The complete mitochondrial genome of Eclipophleps carinata (Orthoptera: Acridoidea: Gomphoceridae).

ABSTRACT: The complete Eclipophleps carinata mitogenome was sequenced through Illumina HiSeq 2500 platform and the resulting data were analyzed in this paper. The mitochondrial genome of E. carinata is a typical circular DNA molecule of 15,781 bp with 37 genes and 74.5% A + T content, which encoded 13 protein-coding genes (PCGs), 22 tRNA genes, two rRNA genes, and the control region. The E. carinata mitochondrial genome and 27 mitochondrial genomes (downloaded from NCBI) were employed to construct phylogenetic tree, in which Ruspolia dubia and Teleogryllus occipitails were the outgroups. Phylogenetic reconstruction validated the taxonomic status of E. carinata, which was placed in the monophyletic Gomphocerinae in Acrididae.

Project description:Calliptamus abbreviatus Ikovnnikov (Orthoptera: Acridoidea) is one of the important pests in the grasslands in northern China. The complete mitochondrial genome of this insect was sequenced. This genome is 16,615 bp long, with an AT content of 73.3%, containing 37 typical animal mitochondrial genes and an AT-rich region. All 13 PCGs share the start codon ATN, and the usual termination codons (TAA) are found from 13 protein-coding genes. All of the 22 typical animal tRNA genes were found in C. abbreviatus mt-genome, and most of the tRNAs could be folded into the classic cloverleaf secondary structure except for tRNA-Ser (AGN), which lacks the dihydrouracil (DHU) stem. The sizes of the large and small ribosomal RNA genes are 1555 and 799 bp long, respectively. The AT content of the AT-rich region is 87.0%. Phylogenetic analysis supports that the coleopteran insects from the same family cluster in the same group.

The completely assembled mitochondrial genome is 15,483 bp long. The genome follows the typical invertebrate mitochondrial gene arrangement, and includes 13 protein-coding genes, 22 tRNA genes, two rRNA genes and one A + T-rich region. Two tandem repeats were also identified within the A + T-rich region.

Complete Mitochondrial Genome Sequence of Acrida cinerea (Acrididae: Orthoptera) and Comparative Analysis of Mitochondrial Genomes in Orthoptera.

Project description:The complete 15,599-bp mitogenome of Acrida cinerea was determined and compared with that of the other 20 orthopterans. It displays characteristic gene content, genome organization, nucleotide composition, and codon usage found in other Caelifera mitogenomes. Comparison of 21 orthopteran sequences revealed that the tRNAs encoded by the H-strand appear more conserved than those by the L-stand. All tRNAs form the typical clover-leaf structure except trnS (agn), and most of the size variation among tRNAs stemmed from the length variation in the arm and loop of TΨC and the loop of DHU. The derived secondary structure models of the rrnS and rrnL from 21 orthoptera species closely resemble those from other insects on CRW except a considerably enlarged loop of helix 1399 of rrnS in Caelifera, which is a potentially autapomorphy of Caelifera. In the A+T-rich region, tandem repeats are not only conserved in the closely related mitogenome but also share some conserved motifs in the same subfamily. A stem-loop structure, 16 bp or longer, is likely to be involved in replication initiation in Caelifera and Grylloidea. A long T-stretch (>17 bp) with conserved stem-loop structure next to rrnS on the H-strand, bounded by a purine at either end, exists in the three species from Tettigoniidae.

Project description:In this paper, we determined the complete mitochondrial genome of Chinese Daphnia carinata for the first time by the long and accurate polymerase chain reaction and primer-walking methods. It was 15,245 bp in length, with an A + T content of 70.35%, containing 37 typical animal mitochondrial genes and an A + T-rich region. The COI gene started with ACTA. All the 22 typical tRNA genes had a classical cloverleaf structure except for trnS1, in which the D-stem pairings in the DHU arm were absent.

The complete mitochondrial genome (mitogenome) of Fruhstorferiola tonkinensis is 15,637 bp in length, including 13 protein-coding genes (PCGs), 22 transfer RNA (tRNA) genes, 2 ribosomal RNA genes and 1 A + T-rich region. The gene order and arrangement are identical to other Acridoidae species. Most PCGs (except for the ATP6) start with the typical ATN codons, while most stop codons are TAA, TAG. The combined dataset of 13 PCGs and 2 rRNAs from 17 grasshoppers (including 15 Catantopidae species and 2 outgroups) is used to construct phylogenetic tree and analyse phylogenetic relationship of Catantopidae.

Project description:The complete mitochondrial genome of Fruhstorferiola huayinensis is 16 227 bp in length, and consisted of 13 protein-coding genes (PCGs), 22 tRNA genes, 2 rRNA genes and a control region. In control region, a tRNAIle-like sequence was identified. Additionally, a 259 bp long tandem repeat was identified in the control region. Using the 13 PCGs and 2 rRNA of F. huayinensis, together with 14 other close-related and 2 outgroup species, we constructed BI phylogenetic tree to verify the accuracy and utility of new determined mitogenome sequences.

Using the 13 PCGs and two rRNA of P. infumata, together with 10 other close-related and two outgroup species, we constructed Bayesian inference (BI) phylogenetic tree to validate the mitogenome sequences of P. infumata.

An annotated checklist of grasshoppers (Orthoptera, Acridoidea) from Mongolia.

Project description:BackgroundGrasshoppers (Acridoidea, Orthoptera) are the dominant herbivores in grassland ecosystems worldwide. They can increase rangeland productivity by stimulating plant growth and accelerating nutrient cycling. This article presents a comprehensive checklist of grasshoppers in Mongolia. Until then, the available information was very scattered, based on old studies of Mongolian grasshoppers, recorded in a few international catalogues and databases, individual records and research work on agroecosystem communities. However, the available information on the composition of the Orthopteran fauna in Mongolia was sometimes unclear or non-existent and these dubious data were excluded from the present study. In addition, the grasshopper distribution analysis used the standardised personal collection of D. Altanchimeg. We also present a list of grasshoppers, as well as their distribution and abundance, in countries adjacent to Mongolia, such as Russia, China and South Korea. The surveys covered six types of natural zones: high mountain, taiga, forest-steppe, steppe, desert steppe and desert; desert steppe and steppe zones are the most widely distributed. We hope to have contributed significantly to the study of the distribution of grasshopper species in all these natural zones.New informationIn this study, a total of three families of Acridoidea belonging to eight subfamilies, 17 tribes, 52 genera and 128 species are reported for the various natural zones. The recorded species belong to eight subfamilies: Gomphocerinae are the most numerous with 56 species recorded, followed by Oedipodinae (51 species), Thrinchinae (nine species), Melanoplinae (six species), Calliptaminae (three species), Dericorythinae, Acridinae, Egnatiinae (one species each).

The complete mitochondrial genome of Ruspolia yunnana (Orthoptera: Tettigoniidae: Conocephalinae).

Project description:The complete mitochondrial genome of Ruspolia yunnana was 15,794 bp and consisted of 37 genes and a control region. The A + T content of the mitogenome was as high as 71.3%. Most protein-coding genes started with the codon ATN and ended with stop codons TAA. Phylogenetic analyses based on mitogenomes suggested that Ruspolia yunnana and Ruspolia dubia have the closest genetic relationship. In general, this study provided meaningful genetic information for Ruspolia yunnana and validated the phylogenetic relationships within the Conocephalinae.

The complete mitochondrial genome of H. tonkinensis is 15,625 bp in length and consists of 13 protein-coding genes, 22 tRNA genes, 2 rRNA genes and 1 A + T-rich region. The gene order of the mitogenome is identical with most orthopteran insects. Most protein-coding genes start with typical ATN codon except for COX1, which initiates with ACC codon instead. While all PCGs use complete stop codons (TAA and TAG). In addition, 13 related species and 2 outgroup taxa were used to construct the phylogenetic tree to further validate the mitogenome of H. tonkinensis. The result showed that H. tonkinensis is sister group to a clade of Oxya and Pseudoxya.
