The mitochondrial genome of Asyndetus clavipes (Diptera: Dolichopodidae).

ABSTRACT: The long-legged fly Asyndetus clavipes belongs to the subfamily Diaphorinae of Dolichopodidae. The mitogenome of A. clavipes was sequenced, the first representative complete mitogenome from this subfamily. This mitogenome is 15,385 bp in size, includes 13 protein-coding genes, 22 transfer RNAs, and two ribosomal RNAs. All genes have the same location and coding strand as in other published species of Dolichopodidae. Nucleotide composition is biased toward A and T, which together made up 78.6% of the entire genome. Bayesian inference strongly supported the monophyly of Empidoidea, Empididae and Dolichopodidae, with the phylogenetic relationships within Empidoidea: ((Dolichopodinae + Neurigoninae) + Diaphorinae) + ((Trichopezinae + (Empidinae + Oreogetoninae)) + Ocydromiinae).

Project description:The mitochondrial genome of Gymnopternus bomiensis (Yang, 1996) (Diptera: Dolichopodidae) has been reported in this study. This is the first mitogenome representative of Gymnopternus. The sequenced region is determined to be 15,212 bp, including 13 protein-coding genes, 2 ribosomal RNAs, 22 transfer RNAs, and a partial A-T rich region. The nucleotide composition biased toward A and T, and the overall A + T% was up to 74.0％. Additionally, we reconstructed the phylogeny of relative species using 13 PCRs and two rRNAs. Bayesian inference analysis strongly supported the monophyly of Dolichopodinae. It also suggested that Gymnopternus is the sister group of Dolichopus.

The newly sequenced mitogenome of L. dentalis is a new representative of the subfamily. The nearly complete mitogenome is 15,124 bp in length, consisting of 13 protein-coding genes (PCGs), 2 ribosomal RNA genes (rRNAs), and 22 transfer RNA genes (tRNAs). All genes have similar locations and encoding directions with that of other published mitogenomes of Dolichopodidae. The nucleotide composition biases toward A and T with the overall A + T % is 73.9%. All protein-coding genes initiate with standard start codon ATN except COI and NAD1, and TAA/TAG are conventionally used as stop codons. All tRNAs, ranging from 62 to 71 bp, have a clover-leaf structure. Based on the result of the phylogenetic analysis, Dolichopodidae and Empididae were monophyletic, and the relationships among subfamilies of Dolichopodidae were Diaphorinae + (Peloropeodinae + (Xanthochlorinae + (Medeterinae + Dolichopodinae))). The monophyly of the subfamily Dolichopodinae and the sister relationship between Dolichopus and Lichtwardtia were also strongly supported.

The complete mitochondrial genome of Trichonephila clavipes (Araneae: Araneidae).

Project description:Trichonephila clavipes (Linnaeus, 1767) is known as a golden silk orb-weaver and belongs to the family Araneidae. T. clavipes is one of the few spider species whose genome has been reported and model organism for a molecular biology. Here, we present the complete mitochondrial genome sequence (mtDNA) of T. clavipes. The sequence was obtained using a long-read Nanopore technology and corrected with an Illumina technology. The circular genome is 14,902 bp in length, and the AT content was 77.21%. The T. clavipes mitochondrial genome contains 13 protein-coding genes (PCGs), 22 tRNA genes, and 2 rRNA genes. The majority of PCGs were found on the heavy strain.

The Dolichopodidae (Diptera) of Montserrat, West Indies.

Project description:The long-legged flies (Dolichopodidae) of the island of Montserrat in the Lesser Antilles have been surveyed and include 63 species in 27 genera. The following eleven new species are described and illustrated: Amblypsilopus marskeae sp. nov., Medetera iviei sp. nov., Medetera montserratensis sp. nov., Systenus ladonnae sp. nov., Thrypticus mediofuscus sp. nov., Chrysotus antillensis sp. nov., Chrysotus callichromoides sp. nov., Chrysotus interfrons sp. nov., Chrysotus montserratensis sp. nov., Diaphorus robinsoni sp. nov., and Sympycnus montserratensis sp. nov. Six species have only been found on Montserrat (ca. 10% endemicity). Keys are provided to the genera and species on Montserrat, their known distribution summarized, and additional new island records provided for many species. Asyndetus wirthi Robinson is synonymized with A. interruptus (Loew) and Achradocera apicalis (Aldrich) is removed from synonymy with A. barbata (Loew). Diaphorus flavipes Aldrich is transferred to Chrysotus as a new combination. A new replacement name, Chrysotus milvadu nom. nov., is provided for the Nearctic Chrysotus parvulus Van Duzee. Lectotypes are designated for Achradocera apicalis (Aldrich) and Asyndetus fratellus Aldrich. The fauna of Montserrat is summarized and compared with that of Dominica. Collecting methods are compared and threats to the dolichopodid fauna of Montserrat discussed.
