Today's computing challenges: opportunities for computer hardware design.

ABSTRACT: Due to the explosive increase of digital data creation, demand on advancement of computing capability is ever increasing. However, the legacy approaches that we have used for continuous improvement of three elements of computer (process, memory, and interconnect) have started facing their limits, and therefore are not as effective as they used to be and are also expected to reach the end in the near future. Evidently, it is a large challenge for computer hardware industry. However, at the same time it also provides great opportunities for the hardware design industry to develop novel technologies and to take leadership away from incumbents. This paper reviews the technical challenges that today's computing systems are facing and introduces potential directions for continuous advancement of computing capability, and discusses where computer hardware designers find good opportunities to contribute.

Emerging opportunities and challenges for the future of reservoir computing.

Project description:Reservoir computing originates in the early 2000s, the core idea being to utilize dynamical systems as reservoirs (nonlinear generalizations of standard bases) to adaptively learn spatiotemporal features and hidden patterns in complex time series. Shown to have the potential of achieving higher-precision prediction in chaotic systems, those pioneering works led to a great amount of interest and follow-ups in the community of nonlinear dynamics and complex systems. To unlock the full capabilities of reservoir computing towards a fast, lightweight, and significantly more interpretable learning framework for temporal dynamical systems, substantially more research is needed. This Perspective intends to elucidate the parallel progress of mathematical theory, algorithm design and experimental realizations of reservoir computing, and identify emerging opportunities as well as existing challenges for large-scale industrial adoption of reservoir computing, together with a few ideas and viewpoints on how some of those challenges might be resolved with joint efforts by academic and industrial researchers across multiple disciplines.

Computing Components of Everyday Stress Responses: Exploring Conceptual Challenges and New Opportunities.

Project description:Repeated assessments in everyday life enables collecting ecologically valid data on dynamic, within-persons processes. These methods have widespread utility and application and have been extensively used for the study of stressors and stress responses. Enhanced conceptual sophistication of characterizing intraindividual stress responses in everyday life would help advance the field. This article provides a pragmatic overview of approaches, opportunities, and challenges when intensive ambulatory methods are applied to study everyday stress responses in "real time." We distinguish between three stress-response components (i.e., reactivity, recovery, and pileup) and focus on several fundamental questions: (a) What is the appropriate stress-free resting state (or "baseline") for an individual in everyday life? (b) How does one index the magnitude of the initial response to a stressor (reactivity)? (c) Following a stressor, how can recovery be identified (e.g., when the stress response has completed)? and (d) Because stressors may not occur in isolation, how can one capture the temporal clustering of stressors and/or stress responses (pileup)? We also present initial ideas on applying this approach to intervention research. Although we focus on stress responses, these issues may inform many other dynamic intraindividual constructs and behaviors (e.g., physical activity, physiological processes, other subjective states) captured in ambulatory assessment.

Power-saving design opportunities for wireless intracortical brain-computer interfaces.

Project description:The efficacy of wireless intracortical brain-computer interfaces (iBCIs) is limited in part by the number of recording channels, which is constrained by the power budget of the implantable system. Designing wireless iBCIs that provide the high-quality recordings of today's wired neural interfaces may lead to inadvertent over-design at the expense of power consumption and scalability. Here, we report analyses of neural signals collected from experimental iBCI measurements in rhesus macaques and from a clinical-trial participant with implanted 96-channel Utah multielectrode arrays to understand the trade-offs between signal quality and decoder performance. Moreover, we propose an efficient hardware design for clinically viable iBCIs, and suggest that the circuit design parameters of current recording iBCIs can be relaxed considerably without loss of performance. The proposed design may allow for an order-of-magnitude power savings and lead to clinically viable iBCIs with a higher channel count.

Immuno-oncology Clinical Trial Design: Limitations, Challenges, and Opportunities.

Project description:Recent advances in immuno-oncology and regulatory approvals have been rapid and paradigm shifting in many difficult-to-treat malignancies. Despite immune checkpoint inhibitor therapy becoming the standard of care across multiple tumor types, there are many unanswered questions that need to be addressed before this therapeutic modality can be fully harnessed. Areas of limitations include treatment of patients not sufficiently represented in clinical trials, uncertainty of the optimal treatment dosing and duration, and lack of understanding regarding long-term immune related toxicities and atypical tumor responses. Patients such as those with autoimmune disease, chronic viral infections, limited performance status, and brain metastases were often excluded from initial trials due to concerns of safety. However, limited data suggest that some of these patients can benefit from therapy with manageable toxicities; thus, future studies should incorporate these patients to clearly define safety and efficacy. There are still controversies regarding the optimal dosing strategy that can vary from weight-based to flat dosing, with undefined treatment duration. Further elucidation of the optimal dosing approach and evaluation of predictive biomarkers should be incorporated in the design of future trials. Finally, there are long-term immune-mediated toxicities, atypical tumor responses such as pseudoprogression and endpoints unique to immuno-oncology that are not adequately captured by traditional trial designs; thus, novel study designs are needed. In this article, we discuss in detail the above challenges and propose needed areas of research for exploration and incorporation in the next generation of immuno-oncology clinical trials.

Deep Bayesian active learning using in-memory computing hardware.

Project description:Labeling data is a time-consuming, labor-intensive and costly procedure for many artificial intelligence tasks. Deep Bayesian active learning (DBAL) boosts labeling efficiency exponentially, substantially reducing costs. However, DBAL demands high-bandwidth data transfer and probabilistic computing, posing great challenges for conventional deterministic hardware. Here we propose a memristor stochastic gradient Langevin dynamics in situ learning method that uses the stochastic of memristor modulation to learn efficiency, enabling DBAL within the computation-in-memory (CIM) framework. To prove the feasibility and effectiveness of the proposed method, we implemented in-memory DBAL on a memristor-based stochastic CIM system and successfully demonstrated a robot's skill learning task. The inherent stochastic characteristics of memristors allow a four-layer memristor Bayesian deep neural network to efficiently identify and learn from uncertain samples. Compared with cutting-edge conventional complementary metal-oxide-semiconductor-based hardware implementation, the stochastic CIM system achieves a remarkable 44% boost in speed and could conserve 153 times more energy.

Opportunities and challenges in design and optimization of protein function.

Project description:The field of protein design has made remarkable progress over the past decade. Historically, the low reliability of purely structure-based design methods limited their application, but recent strategies that combine structure-based and sequence-based calculations, as well as machine learning tools, have dramatically improved protein engineering and design. In this Review, we discuss how these methods have enabled the design of increasingly complex structures and therapeutically relevant activities. Additionally, protein optimization methods have improved the stability and activity of complex eukaryotic proteins. Thanks to their increased reliability, computational design methods have been applied to improve therapeutics and enzymes for green chemistry and have generated vaccine antigens, antivirals and drug-delivery nano-vehicles. Moreover, the high success of design methods reflects an increased understanding of basic rules that govern the relationships among protein sequence, structure and function. However, de novo design is still limited mostly to α-helix bundles, restricting its potential to generate sophisticated enzymes and diverse protein and small-molecule binders. Designing complex protein structures is a challenging but necessary next step if we are to realize our objective of generating new-to-nature activities.

Challenges and Opportunities in Adapting Clinical Trial Design for Immunotherapies.

Project description:Immunotherapy adds an exciting new dimension to the treatment of cancer, joining other approaches as a key pillar in the oncotherapeutics armamentarium. Immuno-oncology agents harbor unique mechanisms of antitumor activity by leveraging the host immune system, which may result in response patterns, resistance kinetics, and toxicity profiles that differ from other systemic therapies. These features have led to many discussions on ways to optimally integrate immunotherapy into cancer clinical trials. This overview provides an introduction to the four CCR Focus articles that ensue, with special thoughts paid to clinical trial endpoints, biomarker development and validation, combination strategies, and limitations that arise with increasing use of these agents. In addition, this overview examines design concepts that may be applied to invigorate clinical trials and to maximize their impact in the immuno-oncology era.

Ultra-fast data-mining hardware architecture based on stochastic computing.

Project description:Minimal hardware implementations able to cope with the processing of large amounts of data in reasonable times are highly desired in our information-driven society. In this work we review the application of stochastic computing to probabilistic-based pattern-recognition analysis of huge database sets. The proposed technique consists in the hardware implementation of a parallel architecture implementing a similarity search of data with respect to different pre-stored categories. We design pulse-based stochastic-logic blocks to obtain an efficient pattern recognition system. The proposed architecture speeds up the screening process of huge databases by a factor of 7 when compared to a conventional digital implementation using the same hardware area.

Molecular Energy Landscapes of Hardware-Efficient Ansatze in Quantum Computing.

Project description:Rapid advances in quantum computing have opened up new opportunities for solving the central electronic structure problem in computational chemistry. In the noisy intermediate-scale quantum (NISQ) era, where qubit coherence times are limited, it is essential to exploit quantum algorithms with sufficiently short quantum circuits to maximize qubit efficiency. The procedural construction of hardware-efficient ansätze provides one approach to design such circuits. However, refining the accuracy of the global minimum by increasing circuit depth may lead to a proliferation of local minima that hinders global optimization. To investigate this phenomenon, we explore the energy landscapes of hardware-efficient circuits to identify ground-state energies of the hydrogen, lithium hydride, and beryllium hydride molecules. We also propose a simple dimensionality reduction procedure that reduces quantum gate depth while retaining high accuracy for the global minimum, simplifying the energy landscape, and hence speeding up optimization from both software and hardware perspectives.

Integrating reconfigurable hardware-based grid for high performance computing.

Project description:FPGAs have shown several characteristics that make them very attractive for high performance computing (HPC). The impressive speed-up factors that they are able to achieve, the reduced power consumption, and the easiness and flexibility of the design process with fast iterations between consecutive versions are examples of benefits obtained with their use. However, there are still some difficulties when using reconfigurable platforms as accelerator that need to be addressed: the need of an in-depth application study to identify potential acceleration, the lack of tools for the deployment of computational problems in distributed hardware platforms, and the low portability of components, among others. This work proposes a complete grid infrastructure for distributed high performance computing based on dynamically reconfigurable FPGAs. Besides, a set of services designed to facilitate the application deployment is described. An example application and a comparison with other hardware and software implementations are shown. Experimental results show that the proposed architecture offers encouraging advantages for deployment of high performance distributed applications simplifying development process.
