and mean square of error were 0.911 and 0.007, respectively. This is the first forensically focused study on the development of L.sericata for the estimation of minimum postmortem interval in South Korea. In future studies, we intend to study the development of other necrophagous fly species and to identify parameters for the determination of age at post-feeding and pupal stages.

Cloning, expression and molecular analysis of recombinant Netrin-A protein of Lucilia sericata Meigen (Diptera: Calliphoridae) larvae.

Project description:ObjectivesLucilia sericata (Diptera: Calliphoridae) is used in larval therapy for wound healing. Netrin-A is an enzyme secreted from the salivary glands of these larvae, and has a central role in neural regeneration and angiogenesis. This study aimed to produce the recombinant Netrin-A protein from Lucilia sericata larvae by the baculovirus expression vector system in the Sf9 insect cell line.MethodsThe coding sequence of Netrin-A was cloned, amplified in the pTG19 vector, and then cloned in the pFastBac HTA vector. It was then transformed into DH10Bac, and the recombinant Bacmid was subsequently transfected into Sf9 cells. The recombinant Netrin-A was purified by Ni-NTA agarose. The evaluation was done using SDS-PAGE and western blot, respectively. Finally, its concentration was calculated with the Bradford assay.ResultsThe molecular weight of this protein was 52 kDa with 404 amino acids. The signal peptide was located between amino acids 24 and 25. The concentration of Netrin-A was calculated to be 48.8 μg/ml. It reaffirmed the characterized gene codes of Lucilia sericata Netrin-A in a previous study.ConclusionsThe generation of recombinant Netrin-A could be used in larval therapy, and as a biomarker in certain diseases. The netrin-A of Lucilia sericata was unprecedentedly cloned and expressed in a eukaryotic cell line. Given that this larva is FDA-approved, and non-pathogenic, it conduces to research on the development of maggot therapy in future.
