First report of the bent seed gall nematode, Anguina agrostis (Steinbuch, 1799) Filipjev, 1936 from Poa palustris L. in Wyoming, USA.

ABSTRACT: In September 2020, several plants of fowl bluegrass, Poa palustris with seed galls were collected on a bank of river in Teton County, Wyoming, USA. Isolated nematodes were identified by both morphological and molecular methods as Anguina agrostis. This is a first report of A. agrostis in Wyoming and its report on fowl bluegrass.

Molecular Phylogenetic Characterization of Anguina Tritici (Steinbuch, 1799) Filipjev, 1936 (Rhabditida: Anguinidae) on Barley from Iraq.

Project description:During a nematological survey in Iraq, in the Bashika area, Ninevah province, an anguinid nematode population was isolated from galls of infected barley plants. The morphological characteristics indicated that the recovered species is identical to Anguina tritici. The barley population of A. tritici was molecularly characterized by sequencing two ribosomal regions (ITS and 18S rRNA genes), and their phylogenetic analyses revealed the newly generated sequences are in sister relation to corresponding sequences of A. tritici from wheat in the Bayesian tree, providing further evidence that the host plant can contribute to the separation of new isolates of plant parasitic nematodes.

Nematode Genome Announcement: A Draft Genome of Seed Gall Nematode, Anguina tritici.

Project description:Anguina tritici is the first plant-parasitic nematode described in literature, dating back to the year 1743. It is responsible for causing earcockle (seed gall) and tundu diseases in wheat and rye. Notably, this nematode has been observed to survive in an anhydrobiotic state for up to 32 years within wheat seed galls. These exceptional characteristics have inspired the sequencing of the A. tritici genome. In this study, we present the initial draft genome of A. tritici, obtained using the Illumina MiSeq platform with coverage of 60-fold. The genome is estimated to have a size of 164 Mb and comprises 39,965 protein-coding genes, exhibiting a GC content of 39.1%. The availability of this genome data will serve as a foundation for future functional biological investigations, particularly for genes whose functions remain unknown to this day.

Fungus-originated glucanase and monooxygenase genes in creeping bent grass (Agrostis stolonifera L.).

Project description:Recent studies have revealed presence of fungus-originated genes in genomes of cool-season grasses, suggesting occurrence of multiple ancestral gene transfer events between the two distant lineages. The current article describes identification of glucanase-like and monooxygenase-like genes from creeping bent grass, as lateral gene transfer candidates. An in silico analysis suggested presence of the glucanase-like gene in Agrostis, Deyeuxia, and Polypogon genera, but not in other species belonging to the clade 1 of the Poeae tribe. Similarly, the monooxygenase-like gene was confined to Agrostis and Deyeuxia genera. A consistent result was obtained from PCR-based screening. The glucanase-like gene was revealed to be ubiquitously expressed in young seedlings of creeping bent grass. Although expression of the monooxygenase-like gene was suggested in plant tissues, the levels were considerably lower than those of the glucanase-like gene. A phylogenetic analysis revealed close relationships of the two genes between the corresponding genes in fungal endophyte species of the Epichloë genus, suggesting that the genes originated from the Epichloë lineage.

A First Report of Anguina pacificae in Ireland.

Project description:Anguina pacificae is a significant pest of Poa annua golf course greens in northern California. This study presents the first confirmed case of an A. pacificae infestation outside of North America, where the nematode's distribution is further restricted to a relatively limited coastal region. Species confirmation was made by morphometric and molecular methods and comparisons to closely related species including the European species, Anguina agropyri. The A. pacificae population detected on an Irish golf course was monitored over a 2-yr period and the life cycle compared with Californian population dynamics. A. pacificae was assessed for the potential risk of spreading to the local agricultural sector, in addition, the biosecurity risks from A. pacificae and plant parasitic nematodes in general were reviewed for northwest Europe.

An updated checklist and key to the open-panicled species of Poa L. (Poaceae) in Peru including three new species, Poa ramoniana, Poa tayacajaensis, and Poa urubambensis.

Project description:We provide an updated checklist and key to the 30 Poa species with open panicles from Peru which includes previously circumscribed Dissanthelium and Aphanelytrum species, new taxon records, and three undescribed species. Poa compressa, Poa grisebachii, and Poa leioclada are recorded from Peru for the first time. A number of species are placed in synonymy: Poa carazensis, Poa ferreyrae and Poa tovarii are synonymized under the name Poa fibrifera; Poa adusta (tentatively) and Poa pilgeri are synonymized under Poa candamoana; Poa superata is synonymized under Poa grisebachii; and Poa paramoensis is synonymized under Poa huancavelicae. Included within this treatment are three new species, Poa ramoniana, Poa tayacajaensis and Poa urubambensis, which are described and illustrated. Poa ramoniana, found growing near lakes in high elevation Puna grasslands of Junín, is similar to a small form of Poa glaberrima, but differs in having rhizomes and growing to only 5 cm tall. Poa tayacajaensis, found from shrublands on Andean slopes of Huancavelica and Huánuco, bears similarities to Poa aequatoriensis but differs in having shorter lemmas which are pubescent between the veins, densely scabrous sheaths with smooth, glabrous throats, and shorter ligules. Poa urubambensis, a common element of the undisturbed Polylepis forest understory of the Cordillera Urubamba, Cusco, is distinct from all other members of open-panicled Poa's by having glabrous lemmas with a smooth and glabrous callus, and notably small anthers. The type material for the name Poa adusta is discussed and a lectotype is selected.

A Draft Transcriptome Announcement of Anguina tritici.

Project description:Anguina tritici, the wheat seed gall nematode, causes the 'ear-cockle' or seed gall disease of wheat (Triticum sp.), leading to an extensive decline of yield (30-70%) in underdeveloped wheat cultivating countries of the world. The nematode is known to survive in anhydrobiotic conditions for up to 32 years. Here, we present the first transcriptome assembly of A. tritici, which will be a valuable resource for understanding the genes responsible for nematode survival and above-ground plant parasitism. The final 133.2 Mb assembly consists of 105606 open reading frames (including isoforms) with the following BUSCO scores against Nematoda database: 80.3% complete (16.4% single copy and 63.9% duplicated), 2.1% fragmented, and 17.6% missing.
