Discontinuous nanoporous membranes reduce non-specific fouling for immunoaffinity cell capture.

ABSTRACT: The microfluidic isolation of target cells using adhesion-based surface capture has been widely explored for biology and medicine. However, high-throughput processing can be challenging due to interfacial limitations such as transport, reaction, and non-specific fouling. Here, it is shown that antibody-functionalized capture surfaces with discontinuous permeability enable efficient target cell capture at high flow rates by decreasing fouling. Experimental characterization and theoretical modeling reveal that "wall effects" affect cell-surface interactions and promote excess surface accumulation. These issues are partially circumvented by reducing the transport and deposition of cells near the channel walls. Optimized microfluidic devices can be operated at higher cell concentrations with significant improvements in throughput.

Depletion attractions drive bacterial capture on both non-fouling and adhesive surfaces, enhancing cell orientation.

Project description:Depletion attractions, occurring between surfaces immersed in a polymer solution, drive bacteria adhesion to a variety of surfaces. The latter include the surfaces of non-fouling coatings such as hydrated polyethylene glycol (PEG) layers but also, as demonstrated in this work, surfaces that are bacteria-adhesive, such as that of glass. Employing a flagella free E. coli strain, we demonstrate that cell adhesion on glass is enhanced by dissolved polyethylene oxide (PEO), exhibiting a faster rate and greater numbers of captured cells compared with the slower capture of the same cells on glass from a buffer solution. After removal of depletant, any cell retention appears to be governed by the substrate, with cells immediately released from non-fouling PEG surfaces but retained on glass. A distinguishing feature of cells captured by depletion on PEG surfaces is their orientation parallel to the surface and very strong alignment with flow. This suggests that, in the moments of capture, cells are able to rotate as they adhere. By contrast on glass, captured cells are substantially more upright and less aligned by flow. On glass the free polymer exerts forces that slightly tip cells towards the surface. Free polymer also holds cells still on adhesive and non-fouling surfaces alike but, upon removal of free PEO, cells retained on glass tend to be held by one end and exhibit a Brownian type rotational rocking.

Tunable molecular separation by nanoporous membranes.

Project description:Metal-organic frameworks offer tremendous potential for efficient separation of molecular mixtures. Different pore sizes and suitable functionalizations of the framework allow for an adjustment of the static selectivity. Here we report membranes which offer dynamic control of the selectivity by remote signals, thus enabling a continuous adjustment of the permeate flux. This is realized by assembling linkers containing photoresponsive azobenzene-side-groups into monolithic, crystalline membranes of metal-organic frameworks. The azobenzene moieties can be switched from the trans to the cis configuration and vice versa by irradiation with ultraviolet or visible light, resulting in a substantial modification of the membrane permeability and separation factor. The precise control of the cis:trans azobenzene ratio, for example, by controlled irradiation times or by simultaneous irradiation with ultraviolet and visible light, enables the continuous tuning of the separation. For hydrogen:carbon-dioxide, the separation factor of this smart membrane can be steplessly adjusted between 3 and 8.

Versatile ultrathin nanoporous silicon nitride membranes.

Project description:Single- and multiple-nanopore membranes are both highly interesting for biosensing and separation processes, as well as their ability to mimic biological membranes. The density of pores, their shape, and their surface chemistry are the key factors that determine membrane transport and separation capabilities. Here, we report silicon nitride (SiN) membranes with fully controlled porosity, pore geometry, and pore surface chemistry. An ultrathin freestanding SiN platform is described with conical or double-conical nanopores of diameters as small as several nanometers, prepared by the track-etching technique. This technique allows the membrane porosity to be tuned from one to billions of pores per square centimeter. We demonstrate the separation capabilities of these membranes by discrimination of dye and protein molecules based on their charge and size. This separation process is based on an electrostatic mechanism and operates in physiological electrolyte conditions. As we have also shown, the separation capabilities can be tuned by chemically modifying the pore walls. Compared with typical membranes with cylindrical pores, the conical and double-conical pores reported here allow for higher fluxes, a critical advantage in separation applications. In addition, the conical pore shape results in a shorter effective length, which gives advantages for single biomolecule detection applications such as nanopore-based DNA analysis.

Ionization behavior of nanoporous polyamide membranes.

Project description:Escalating global water scarcity necessitates high-performance desalination membranes, for which fundamental understanding of structure-property-performance relationships is required. In this study, we comprehensively assess the ionization behavior of nanoporous polyamide selective layers in state-of-the-art nanofiltration (NF) membranes. In these films, residual carboxylic acids and amines influence permeability and selectivity by imparting hydrophilicity and ionizable moieties that can exclude coions. We utilize layered interfacial polymerization to prepare physically and chemically similar selective layers of controlled thickness. We then demonstrate location-dependent ionization of carboxyl groups in NF polyamide films. Specifically, only surface carboxyl groups ionize under neutral pH, whereas interior carboxyl ionization requires pH >9. Conversely, amine ionization behaves invariably across the film. First-principles simulations reveal that the low permittivity of nanoconfined water drives the anomalous carboxyl ionization behavior. Furthermore, we report that interior carboxyl ionization could improve the water-salt permselectivity of NF membranes over fourfold, suggesting that interior charge density could be an important tool to enhance the selectivity of polyamide membranes. Our findings highlight the influence of nanoconfinement on membrane transport properties and provide enhanced fundamental understanding of ionization that could enable novel membrane design.

Discontinuous and non-discontinuous subgenomic RNA transcription in a nidovirus.

Project description:Arteri-, corona-, toro- and roniviruses are evolutionarily related positive-strand RNA viruses, united in the order Nidovirales. The best studied nidoviruses, the corona- and arteriviruses, employ a unique transcription mechanism, which involves discontinuous RNA synthesis, a process resembling similarity-assisted copy-choice RNA recombination. During infection, multiple subgenomic (sg) mRNAs are transcribed from a mirror set of sg negative-strand RNA templates. The sg mRNAs all possess a short 5' common leader sequence, derived from the 5' end of the genomic RNA. The joining of the non-contiguous 'leader' and 'body' sequences presumably occurs during minus-strand synthesis. To study whether toroviruses use a similar transcription mechanism, we characterized the 5' termini of the genome and the four sg mRNAs of Berne virus (BEV). We show that BEV mRNAs 3-5 lack a leader sequence. Surprisingly, however, RNA 2 does contain a leader, identical to the 5'-terminal 18 residues of the genome. Apparently, BEV combines discontinuous and non-discontinuous RNA synthesis to produce its sg mRNAs. Our findings have important implications for the understanding of the mechanism and evolution of nidovirus transcription.

Gas permeation through graphdiyne-based nanoporous membranes.

Project description:Nanoporous membranes based on two dimensional materials are predicted to provide highly selective gas transport in combination with extreme permeance. Here we investigate membranes made from multilayer graphdiyne, a graphene-like crystal with a larger unit cell. Despite being nearly a hundred of nanometers thick, the membranes allow fast, Knudsen-type permeation of light gases such as helium and hydrogen whereas heavy noble gases like xenon exhibit strongly suppressed flows. Using isotope and cryogenic temperature measurements, the seemingly conflicting characteristics are explained by a high density of straight-through holes (direct porosity of ∼0.1%), in which heavy atoms are adsorbed on the walls, partially blocking Knudsen flows. Our work offers important insights into intricate transport mechanisms playing a role at nanoscale.

Ultrafast ion sieving using nanoporous polymeric membranes.

Project description:The great potential of nanoporous membranes for water filtration and chemical separation has been challenged by the trade-off between selectivity and permeability. Here we report on nanoporous polymer membranes with an excellent balance between selectivity and permeability of ions. Our membranes are fabricated by irradiating 2-μm-thick polyethylene terephthalate Lumirror® films with GeV heavy ions followed by ultraviolet exposure. These membranes show a high transport rate of K+ ions of up to 14 mol h-1 m-2 and a selectivity of alkali metal ions over heavy metal ions of >500. Combining transport experiments and molecular dynamics simulations with a polymeric nanopore model, we demonstrate that the high permeability is attributable to the presence of nanopores with a radius of ~0.5 nm and a density of up to 5 × 1010 cm-2, and the selectivity is ascribed to the interaction between the partially dehydrated ions and the negatively charged nanopore wall.

Anti-fouling graphene-based membranes for effective water desalination.

Project description:The inability of membranes to handle a wide spectrum of pollutants is an important unsolved problem for water treatment. Here we demonstrate water desalination via a membrane distillation process using a graphene membrane where water permeation is enabled by nanochannels of multilayer, mismatched, partially overlapping graphene grains. Graphene films derived from renewable oil exhibit significantly superior retention of water vapour flux and salt rejection rates, and a superior antifouling capability under a mixture of saline water containing contaminants such as oils and surfactants, compared to commercial distillation membranes. Moreover, real-world applicability of our membrane is demonstrated by processing sea water from Sydney Harbour over 72 h with macroscale membrane size of 4 cm2, processing ~0.5 L per day. Numerical simulations show that the channels between the mismatched grains serve as an effective water permeation route. Our research will pave the way for large-scale graphene-based antifouling membranes for diverse water treatment applications.

Evaluating Protein Fouling on Membranes Patterned by Woven Mesh Fabrics.

Project description:Membrane surface patterning is one approach used to mitigate fouling. This study used a combination of flux decline measurements and visualization experiments to evaluate the effectiveness of a microscale herringbone pattern for reducing protein fouling on polyvinylidene fluoride (PVDF) ultrafiltration membranes. Thermal embossing with woven mesh stamps was used for the first time to pattern membranes. Embossing process parameters were studied to identify conditions replicating the mesh patterns with high fidelity and to determine their effect on membrane permeability. Permeability increased or remained constant when patterning at low pressure (≤4.4 MPa) as a result of increased effective surface area; whereas permeability decreased at higher pressures due to surface pore-sealing of the membrane active layer upon compression. Flux decline measurements with dilute protein solutions showed monotonic decreases over time, with lower rates for patterned membranes than as-received membranes. These data were analyzed by the Hermia model to follow the transient nature of fouling. Confocal laser scanning microscopy (CLSM) provided complementary, quantitative, spatiotemporal information about protein deposition on as-received and patterned membrane surfaces. CLSM provided a greater level of detail for the early (pre-monolayer) stage of fouling than could be deduced from flux decline measurements. Images show that the protein immediately started to accumulate rapidly on the membranes, likely due to favorable hydrophobic interactions between the PVDF and protein, followed by decreasing rates of fouling with time as protein accumulated on the membrane surface. The knowledge generated in this study can be used to design membranes that inhibit fouling or otherwise direct foulants to deposit selectively in regions that minimize loss of flux.
