Project description:This study explored the mechanical factors that determine accuracy of a baseball pitching. In particular, we focused on the mechanical parameters at ball release, referred to as release parameters. The aim was to understand which parameter has the most deterministic influence on pitch location by measuring the release parameters during actual pitching and developing a simulation that predicts the pitch location from given release parameters. By comparing the fluctuation of the simulated pitch location when varying each release parameter, it was found that the elevation pitching angle and speed significantly influenced the vertical pitch location, and the azimuth pitching angle significantly influenced the horizontal pitch location. Moreover, a regression model was obtained to predict the pitch location, and it became clear that the significant predictors for the vertical pitch location were the elevation pitching angle, the speed, and spin axis, and those for the horizontal pitch location were the azimuth pitching angle, the spin axis, and horizontal release point. Therefore, it was suggested that the parameter most affecting pitch location weas pitching angle. On the other hand, multiple regression analyses revealed that the relation between release parameters varied between pitchers. The result is expected to contribute to an understanding of the mechanisms underlying accurate ball control skill in baseball pitching.

Project description:Theory predicts the optimal timing of sex change will be the age or size at which half of an individual's expected fitness comes through reproduction as a male and half through reproduction as a female. In this way, sex allocation across the lifetime of a sequential hermaphrodite parallels the sex allocation of an outbreeding species exhibiting a 1:1 ratio of sons to daughters. However, the expectation of a 1:1 sex ratio is sensitive to variation in individual condition. If individuals within a population vary in condition, high-condition individuals are predicted to make increased allocations to the sex with the higher variance in reproductive success. An oft-cited example of this effect is seen in red deer, Cervus elaphus, in which mothers of high condition are more likely to produce sons, while those in low condition are more likely to produce daughters. Here, we show that individual condition is predicted to similarly affect the pattern of sex allocation, and thus the allocation of reproductive effort, in sequential hermaphrodites. High-condition sex-changers are expected to obtain more than half of their fitness in the high-payoff second sex and, as a result, are expected to reduce the allocation of reproductive effort in the initial sex. While the sex ratio in populations of sequential hermaphrodites is always skewed towards an excess of the initial sex, condition dependence is predicted to increase this effect.

Project description:We study the emergent behaviors of a population of swarming coupled oscillators, dubbed swarmalators. Previous work considered the simplest, idealized case: identical swarmalators with global coupling. Here we expand this work by adding more realistic features: local coupling, non-identical natural frequencies, and chirality. This more realistic model generates a variety of new behaviors including lattices of vortices, beating clusters, and interacting phase waves. Similar behaviors are found across natural and artificial micro-scale collective systems, including social slime mold, spermatozoa vortex arrays, and Quincke rollers. Our results indicate a wide range of future use cases, both to aid characterization and understanding of natural swarms, and to design complex interactions in collective systems from soft and active matter to micro-robotics.

Project description:Americans on the political left and right are engaged in a Culture War with one another, one that is often characterized by mutual fear, antipathy, and avoidance. Are there safe havens from the socially straining effects of this Culture War, times and places where Americans of different political stripes gather and put aside their political differences? Previous research (Chen & Rohla, 2018) implied that there might not be insofar as even intimate family gatherings seem to be subject to Culture War tensions. They found that politically diverse Thanksgiving Dinners were 35-70 minutes shorter than politically uniform ones, representing a 14-27% reduction in overall dinner duration. Noting analytical and methodological limitations in the prior analysis, we conducted two pre-registered studies to test whether diverse dinners are shorter than uniform ones and to attempt to conceptually replicate and extend this prior analysis. Individual analyses yielded mixed results, with null models generally supported but effect estimates generally overlapping with those of Chen and Rohla (2018). A mega-analysis found that, when controlling for various covariates, politically diverse dinners were 24 minutes shorter than politically uniform ones, 95% confidence interval = [9, 39], representing a 6% decrease in the total dinner time [2%-10%]. This final result successfully replicates Chen and Rohla (2018) both in terms of effect overlap and direct-and-significance criteria while nonetheless favoring the conclusion that politics is not straining family ties as much as previously thought.

Project description:Therapies that restore dystrophin expression are presumed to correct Duchenne muscular dystrophy (DMD), with antisense-mediated exon skipping being the leading approach. Here we aimed to determine whether exon skipping using a peptide-phosphorodiamidate morpholino oligonucleotide (PPMO) conjugate results in dose-dependent restoration of uniform dystrophin localization, together with correction of putative DMD serum and muscle biomarkers. Dystrophin-deficient mdx mice were treated with a PPMO (Pip9b2-PMO) designed to induce Dmd exon 23 skipping at single, ascending intravenous doses (3, 6, or 12 mg/kg) and sacrificed 2 weeks later. Dose-dependent exon skipping and dystrophin protein restoration were observed, with dystrophin uniformly distributed at the sarcolemma of corrected myofibers at all doses. Serum microRNA biomarkers (i.e., miR-1a-3p, miR-133a-3p, miR-206-3p, miR-483-3p) and creatinine kinase levels were restored toward wild-type levels after treatment in a dose-dependent manner. All biomarkers were strongly anti-correlated with both exon skipping level and dystrophin expression. Dystrophin rescue was also strongly positively correlated with muscle stiffness (i.e., Young's modulus) as determined by atomic force microscopy (AFM) nanoindentation assay. These data demonstrate that PPMO-mediated exon skipping generates myofibers with uniform dystrophin expression and that both serum microRNA biomarkers and muscle AFM have potential utility as pharmacodynamic biomarkers of dystrophin restoration therapy in DMD.

Project description:Key pointsDisruptions in circadian rhythms across an organism are associated with negative health outcomes, such as cardiometabolic and neurodegenerative diseases. Exercise has been proposed as a time cue for the circadian clock in rodents and humans. In this study, we assessed the effect of a single bout of endurance exercise on the skeletal muscle clock in vivo and a bout of muscle contractions in vitro. Timing of exercise or contractions influences the directional response of the muscle clock phase in vivo and in vitro. Our findings demonstrate that muscle contractions, as a component of exercise, can directly modulate the expression of muscle clock components in a time-of-day dependent manner.AbstractExercise has been proposed to be a zeitgeber for the muscle circadian clock mechanism. However, this is not well defined and it is unknown if exercise timing induces directional shifts of the muscle clock. Our purpose herein was to assess the effect of one bout of treadmill exercise on skeletal muscle clock phase changes. We subjected PERIOD2::LUCIFERASE mice (n = 30F) to one 60 min treadmill exercise bout at three times of day. Exercise at ZT5, 5 h after lights on, induced a phase advance (100.2 ± 25.8 min; P = 0.0002), whereas exercise at ZT11, 1 h before lights off, induced a phase delay (62.1 ± 21.1 min; P = 0.0003). Exercise at ZT17, middle of the dark phase, did not alter the muscle clock phase. Exercise induces diverse systemic changes so we developed an in vitro model system to examine the effects of contractile activity on muscle clock phase. Contractions applied at peak or trough Bmal1 expression induced significant phase delays (applied at peak: 27.2 ± 10.2 min; P = 0.0017; applied at trough: 64.6 ± 6.5 min, P < 0.0001). Contractions applied during the transition from peak to trough Bmal1 expression induced a phase advance (49.8 ± 23.1 min; P = 0.0051). Lastly, contractions at different times of day resulted in differential changes of core clock gene expression, demonstrating an exercise and clock interaction, providing insight into potential mechanisms of exercise-induced phase shifts. These data demonstrate that muscle contractions, as part of exercise, are sufficient to shift the muscle circadian clock phase, likely through changes in core clock gene expression. Additionally, our findings that exercise induces directional muscle clock phase changes confirms that exercise is a bona fide environmental time cue for skeletal muscle.

Project description:Adult skeletal muscles retain an adaptive capacity to switch between slow- and fast-twitch properties that largely depend on motoneuron activity. The NFAT (nuclear factor of activated T cells) family of calcium-dependent transcription factors has been implicated in the up-regulation of genes encoding slow contractile proteins in response to slow-patterned motoneuron depolarization. Here, we demonstrate an unexpected, novel function of NFATc1 in slow-twitch muscles. Using the troponin I fast (TnIf) intronic regulatory element (FIRE), we identified sequences that down-regulate its function selectively in response to patterns of electrical activity that mimic slow motoneuron firing. A bona fide NFAT binding site in the TnIf FIRE was identified by site-directed mutations and by electrophoretic mobility and supershift assays. The activity-dependent transcriptional repression of FIRE is mediated through this NFAT site and, importantly, its mutation did not alter the up-regulation of TnIf transcription by fast-patterned activity. siRNA-mediated knockdown of NFATc1 in adult muscles resulted in ectopic activation of the FIRE in the slow soleus, without affecting enhancer activity in the fast extensor digitorum longus muscle. These findings demonstrate that NFAT can function as a repressor of fast contractile genes in slow muscles and they exemplify how an activity pattern can increase or decrease the expression of distinct contractile genes in a use-dependent manner as to enhance phenotypic differences among fiber types or induce adaptive changes in adult muscles.

Project description:Why do professional athletes and musicians exhibit individually different motion patterns? For example, baseball pitchers generate various pitching forms, e.g., variable wind-up, cocking, and follow-through forms. However, they commonly rotate their wrists and fingers at increasingly high speeds via shoulder and trunk motions. Despite the universality of common and individually different motion patterns in skilled movements, the abovementioned question remains unanswered. Here, we focus on a motion required to hit a snare drum, including the indirect phase of task achievement (i.e., the early movement and mid-flight phases) and the direct phase of task achievement (i.e., the hit phase). We apply tensor decomposition to collected kinematic data for the drum-hitting motion, enabling us to decompose high-dimensional and time-varying motion data into individually different and common movement patterns. As a result, individually different motion patterns emerge during the indirect phase of task achievement, and common motion patterns are evident in the direct phase of task achievement. Athletes and musicians are thus possibly allowed to perform individually different motion patterns during the indirect phase of task achievement. Additionally, they are required to exhibit common patterns during the direct phase of task achievement.

Project description:The intracellular signals that convert fast and slow motor neuron activity into muscle fiber type specific transcriptional programs have only been partially defined. The calcium/calmodulin-dependent phosphatase calcineurin (Cn) has been shown to mediate the transcriptional effects of motor neuron activity, but precisely how 4 distinct muscle fiber types are composed and maintained in response to activity is largely unknown. Here, we show that 4 nuclear factor of activated T cell (NFAT) family members act coordinately downstream of Cn in the specification of muscle fiber types. We analyzed the role of NFAT family members in vivo by transient transfection in skeletal muscle using a loss-of-function approach by RNAi. Our results show that, depending on the applied activity pattern, different combinations of NFAT family members translocate to the nucleus contributing to the transcription of fiber type specific genes. We provide evidence that the transcription of slow and fast myosin heavy chain (MyHC) genes uses different combinations of NFAT family members, ranging from MyHC-slow, which uses all 4 NFAT isoforms, to MyHC-2B, which only uses NFATc4. Our data contribute to the elucidation of the mechanisms whereby activity can modulate the phenotype and performance of skeletal muscle.

Project description:Individuals with lesions of central motor pathways frequently suffer from sustained involuntary muscle activity. This symptom shares clinical characteristics with dystonia but is observable in individuals classified as spastic. The term spastic dystonia has been introduced, although the underlying mechanisms of involuntary activity are not clarified and vary between individuals depending on the disorder. This study aimed to investigate the nature and pathophysiology of sustained involuntary muscle activity in adults with cerebral palsy and stroke. Seventeen adults with cerebral palsy (Gross Motor Function Classification System I-V), 8 adults with chronic stroke and 14 control individuals participated in the study. All individuals with cerebral palsy or stroke showed increased resistance to passive movement with Modified Ashworth Scale >1. Two-minute surface EMG recordings were obtained from the biceps muscle during attempted rest in three positions of the elbow joint; a maximally flexed position, a 90-degree position and a maximally extended position. Cross-correlation analysis of sustained involuntary muscle activity from individuals with cerebral palsy and stroke, and recordings of voluntary isometric contractions from control individuals were performed to examine common synaptic drive. In total, 13 out of 17 individuals with cerebral palsy and all 8 individuals with stroke contained sustained involuntary muscle activity. In individuals with cerebral palsy, the level of muscle activity was not affected by the joint position. In individuals with stroke, the level of muscle activity significantly (P < 0.05) increased from the flexed position to the 90 degree and extended position. Cumulant density function indicated significant short-term synchronization of motor unit activities in all recordings. All groups exhibited significant coherence in the alpha (6-15 Hz), beta (16-35 Hz) and early gamma band (36-60 Hz). The cerebral palsy group had lower alpha band coherence estimates, but higher gamma band coherence estimates compared with the stroke group. Individuals with increased resistance to passive movement due to cerebral palsy or stroke frequently suffer sustained involuntary muscle activity, which cannot exclusively be described by spasticity. The sustained involuntary muscle activity in both groups originated from a common synaptic input to the motor neuron pool, but the generating mechanisms could differ between groups. In cerebral palsy it seemed to originate more from central mechanisms, whereas peripheral mechanisms likely play a larger role in stroke. The sustained involuntary muscle activity should not be treated simply like the spinal stretch reflex mediated symptom of spasticity and should not either be treated identically in both groups.
