Energy-saving mechanism research on beam-pumping unit driven by hydraulics.

ABSTRACT: Aiming to solve the problems of long transmission chain, large movement inertia of components and high energy consumption of pumping units, this proposes a new pumping unit with direct balance and hydraulic drive. Through mathematical modeling and simulation analysis to compare the suspension dynamic characteristics and balance characteristics of the hydraulically driven pumping unit with the conventional one. It turns out that the suspension maximum speed drop 21.14%, the maximum acceleration drops 28.88% and the root mean square torque drops 92.9% on the suspension of the hydraulically driven pumping unit. The experimental results proves that the hydraulically driven pumping unit has significant energy saving efficiency, and achieves more than 30.9% of active power saving rate. Theoretical and practical research results show that hydraulically driven pumping unit is reliable and better energy saving, which provides a basis in theory and engineering practice in application.

Saving energy in turbulent flows with unsteady pumping.

Project description:Viscous dissipation causes significant energy losses in fluid flows; in ducts, laminar flows provide the minimum resistance to the motion, whereas turbulence substantially increases the friction at the wall and the consequent energy requirements for pumping. Great effort is currently being devoted to find new strategies to reduce the energy losses induced by turbulence. Here we propose a simple and novel drag-reduction technique which achieves substantial energy savings in internal flows. Our approach consists in driving the flow with a temporally intermittent pumping, unlike the common practice of a constant pumping. We alternate "pump on" phases where the flow accelerates, and "pump off" phases where the flow decays freely. The flow cyclically enters a quasi-laminar state during the acceleration, and transitions to a more classic turbulent state during the deceleration. Our numerical results demonstrate that important energy savings can be achieved by simply modulating the power injection into the system over time. The physical understanding of this process can help the industry in reducing the waste of energy, creating economical benefits and preserving the environment by reducing harmful emissions.

Molecular mechanism of light-driven sodium pumping.

Project description:The light-driven sodium-pumping rhodopsin KR2 from Krokinobacter eikastus is the only non-proton cation active transporter with demonstrated potential for optogenetics. However, the existing structural data on KR2 correspond exclusively to its ground state, and show no sodium inside the protein, which hampers the understanding of sodium-pumping mechanism. Here we present crystal structure of the O-intermediate of the physiologically relevant pentameric form of KR2 at the resolution of 2.1 Å, revealing a sodium ion near the retinal Schiff base, coordinated by N112 and D116 of the characteristic NDQ triad. We also obtained crystal structures of D116N and H30A variants, conducted metadynamics simulations and measured pumping activities of putative pathway mutants to demonstrate that sodium release likely proceeds alongside Q78 towards the structural sodium ion bound between KR2 protomers. Our findings highlight the importance of pentameric assembly for sodium pump function, and may be used for rational engineering of enhanced optogenetic tools.

Light-driven anion-pumping rhodopsin with unique cytoplasmic anion-release mechanism.

Project description:Microbial rhodopsins are photoreceptive membrane proteins found in microorganisms with an all-trans-retinal chromophore. The function of many microbial rhodopsins is determined by three residues in the third transmembrane helix called motif residues. Here, we report a group of microbial rhodopsins with a novel Thr-Thr-Gly (TTG) motif. The ion-transport assay revealed that they function as light-driven inward anion pumps similar to halorhodopsins previously found in archaea and bacteria. Based on the characteristic glycine residue in their motif and light-driven anion-pumping function, these new rhodopsins are called glycylhalorhodopsins (GHRs). X-ray crystallographic analysis found large cavities on the cytoplasmic side, which are produced by the small side-chain volume of the glycine residue in the motif. The opened structure of GHR on the cytoplasmic side is related to the anion releasing process to the cytoplasm during the photoreaction compared to canonical halorhodopsin from Natronomonas pharaonis (NpHR). GHR also transports SO42- and the extracellular glutamate residue plays an essential role in extracellular SO42- uptake. In summary, we have identified TTG motif-containing microbial rhodopsins that display an anion-releasing mechanism.

How do energy price, energy-saving policies, and crisis affect energy-saving behavior?

Project description:In order to reduce CO2 emissions into the environment, energy conservation is considered a long-term strategy. To encourage people to save energy, the government may need to implement certain policies. Vietnam is a developing country dependent on fossil fuels and heavily affected by climate change. At the same time, there has been no research evaluating the impact of energy prices, government policies, and crises on energy-saving behavior in Vietnam. Therefore, this study uses the theory of planned behavior, technological acceptance behavior, and social exchange to evaluate these relationships. This research will answer the keys research question such as how would the policies which related to price, energy-saving activities effect on energy-saving behavior of households; How would a crisis like covid-19 affect energy-saving behavior? The research conducted surveys and analysis on 1166 respondents. The results of the PLS-SEM model analysis indicate that energy-saving policies do not directly affect the intention to save energy but have a positive impact on energy-saving behavior. Both energy prices and crises have a positive impact on both the intention and behavior of energy-saving. The study has made a significant contribution to the theory by confirming the existence of social exchange theory and the protection motivation theory in explaining energy-saving behavior.

The authors collected data on state policies and energy prices concerning energy-saving behavior during crises, with a particular focus on the Covid-19 crisis. The data was gathered from 1216 respondents, who are households. The data's reliability was assessed using Smart-PLS software. The data will provide research ideas related to state policies, energy prices, and energy-saving behavior associated with crises similar to Covid-19.

Research on energy-saving control of agricultural hybrid tractors integrating working condition prediction.

Project description:To address the issues of tractors using too much fuel and not being energy efficient, a predictive control strategy based on Pontryagin's minimum principle integrating working condition prediction is proposed for agricultural hybrid tractors. The Dongfanghong 1804 tractor is being used for research. Firstly, the main parameters of the hybrid drive system are determined and modeled. Secondly, based on the adaptive cubic exponential forecasting method, the working condition information for a period of time in the future is predicted through historical working condition information. Furthermore, combining the predicted working conditions information, the goal is to minimize the total energy consumption cost of the entire machine. Motor power and diesel engine power are control variables. The battery state of charge is a state variable. Subsequently, a predictive control strategy based on Pontryagin's minimum principle integrating working condition prediction is proposed. Finally, the simulation test is carried out based on the MATLAB simulation platform. Research indicates: under plowing conditions, compared with the power following control strategy, the proposed predictive control strategy can effectively manage the performance of the diesel engine and motor, ensuring they operate at their most efficient level. The total energy consumption costs of the power following control and predictive control strategies are 37.17 China Yuan (CNY) and 33.67 CNY, respectively. The cost of energy used is decreased by 9. 42%, which helps make tractor field plowing more efficient and economical.

Increased cardiac myosin super-relaxation as an energy saving mechanism in hibernating grizzly bears.

Project description:AimThe aim of the present study was to define whether cardiac myosin contributes to energy conservation in the heart of hibernating mammals.MethodsThin cardiac strips were isolated from the left ventricles of active and hibernating grizzly bears; and subjected to loaded Mant-ATP chase assays, X-ray diffraction and proteomics.Main findingsHibernating grizzly bears displayed an unusually high proportion of ATP-conserving super-relaxed cardiac myosin molecules that are likely due to altered levels of phosphorylation and rod region stability.ConclusionsCardiac myosin depresses the heart's energetic demand during hibernation by modulating its function.

Pumping mechanism of NM-R3, a light-driven bacterial chloride importer in the rhodopsin family.

Project description:A newly identified microbial rhodopsin, NM-R3, from the marine flavobacterium Nonlabens marinus, was recently shown to drive chloride ion uptake, extending our understanding of the diversity of mechanisms for biological energy conversion. To clarify the mechanism underlying its function, we characterized the crystal structures of NM-R3 in both the dark state and early intermediate photoexcited states produced by laser pulses of different intensities and temperatures. The displacement of chloride ions at five different locations in the model reflected the detailed anion-conduction pathway, and the activity-related key residues-Cys105, Ser60, Gln224, and Phe90-were identified by mutation assays and spectroscopy. Comparisons with other proteins, including a closely related outward sodium ion pump, revealed key motifs and provided structural insights into light-driven ion transport across membranes by the NQ subfamily of rhodopsins. Unexpectedly, the response of the retinal in NM-R3 to photostimulation appears to be substantially different from that seen in bacteriorhodopsin.

Low-Energy Laser-Driven Ultrashort Pulsed Electron Beam Irradiation-Induced Immune Response in Rats.

Project description:The development of new laser-driven electron linear accelerators, providing unique ultrashort pulsed electron beams (UPEBs) with low repetition rates, opens new opportunities for radiotherapy and new fronts for radiobiological research in general. Considering the growing interest in the application of UPEBs in radiation biology and medicine, the aim of this study was to reveal the changes in immune system in response to low-energy laser-driven UPEB whole-body irradiation in rodents. Forty male albino Wistar rats were exposed to laser-driven UPEB irradiation, after which different immunological parameters were studied on the 1st, 3rd, 7th, 14th, and 28th day after irradiation. According to the results, this type of irradiation induces alterations in the rat immune system, particularly by increasing the production of pro- and anti-inflammatory cytokines and elevating the DNA damage rate. Moreover, such an immune response reaches its maximal levels on the third day after laser-driven UPEB whole-body irradiation, showing partial recovery on subsequent days with a total recovery on the 28th day. The results of this study provide valuable insight into the effect of laser-driven UPEB whole-body irradiation on the immune system of the animals and support further animal experiments on the role of this novel type of irradiation.

Dogs lap using acceleration-driven open pumping.

Project description:Dogs lap because they have incomplete cheeks and cannot suck. When lapping, a dog's tongue pulls a liquid column from the bath, suggesting that the hydrodynamics of column formation are critical to understanding how dogs drink. We measured lapping in 19 dogs and used the results to generate a physical model of the tongue's interaction with the air-fluid interface. These experiments help to explain how dogs exploit the fluid dynamics of the generated column. The results demonstrate that effects of acceleration govern lapping frequency, which suggests that dogs curl the tongue to create a larger liquid column. Comparing lapping in dogs and cats reveals that, despite similar morphology, these carnivores lap in different physical regimes: an unsteady inertial regime for dogs and steady inertial regime for cats.
