Direct Synthesis of Unsymmetrical Dithioacetals.

ABSTRACT: Dithioacetals are a frequently used motif in synthetic organic chemistry and have recently seen increasing attention as structural motif in promising antiviral agents against plant pathogens. Most existing reports, however, only discuss symmetrical dithioacetals. Examples of mixed dithioacetals are scarce and no general method for the selective synthesis of these compounds exists. Herein, a synthetically simple general one-step protocol was developed for the synthesis of a broad range of unsymmetrical dithioacetals consisting of one aromatic and one aliphatic thiol moiety from the corresponding aldehyde/thiol mixture. The mixed S,S-acetals were obtained in high yields, and a great variety of functional groups was tolerated. Kinetic control enabled an excellent selectivity in regard to the unsymmetrical dithioacetal.

Project description:The first non-directed dehydrogenative phenone coupling method of methylarenes with aromatic C-H bonds, displaying a large substrate scope, is herein reported. This reaction represents a far more direct atom- and step-efficient alternative to the classical Friedel-Crafts or Suzuki-Miyaura derived acylation reactions. Project description:We demonstrate the use of the symmetrical diethyl(dimethyl)difluoromethylene bisphosphonate reagent for the synthesis of terminal and unsymmetrical difluoromethylene bisphosphonates, close analogues of biologically important molecules. The difference in reactivity of the methyl and ethyl groups in the symmetrical diethyl(dimthyl)difluoromethylene bisphosphonate is exploited in a stepwise demethylation-condensation sequence to functionalize either side of the reagent to allow the generation of a series of close bioisosteres of natural pyrophosphate molecules, including ADPr, CDP-glycerol and CDP-ribitol.

Investigations on Gold-Catalyzed Thioalkyne Activation Toward Facile Synthesis of Ketene Dithioacetals.

Project description:Nucleophilic addition to thioalkynes was investigated under various catalytic conditions with gold(I) complexes being identified as the optimal catalysts. Structural evaluation of the product revealed an unexpected cis-addition, arising from a gold-associated thioketene intermediate. Based on this interesting mechanistic insight, a gold(I)-catalyzed thioether addition to thioalkynes was developed as a novel approach to prepare ketene dithioacetals with good yields and high efficiency.

The hydroamination of allyl amine derivatives is a powerful approach for the synthesis of substituted 1,2-diamines. Herein, the rhodium-catalyzed hydroamination of primary and secondary allylic amines using diverse amine nucleophiles, including primary, secondary, acyclic, and cyclic aliphatic amines to access a wide range of unsymmetrical vicinal diamines, is presented. The utility of this methodology is further demonstrated through the rapid synthesis of several bioactive molecules and analogs.

Synthesis and Biological Activity of Unsymmetrical Monoterpenylhetaryl Disulfides.

Project description:New unsymmetrical monoterpenylhetaryl disulfides based on heterocyclic disulfides and monoterpene thiols were synthesized for the first time in 48-88% yields. Hydrolysis of disulfides with fragments of methyl esters of 2-mercaptonicotinic acid was carried out in 73-95% yields. The obtained compounds were evaluated for antioxidant, antibacterial, antifungal activity, cytotoxicity and mutagenicity.

HPLC purification technique: synthesis of unsymmetrical thiobarbituric acids.

Project description:Synthesis of thiobarbituric acids by the reaction of 1,3-disubstituted thioureas and malonic acid in acetyl chloride-acetic acid medium and synthesis of cyclized pyrimidin-7-one by the interaction of 1-(2-hydroxyethyl)-aryl thioureas, with malonic acid in p-tolyl sulphonic acid and acetyl chloride-acetic acid medium at room temperature stirring has been reported. Unsymmetrical Thienopentalenes: Synthesis, Optoelectronic Properties, and (Anti)aromaticity Analysis.

Project description:The synthesis and properties of a series of unsymmetrical thienopentalenes are explored, including both monoareno and diareno derivatives. For the synthesis of monoareno pentalenes, a carbopalladation cascade reaction between alkynes and gem-dibromoolefins was applied. Diareno pentalene derivatives were accessed via gold-catalyzed cyclization of diynes. Thiophene was fused to pentalene in two different geometries via its 2,3 and 3,4 bonds. 2,3-Fusion resulted in increased antiaromaticity of the pentalene unit compared to the 3,4-fusion both in the monoareno and diareno framework. Monothienopentalenes that contained the destabilizing 2,3-fusion could not be isolated. For diareno derivatives, the aromatic character of the different aryl groups fused to the pentalene was not independent. Destabilizing fusion on one side resulted in alleviated aromaticity on the other side and vice versa. The synthesized molecules were characterized experimentally by 1H NMR and UV-vis spectroscopies, cyclic voltammetry, and X-ray crystallography, and their aromatic character was assessed using magnetic (NICS and ACID) and electronic indices (MCI and FLU).

Aroylketene dithioacetal chemistry: facile synthesis of 4-aroyl-3-methylsulfanyl-2-tosylpyrroles from aroylketene dithioacetals and TosMIC.

Project description:The cycloaddition of the von Leusen's reagent (p-tolylsulfonyl)methyl isocyanide (TosMIC) to alpha-aroylketene dithioacetals (AKDTAs) in the presence of sodium hydride in THF at rt resulted in a facile synthesis of the 4-aroyl-3-methylsulfanyl-2-tosylpyrroles 3 in good yield along with a minor amount of 4-[(4-methylphenyl)sulfonyl]-1-[(methylsulfanyl)methyl]-1H-imidazole 4. This study lead to the synthesis of 2,3,4-trisubstiuted pyrroles having 1-naphthoyl/2-naphthoyl/ferrocenoyl/pyrenoyl scaffolds on C-4 of the pyrrole ring. In this transformation, the AKDTAs behave as a 3-(methylsulfanyl)-1-aryl-2-propyn-1-one (ArCOC identical withCSMe) equivalent. Competition experiments clearly showed that the cycloaddition of TosMIC to "push-pull" alkenes like AKDTAs is slower compared to general alpha,beta-unsaturated carbonyl compounds.
