Chemically Fueled Volume Phase Transition of Polyacid Microgels.

ABSTRACT: Microgels are soft colloids that show responsive behavior and are easy to functionalize for applications. They are considered key components for future smart colloidal material systems. However, so far microgel systems have almost exclusively been studied in classical responsive switching settings using external triggers, while internally organized, autonomous control mechanisms as found in supramolecular chemistry and DNA nanotechnology relying on fuel-driven out-of-equilibrium concepts have not been implemented into microgel systems. Here, we introduce chemically fueled transient volume phase transitions (VPTs) for poly(methacrylic acid) (PMAA) microgels, where the collapsed hydrophobic state can be programmed using the fuel concentration in a cyclic reaction network. We discuss details of the system behavior as a function of pH and fuel amount, unravel kinetically trapped regions and showcase transient encapsulation and time-programmed release as a first application.

Deuteration-Induced Volume Phase Transition Temperature Shift of PNIPMAM Microgels.

Project description:The effect of deuteration on the volume phase transition (VPT) temperature of poly (N-isopropylmethacrylamide) (pNIPMAM) microgels in aqueous suspension is determined via IR spectroscopy and size measurements by photon correlation spectroscopy (PCS). We study the effect of a hydrogenated and a deuterated solvent (H₂O/D₂O), and of the hydrogenated and (partially) deuterated monomer. Deuteration of the monomer or copolymerization with deuterated monomers shifts the volume phase transition temperature (VPTT) by up to 8.4 K to higher temperatures, in good agreement with known results for pNIPAM microgels. Moreover, the shape of the swelling curve is found to depend on deuteration, with the highest deuteration leading to the sharpest VPT. Finally, the quantitative agreement between FTIR spectroscopy and PCS evidences the spatial homogeneity of the microgel particles. Our results are rationalized in terms of the effect of deuteration on hydrogen bonding. They shall be of primary importance for any experimental measurements close to the VPT involving isotopic substitution, and in particular contrast variation small angle neutron scattering.

Two-step deswelling in the Volume Phase Transition of thermoresponsive microgels.

Project description:Thermoresponsive microgels are one of the most investigated types of soft colloids, thanks to their ability to undergo a Volume Phase Transition (VPT) close to ambient temperature. However, this fundamental phenomenon still lacks a detailed microscopic understanding, particularly regarding the presence and the role of charges in the deswelling process. This is particularly important for the widely used poly(N-isopropylacrylamide)-based microgels, where the constituent monomers are neutral but charged groups arise due to the initiator molecules used in the synthesis. Here, we address this point combining experiments with state-of-the-art simulations to show that the microgel collapse does not happen in a homogeneous fashion, but through a two-step mechanism, entirely attributable to electrostatic effects. The signature of this phenomenon is the emergence of a minimum in the ratio between gyration and hydrodynamic radii at the VPT. Thanks to simulations of microgels with different cross-linker concentrations, charge contents, and charge distributions, we provide evidence that peripheral charges arising from the synthesis are responsible for this behavior and we further build a universal master curve able to predict the two-step deswelling. Our results have direct relevance on fundamental soft condensed matter science and on applications where microgels are involved, ranging from materials to biomedical technologies.

Accounting for Cooperativity in the Thermotropic Volume Phase Transition of Smart Microgels.

Project description:A full quantitative description of the swelling of smart microgels is still problematic in many cases. The original approach of Flory and Huggins for the monomer-solvent interaction parameter χ cannot be applied to some microgels. The reason for this obviously is that the cross-linking enhances the cooperativity of the volume phase transitions, since all meshes of the network are mechanically coupled. This was ignored in previous approaches, arguing with distinct transition temperatures for different meshes to describe the continuous character of the transition of microgels. Here, we adjust the swelling curves of a series of smart microgels using the Flory-Rehner description, where the polymer-solvent interaction parameter χ is modeled by a Hill-like equation for a cooperative thermotropic transition. This leads to a very good description of all measured microgel swelling curves and yields the physically meaningful Hill parameter ν. A linear decrease of ν is found with increasing concentration of the cross-linker N,N'-methylenebisacrylamide in the microgel particles p(NIPAM), p(NNPAM), and p(NIPMAM). The linearity suggests that the Hill parameter ν corresponds to the number of water molecules per network chain that cooperatively leave the chain at the volume phase transition. Driven by entropy, ν water molecules of the solvate become cooperatively "free" and leave the polymer network.

Phase Transitions in Chemically Fueled, Multiphase Complex Coacervate Droplets.

Project description:Membraneless organelles are droplets in the cytosol that are regulated by chemical reactions. Increasing studies suggest that they are internally organized. However, how these subcompartments are regulated remains elusive. Herein, we describe a complex coacervate-based model composed of two polyanions and a short peptide. With a chemical reaction cycle, we control the affinity of the peptide for the polyelectrolytes leading to distinct regimes inside the phase diagram. We study the transitions from one regime to another and identify new transitions that can only occur under kinetic control. Finally, we show that the chemical reaction cycle controls the liquidity of the droplets offering insights into how active processes inside cells play an important role in tuning the liquid state of membraneless organelles. Our work demonstrates that not only thermodynamic properties but also kinetics should be considered in the organization of multiple phases in droplets.

Programmable and Chemically Fueled DNA Coacervates by Transient Liquid-Liquid Phase Separation.

Project description:Multivalency-driven liquid-liquid phase separation (LLPS) is essential in biomolecular condensates to facilitate spatiotemporal regulation of biological functions. Providing programmable model systems would help to better understand the LLPS processes in biology, and furnish new types of compartmentalized synthetic reaction crucibles that exploit biological principles. Herein, we demonstrate a concept for programming LLPS using transient multivalency between ATP-driven sequence-defined functionalized nucleic acid polymers (SfNAPs), which serve as simple models for membrane-less organelles. The ATP-driven SfNAPs are transiently formed by an enzymatic reaction network (ERN) of concurrent ATP-powered DNA ligation and DNA restriction. The lifetimes can be programmed by the ATP concentration, which manifests on the LLPS length scale in tunable lifetimes for the all-DNA coacervates. Critically, the prominent programmability of the DNA-based building blocks allows to encode distinct molecular recognitions for multiple multivalent systems, enabling sorted LLPS and thus multicomponent DNA coacervates, reminiscent of the diverse membraneless organelles in biological systems. The ATP-driven coacervates are capable for multivalent trapping of micron-scale colloids and biomolecules to generate functions as emphasized for rate enhancements in enzymatic cascades. This work supports ATP-driven multivalent coacervation as a valuable mechanism for dynamic multicomponent and function biomolecular condensate mimics and for autonomous materials design in general.

A chemically fueled non-enzymatic bistable network.

Project description:One of the grand challenges in contemporary systems chemistry research is to mimic life-like functions using simple synthetic molecular networks. This is particularly true for systems that are out of chemical equilibrium and show complex dynamic behaviour, such as multi-stability, oscillations and chaos. We report here on thiodepsipeptide-based non-enzymatic networks propelled by reversible replication processes out of equilibrium, displaying bistability. Accordingly, we present quantitative analyses of the bistable behaviour, featuring a phase transition from the simple equilibration processes taking place in reversible dynamic chemistry into the bistable region. This behaviour is observed only when the system is continuously fueled by a reducing agent that keeps it far from equilibrium, and only when operating within a specifically defined parameter space. We propose that the development of biomimetic bistable systems will pave the way towards the study of more elaborate functions, such as information transfer and signalling.

Parasitic behavior in competing chemically fueled reaction cycles.

Project description:Non-equilibrium, fuel-driven reaction cycles serve as model systems of the intricate reaction networks of life. Rich and dynamic behavior is observed when reaction cycles regulate assembly processes, such as phase separation. However, it remains unclear how the interplay between multiple reaction cycles affects the success of emergent assemblies. To tackle this question, we created a library of molecules that compete for a common fuel that transiently activates products. Often, the competition for fuel implies that a competitor decreases the lifetime of these products. However, in cases where the transient competitor product can phase-separate, such a competitor can increase the survival time of one product. Moreover, in the presence of oscillatory fueling, the same mechanism reduces variations in the product concentration while the concentration variations of the competitor product are enhanced. Like a parasite, the product benefits from the protection of the host against deactivation and increases its robustness against fuel variations at the expense of the robustness of the host. Such a parasitic behavior in multiple fuel-driven reaction cycles represents a lifelike trait, paving the way for the bottom-up design of synthetic life.

A chemically fueled supramolecular glue for self-healing gels.

Project description:Chemically fueled supramolecular materials offer unique properties that include spatial and temporal control and even the ability to self-heal. Indeed, a few studies have demonstrated the ability to self-heal, however, the underlying mechanisms remain unclear. Here, we designed a peptide that forms a fibrillar network upon chemical fueling. We were surprised that the hydrogel could self-heal despite the lack of dynamics in the fiber assembly and disassembly. We explain this behavior by a mechanism that involves the chemically fueled peptide molecules that cannot self-assemble due to the lack of nucleation sites. When the fibers are perturbed, new nucleation sites form that help the assembly resulting in the healing of the damaged network. Furthermore, we generalized the behavior for other peptides. We refer to this non-assembling, chemically-fueled peptide as a molecular glue. In future work, we aim to explore whether this self-healing mechanism applies to more complex structures, narrowing the gap between biological and synthetic self-assemblies.

Design rules for reciprocal coupling in chemically fueled assembly.

Project description:Biology regulates the function and assembly of proteins through non-equilibrium reaction cycles. Reciprocally, the assembly of proteins can influence the reaction rates of these cycles. Such reciprocal coupling between assembly and reaction cycle is a prerequisite for behavior like dynamic instabilities, treadmilling, pattern formation, and oscillations between morphologies. While assemblies regulated by chemical reaction cycles gained traction, the concept of reciprocal coupling is under-explored. In this work, we provide two molecular design strategies to tweak the degree of reciprocal coupling between the assembly and reaction cycle. The strategies involve spacing the chemically active site away from the assembly or burying it into the assembly. We envision that design strategies facilitate the creation of reciprocally coupled and, by extension, dynamic supramolecular materials in the future.

Regulating DNA-Hybridization Using a Chemically Fueled Reaction Cycle.

Project description:Molecular machines, such as ATPases or motor proteins, couple the catalysis of a chemical reaction, most commonly hydrolysis of nucleotide triphosphates, to their conformational change. In essence, they continuously convert a chemical fuel to drive their motion. An outstanding goal of nanotechnology remains to synthesize a nanomachine with similar functions, precision, and speed. The field of DNA nanotechnology has given rise to the engineering precision required for such a device. Simultaneously, the field of systems chemistry developed fast chemical reaction cycles that convert fuel to change the function of molecules. In this work, we thus combined a chemical reaction cycle with the precision of DNA nanotechnology to yield kinetic control over the conformational state of a DNA hairpin. Future work on such systems will result in out-of-equilibrium DNA nanodevices with precise functions.
