Recognition of Hydrophilic Cyclic Compounds by a Water-Soluble Cavitand.

ABSTRACT: A water-soluble deep cavitand bearing amides on the upper rim and trimethyl ammonium groups on the feet was synthesized. The open-ended cavity is stabilized by the intramolecular hydrogen bonds formed between the adjacent amides, and the introduction of trimethylammonium imparts to the cavitand good solubility in water. The cavitand exhibits high binding affinity and selectivity to hydrophilic molecules in water. With certain guests, such as cyclohexyl alcohols, amines and acids, the recognition involves the synergistic action of hydrogen bonding with hydrophobic effects. The binding phenomena are interpreted in terms of a fixed solvent cage presented by the host to the guest.

Origin of the blueshift of water molecules at interfaces of hydrophilic cyclic compounds.

Project description:Water molecules at interfaces of materials exhibit enigmatic properties. A variety of spectroscopic studies have observed a high-frequency motion in these water molecules, represented by a blueshift, at both hydrophobic and hydrophilic interfaces. However, the molecular mechanism behind this blueshift has remained unclear. Using Raman spectroscopy and ab initio molecular dynamics simulations, we reveal the molecular mechanism of the blueshift of water molecules around six monosaccharide isomers. In the first hydration shell, we found weak hydrogen-bonded water molecules that cannot have a stable tetrahedral water network. In the water molecules, the vibrational state of the OH bond oriented toward the bulk solvent strongly contributes to the observed blueshift. Our work suggests that the blueshift in various solutions originates from the vibrational motions of these observed water molecules.

Cavitand-Decorated Silicon Columnar Nanostructures for the Surface Recognition of Volatile Nitroaromatic Compounds.

Project description:Nanocolumnar Si substrates (porous silicon (PSi)) have been functionalized with a quinoxaline-bridged (EtQxBox) cavitand in which the quinoxaline moieties are bonded to each other through four ethylendioxy bridges at the upper rim of the cavity. The receptor, which is known to selectively complex aromatic volatile organic compounds (VOCs) even in the presence of aliphatic compounds, has been covalently anchored to PSi. The larger surface area of PSi, compared to that of flat substrates, allowed one to study the recognition process of the surface-grafted receptors through different techniques: Fourier-transform infrared spectroscopy, thermal desorption, and X-ray photoelectron spectroscopy. The experiments proved that surface-grafted cavitands retain the recognition capability toward aromatic VOCs. In addition, the affinities of EtQxBox for various aromatic compounds (i.e., benzene, toluene, nitrobenzene, and p-nitrotoluene) have been studied combining density functional theory computations and thermal desorption experiments. Computational data based on the crystal structures of the complexes indicate that this cavitand possesses a higher affinity toward aromatic nitro-compounds compared to benzene and toluene, making this receptor of particular interest for the detection of explosive taggants. The results of computational studies have been validated also for the surface-grafted receptor through competitive recognition experiments. These experiments showed that EtQxBox-functionalized PSi can recognize nitrobenzene in the presence of a significant excess of aromatic vapors such as benzene (1:300) or toluene (1:100).

Exclusive recognition of sarcosine in water and urine by a cavitand-functionalized silicon surface.

Project description:A supramolecular approach for the specific detection of sarcosine, recently linked to the occurrence of aggressive prostate cancer forms, has been developed. A hybrid active surface was prepared by the covalent anchoring on Si substrates of a tetraphosphonate cavitand as supramolecular receptor and it was proven able to recognize sarcosine from its nonmethylated precursor, glycine, in water and urine. The entire complexation process has been investigated in the solid state, in solution, and at the solid-liquid interface to determine and weight all the factors responsible of the observed specificity. The final outcome is a Si-based active surface capable of binding exclusively sarcosine. The complete selectivity of the cavitand-decorated surface under these stringent conditions represents a critical step forward in the use of these materials for the specific detection of sarcosine and related metabolites in biological fluids.

Guest recognition in a partially bridged deep cavitand.

Project description:The synthesis of a new deep cavitand partially bridged via disulfide bonds is described. Its thermodynamic and kinetic complexation parameters are compared with those of an unbridged analogue. The disulfide bridges cause deviations in ΔH and ΔS but result in only small differences in ΔG of complexation. The bridges increase the activation barrier for guest dissociation and lead to complexes with enhanced kinetic stability.

Recognition and organocatalysis with a synthetic cavitand receptor.

Project description:The cyclization reaction of an epoxyalcohol is catalyzed by a synthetic cavitand receptor with an inwardly directed carboxylic acid function. The receptor features a hydrophobic pocket in which the substrate is bound and positioned to react in a regioselective manner. The nature of this substrate-catalyst complex and its dynamic properties were investigated by NMR methods and with the aid of a model compound lacking the epoxide function. The kinetic parameters of the cyclization reaction were also studied. A catalytic cycle is proposed and diverse inhibition mechanisms are identified that parallel those encounterd in enzymology.

ITC and NMR Analysis of the Encapsulation of Fatty Acids within a Water-Soluble Cavitand and its Dimeric Capsule.

Project description:We report here NMR and ITC studies of the binding of ionizable guests (carboxylate acids) to a deep-cavity cavitand. These studies reveal that the shortest guests favored 1:1 complex formation, but the longer the alkyl chain the more the 2:1 host-guest capsule is favored. For intermediate-sized guests, the equilibrium between these two states is controlled by pH; at low values the capsule containing the carboxylic acid guest is favored, whereas as the pH is raised deprotonation of the guest favors the 1:1 complex. Interestingly, for one host-guest pair the energy required to de-cap the 2:1 capsular complex and form the 1:1 complex is sufficient to shift the pKa of the guest by ~ 3-4 orders of magnitude (4.1-5.4 kcal mol-1). The two largest guests examined form stable 2:1 capsules, with in both cases the guest adopting a relatively high energy J-shaped motif. Furthermore, these 2:1 complexes are sufficiently stable that at high pH guest deprotonation occurs without de-capping of the capsule.

Ultrasound-assisted synthesis of water-soluble monosubstituted diruthenium compounds.

Project description:The elusive monosubstituted diruthenium complexes [Ru2Cl(DAniF)(O2CMe)3] (1), [Ru2Cl(DPhF)(O2CMe)3] (2), [Ru2Cl(D-p-CNPhF)(O2CMe)3] (3), [Ru2Cl(D-o-TolF)(O2CMe)3] (4), [Ru2Cl(D-m-TolF)(O2CMe)3] (5), [Ru2Cl(D-p-TolF)(O2CMe)3] (6) and [Ru2Cl(p-TolA)(O2CMe)3] (7) have been synthesized using for the first time ultrasound-assisted synthesis to carry out a substitution reaction in metal-metal bonded dinuclear compounds (DAniF- = N,N'-bis(4-anisyl)formamidinate; DPhF- = N,N'-diphenylformamidinate; D-p-CNPhF- = N,N'-bis(4-cyanophenyl)formamidinate; D-o/m/p-TolF- = N,N'-bis(2/3/4-tolyl)formamidinate; p-TolA- = N-4-tolylamidate). This is a simpler and greener method than the tedious procedures described in the literature, and it has permitted to obtain water-soluble complexes with good yields in a short period of time. A synthetic study has been implemented to find the best experimental conditions to prepare compounds 1-7. Two different types of ligands, formamidinate and amidate, have been used to check the generality of the method for the preparation of monosubstituted complexes. Five new compounds (2-6) have been obtained using a formamidinate ligand, the synthesis of the previously described compound 1 has been improved, and an unprecedented monoamidate complex has been achieved (7). The crystal structures of compounds 3 and 7 have been solved by single crystal X-ray diffraction. These compounds show the typical paddlewheel structure with three acetate ligands and one formamidinate (3) or amidate (7) bridging ligand at the equatorial positions. The axial positions are occupied by the chloride ligand giving rise to one-dimensional polymer structures that were previously unknown for monosubstituted compounds.

Enantioselective Molecular Recognition in a Flexible Self-Folding Cavitand.

Project description:We report a chiral deep cavitand receptor based on calix[5]arene stabilized by a cooperative network of hydrogen bonds and having a highly flexible structure. The cavitand displays enantioselective molecular recognition with a series of chiral quaternary ammonium salts, providing unprecedented stability ratios between the corresponding diastereomeric host-guest complexes. Molecular dynamics simulations corroborate the higher flexibility of the new host and the emergence of superior induced-fit behavior with regards to resorcin[4]arene derived self-folding cavitands.

Water soluble macrocyclic host for recognition of <i>N</i>-methylquinolinium salts in water.

Project description:In this paper, we reported the synthesis of water soluble macrocyclic arenes 1 containing anionic carboxylate groups. It was found that host 1 could form a 1 : 1 complex with N-methylquinolinium salts in water. Moreover, the complexation and decomplexation of the complexes between host and the guests could be achieved by changing the pH of the solution, and the process could also be observed by naked eye.

A Newly Designed Water Soluble Uranyl-Salophen Complex for Anion Recognition.

Project description:A novel water-soluble uranyl-salophen (salophen=N,N'-disalicylidene-o-phenylenediaminate) complex was obtained. Solubility was achieved in aqueous methyl-β-cyclodextrin solutions, taking advantage of the host-guest interactions established with the adamantyl moieties present on the ligand skeleton. Such an approach facilitates the synthesis of the receptor and the purification processes and, in perspective, can be definitely applicable to other molecular scaffolds. UV/Vis titration experiments demonstrate that the capacity of the uranyl-salophen core to behave as a receptor for anions is retained in water and appears comparable with that previously reported for other water-soluble uranyl-salophen systems. Hence the presence of cyclodextrins does not interfere with molecular recognition processes.
