Single-Atom Switching as a General Approach to Designing Colorimetric and Fluorogenic Probes for Mercury Ions.

ABSTRACT: By performing a single-atom replacement within common fluorophores, we have developed a facile and general strategy to prepare a broad-spectrum class of colorimetric and fluorogenic probes for the selective detection of mercury ions in aqueous environments. Thionation of carbonyl groups from existing fluorophore cores results in a great reduction of fluorescence quantum yield and loss of fluorescence emission. The resulting thiocaged probes are efficiently desulfurized to their oxo derivatives in the presence of mercury ions, leading to pronounced changes in chromogenic and fluorogenic signals. Because these probes exhibit high selectivity, excellent sensitivity, good membrane-permeability, and rapid responses towards mercury ions, they are suitable for visualization of mercury in both aqueous and intracellular environments.

Gold nanozyme-based paper chip for colorimetric detection of mercury ions.

Project description:In this study, we developed a facile gold nanozyme-based paper chip (AuNZ-PAD) for Hg2+ detection. This device has the advantages of being simple, rapid, cost effective, sensitive, selective, high throughput, and applicable to onsite detection. The colorimetric mercury assay on the AuNZ-PAD is established based on the enzyme-like catalytic activity of gold nanoparticles promoted by the formation of Au-Hg amalgam, which is correlated to the intensity of the colorimetric response resulting from the catalytic reaction of 3,3',5,5'-tetramethylbenzidine (TMB) and H2O2. Highly sensitive and selective detection of Hg2+ ions is achieved in both distilled and tap water samples, indicating the feasibility and applicability of our device for the determination of mercury pollution in real samples. Moreover, AuNZ-PAD analysis using a smartphone camera eliminates the need for expensive analytical equipment, thereby increasing the practicality of field monitoring of trace Hg2+ compared with other sensing methods.

Development and applications of fluorogenic probes for mercury(II) based on vinyl ether oxymercuration.

Project description:Mercury is a major threat to the environment and to human health. It is highly desirable to develop a user-friendly kit for on-site mercury detection. Such a method must be able to detect mercury below the threshold levels for drinking water, 1-2 ppb. We developed a fluorescence method based on the oxymercuration of vinyl ethers to detect mercury in dental and environmental samples. Chloride ions interfered with the oxymercuration reaction, but the addition of AgNO(3) solved this problem. Fine electronic and structural tuning led to the development of a more responsive probe that was less sensitive to chloride ion interference. This second-generation probe could detect 1 ppb mercury ions in water.

DNA-Gold Nanozyme-Modified Paper Device for Enhanced Colorimetric Detection of Mercury Ions.

Project description:In this work, a paper device consisted of a patterned paper chip, wicking pads, and a base was fabricated. On the paper chip, DNA-gold nanoparticles (DNA-AuNPs) were deposited and Hg2+ ions could be adsorbed by the DNA-AuNPs. The formed DNA-AuNP/Hg2+ nanozyme could catalyze the tetramethylbenzidine (TMB)-H2O2 chromogenic reaction. Due to the wicking pads, a larger volume of Hg2+ sample could be applied to the paper device for Hg2+ detection and therefore the color response could be enhanced. The paper device achieved a cut-off value of 50 nM by the naked eye for Hg2+ under optimized conditions. Moreover, quantitative measurements could be implemented by using a desktop scanner and extracting grayscale values. A linear range of 50-2000 nM Hg2+ was obtained with a detection limit of 10 nM. In addition, the paper device could be applied in the detection of environmental water samples with high recoveries ranging from 85.7% to 105.6%. The paper-device-based colorimetric detection was low-cost, simple, and demonstrated high potential in real-sample applications.

Simultaneous detection of mercury and cadmium ions: A colorimetric method in aqueous media.

Project description:Hg and Cd are the two most toxic heavy metal ions that could be found in aqueous solutions. In this study, a chemosensor based on 5-(4-((4-nitrophenyl) diazenyl) phenyl)-1,3,4-oxadiazole-2-thiol (DOT) was reported to detect these ions simultaneously. DOT showed high selectivity towards Hg ion by changing the color of the solution from beige to gold-yellow at different concentrations of Hg ion. In comparison, other relevant metals, such as Li+, Na+, K+, Cs+, Mg2+, Ca2+, Al3+, Fe2+, Ag+, Cu2+, Pb2+, Ni2+, Zn2+, Cr3+, Fe3+, Pb4+, Mn2+, and Cd2+ did not affect the color of the DOT solution as the interfering ions. Despite no changes in the color of DOT solution in the presence of Cd ion, a solution containing DOT-Hg complex was changed from gold-yellow to orange by adding Cd ion, providing an approach for detecting Hg and Cd ion simultaneously with UV-Vis and Fluorescent spectroscopy. DOT exhibited a high association constant with a detection limit of 0.05 μM for Hg and Cd ions in an aqueous solution. The results of quantum mechanics (QM) calculations were also consistent with the experimental observations, which indicated that changes in the band gap could explain the various colors of DOT complex with metal ions.

Fluorescein Hydrazide-Appended Metal-Organic Framework as a Chromogenic and Fluorogenic Chemosensor for Mercury Ions.

Project description:In this work, we prepared a fluorescein hydrazide-appended Ni(MOF) (Metal-Organic Framework) [Ni3(BTC)2(H2O)3]·(DMF)3(H2O)3 composite, FH@Ni(MOF). This composite was well-characterized by PXRD (powder X-ray diffraction), FT-IR (Fourier transform infrared spectroscopy), N2 adsorption isotherm, TGA (thermogravimetric analysis), XPS (X-ray photoelectron spectroscopy), and FESEM (field emission scanning electron microscopy). This composite was then tested with different heavy metals and was found to act as a highly selective and sensitive optical sensor for the Hg2+ ion. It was found that the aqueous emulsion of this composite produces a new peak in absorption at 583 nm, with a chromogenic change to a pink color visible to the naked eye upon binding with Hg2+ ions. In emission, it enhances fluorescence with a fluorogenic change to green fluorescence upon complexation with the Hg2+ ion. The binding constant was found to be 9.4 × 105 M-1, with a detection limit of 0.02 μM or 5 ppb. This sensor was also found to be reversible and could be used for seven consecutive cycles. It was also tested for Hg2+ ion detection in practical water samples from ground water, tap water, and drinking water.

Fluorescence Sensing of Zinc and Mercury Ions with Hydrophilic 1,2,3-Triazolyl Fluorene Probes.

Project description:The ability to rapidly detect biologically and environmentally significant metal ions such as zinc and mercury is important to study a number of important cellular and environmental processes. Hydrophilic bis(1,2,3-triazolyl)fluorene-based derivatives, containing a 1,2,3-triazole-based recognition moiety, were synthesized through Click chemistry and characterized by UV-vis absorption, fluorescence emission, and two-photon absorption as new fluorescence sensing probes, selective for Zn(2+) and Hg(2+) ions. The UV-vis absorption and fluorescence emission spectra of the complexes exhibited blue-shifted absorption and emission spectra upon chelation to Zn(2+) and Hg(2+) ions, resulting in ca. two-fold enhancement in fluorescence. Fluorometric titration revealed that 1:2 and 1:3 ligand to metal complexes formed with binding constants of 10(8) and 10(16) for Zn(2+) and Hg(2+), respectively. The two-photon absorption cross sections for the probes and probe-metal ion complexes ranged from 200-350 GM at 800 nm. These novel fluorescent compounds may have potential as new metal ion sensors to probe cellular and biological environments.

Designing neighboring-site activation of single atom via tunnel ions for boosting acidic oxygen evolution.

Project description:Realizing an efficient turnover frequency in the acidic oxygen evolution reaction by modifying the reaction configuration is crucial in designing high-performance single-atom catalysts. Here, we report a "single atom-double site" concept, which involves an activatable inert manganese atom redox chemistry in a single-atom Ru-Mn dual-site platform with tunnel Ni ions as the trigger. In contrast to conventional single-atom catalysts, the proposed configuration allows direct intramolecular oxygen coupling driven by the Ni ions intercalation effect, bypassing the secondary deprotonation step instead of the kinetically sluggish adsorbate evolution mechanism. The strong bonding of Ni ions activates the inert manganese terminal groups and inhibits the cross-site disproportionation process inherent in the Mn scaffolding, which is crucial to ensure the dual-site platform. As a result, the single-atom Ru-Ni-Mn octahedral molecular sieves catalyst delivers a low overpotential, adequate mass activity and good stability.

Label-free colorimetric detection of mercury via Hg(2+) ions-accelerated structural transformation of nanoscale metal-oxo clusters.

Project description:Mercury and its compounds are known to be extremely toxic but widely distributed in environment. Although many works have been reported to efficiently detect mercury, development of simple and convenient sensors is still longed for quick analyzing mercury in water. In this work, a nanoscale metal-oxo cluster, (n-Bu4N)2[Mo5NaO13(OCH3)4(NO)], (MLPOM), organically-derivatized from monolacunary Lindqvist-type polyoxomolybdate, is found to specifically react with Hg(2+) in methanol/water via structural transformation. The MLPOM methanol solution displays a color change from purple to brown within seconds after being mixed with an aqueous solution containing Hg(2+). By comparing the structure of polyoxomolybdate before and after reaction, the color change is revealed to be the essentially structural transformation of MLPOM accelerated by Hg(2+). Based on this discovery, MLPOM could be utilized as a colorimetric sensor to sense the existence of Hg(2+), and a simple and label-free method is developed to selectively detect aqueous Hg(2+). Furthermore, the colorimetric sensor has been applied to indicating mercury contamination in industrial sewage.

Protein encapsulation: a new approach for improving the capability of small-molecule fluorogenic probes.

Project description:Herein, we report a protein-based hybridization strategy that exploits the host-guest chemistry of HSA (human serum albumin) to solubilize the otherwise cell impermeable ONOO- fluorescent probe Pinkment-OAc. Formation of a HSA/Pinkment-OAc supramolecular hybrid was confirmed by SAXS and solution-state analyses. This HSA/Pinkment-OAc hybrid provided an enhanced fluorescence response towards ONOO- versus Pinkment-OAc alone, as determined by in vitro experiments. The HSA/Pinkment-OAc hybrid was also evaluated in RAW 264.7 macrophages and HeLa cancer cell lines, which displayed an enhanced cell permeability enabling the detection of SIN-1 and LPS generated ONOO- and the in vivo imaging of acute inflammation in LPS-treated mice. A remarkable 5.6 fold (RAW 264.7), 8.7-fold (HeLa) and 2.7-fold increased response was seen relative to Pinkment-OAc alone at the cellular level and in vivo, respectively. We anticipate that HSA/fluorescent probe hybrids will soon become ubiquitous and routinely applied to overcome solubility issues associated with hydrophobic fluorescent imaging agents designed to detect disease related biomarkers.

A general screening strategy for peptide-based fluorogenic ligands: probes for dynamic studies of PDZ domain-mediated interactions.

Project description:A systematic and general approach for identifying efficient probes for class I PDZ domains based on environment-sensitive chromophores is presented. A series of peptides derived from the C-terminal sequence of Stargazin was first used with PDZ domains of PSD-95 and Shank3 to identify the optimal position and linker length for the 4-DMAP chromophore. The results were applied to well-characterized ligand sequences for each set of domains to generate high affinity probes that retain their native sequence specificity and yield remarkable fluorescence increases upon binding. These probes constitute efficient tools to study the dynamics and regulatory mechanisms of PDZ domain-mediated interactions.
