Characterization of the mitochondrial genome of Tinda javana (Macquart, 1838) (Diptera: Stratiomyidae: Pachygastrinae).

ABSTRACT: We have assembled and annotated the mitochondrial genome of Tinda javana (Macquart, 1838) (a species of soldier fly) in this study. It is 15,495 bp in length, including 13 protein-coding genes (PCGs), two ribosomal RNA genes (rRNAs), 22 transfer RNA genes (tRNAs), and a large non-coding control region (length: 704 bp). The nucleotide composition of whole mitochondrial genome biases toward A and T (75.5%). Most PCGs use ATN as initiation codon, except for cox1 which starts with CGA. All PCGs end with common termination codon TAA/G. Phylogenetic analyses based on the nucleotide sequence data supported the monophyly of Stratiomyidae and the sister relationship between Pachygastrinae and the clade (Nemotelinae + (Hermetiinae + Sarginae)).

Project description:The complete mitochondrial genome of Parastratiosphecomyia szechuanensis Lindner, 1954 was sequenced and analyzed in this study. The mitochondrial genome is 16,414 bp in length, including the 37 typical insect mitochondrial genes and a large control region. All PCGs end with complete termination codon TAA or TAG. Most PCGs initiated by standard start codon ATN, except for cox1 which starts with TCG. The phylogenetic analysis based on the nucleotide sequence data of 13 PCGs recovered the monophyly of Stratiomyidae and the sister relationship between Xylomyidae and Stratiomyidae.

In this study, the complete mitochondrial genome of H. illucens was investigated. The total number of nucleic acids were 15,698 bp. The genome contains 13 protein-coding genes, 22 tRNA genes, 2 rRNA genes, and a non-coding control region. The phylogenetic relationship of H. illucens and its 12 related species was reconstructed to confirm the taxonomic status of our sample.

Complete mitochondrial genome of the decomposer Ptecticus tenebrifer Walker, 1849 (Insecta: Diptera: Stratiomyidae).

Project description:Ptecticus tenebrifer (Walker, 1849), which is endemic in East Asia, plays a critical role in natural processing of organic waste. In this study, we found that the complete mitochondrial genome of P. tenebrifer is 15681 bp in length obtained from the MGIseq system (MGI) with 150 bp paired-end reads. As with typical animal mitochondrial genomes, it contains 13 protein coding genes, 22 tRNA genes, and 2 rRNA genes. The phylogenetic analysis was performed using the maximum likelihood method with a bootstrap value of 500 and the Tamura-Nei model and reveals the close relationship with P. aurifer.

Updated Italian checklist of Soldier Flies (Diptera, Stratiomyidae).

Project description:An updated checklist for Stratiomyidae of Italy is presented. Previous knowledge and information are put together in a comprehensive way, integrated also with results obtained by sampling with Malaise traps in some of the test areas of the LIFE+ project ManFor C.BD. At the time of writing, with 91 known species, the Italian fauna of Stratiomyidae is the richest in Europe. Neopachygaster meromelas (Dufour, 1841) and Zabrachia minutissima (Zetterstedt, 1838) are new to the Italian fauna. A comprehensive key to the European species of Chorisops Rondani, 1856 is given.

Hexatoma (Eriocera) Macquart (Diptera, Limoniidae) from Xizang, China.

Project description:One new species of the subgenus Eriocera Macquart,1838, Hexatoma (Eriocera) xizangensissp. nov. is described and illustrated from Xizang, China. The following four species are re-described and reported from Xizang for the first time: H. (E.) latigrisea Alexander, 1971, H. (E.) nepalensis (Westwood, 1836), H. (E.) paragnava Alexander, 1973 and H. (E.) perhirsuta Alexander, 1973. A key to the species of Eriocera from Xizang is presented.

Two new species of Oxycera (Diptera, Stratiomyidae) from Ningxia, China.

Project description:Two new speices, Oxycera rozkosnyisp. n. and Oxycera ningxiaensissp. n., are described from Liupanshan Nature Reserve, Ningxia Hui Autonomous Region, Northwest China. All essential diagnostic characters are figured and possible relationships of both taxa are briefly discussed, and a new key to species of Oxycera from China. The type specimens are deposited in the Institute of Entomology, Guizhou University, Guiyang (GUGC).

The family Stratiomyidae in Egypt and Saudi Arabia (Diptera: Stratiomyoidea).

Project description:BackgroundThis study systematically catalogues all known taxa of the family Stratiomyidae in Egypt and Saudi Arabia. It is one in a series of planned studies aiming to catalogue the whole order in both countries.New informationTwenty species, belonging to seven genera and three subfamilies (Pachygastrinae, Stratiomyinae and Nemotelinae), are treated. One of these genera, Oplodontha and two species, Oplodontha pulchriceps Loew and Oxycera turcica Üstüner & Hasbenli, are recorded herein for the first time from Saudi Arabia. A lectotype for Nemotelus matrouhensis Mohammad et al., 2009 is designated. An updated classification, synonymies, type localities, world and local distributions, dates of collection and some coloured photographs are provided.

Proteomics and Lipidomics of Black Soldier Fly (Diptera: Stratiomyidae) and Blow Fly (Diptera: Calliphoridae) Larvae.

Project description:Farming insects has recently emerged as a new source of protein and lipid production. To date, research has mostly focused on food applications of insects. Focusing on nonfood potential of oil and proteins of insects, high-throughput studies of insect lipids and proteins are needed. We performed proteomics and lipidomics investigation on black soldier fly (Hermitia illucens) and blow fly (Lucilia sericata) larvae to investigate new potential and applications. We used mass spectrometry for proteomics and lipidomics analysis of control and treated larvae. Treatment was performed by incubation with a biological decomposer. We provide the list of all fatty acids with their concentration in control and treated larvae. This result showed high levels of lauric acid in black soldier fly, which could even increase after biological decomposition. Proteomics analysis showed the presence of proteins like collagen of cosmetic interest, and proteins with antimicrobial properties such as phenoloxidases and enzymatic activities, such as amylase and trypsin. Insects harbor high potential for nonfood usage as additives, antimicrobial effects, and even pharmaceuticals and cosmetics. These data open avenues for future research in pharmacological and cosmetic approaches to find new molecules of interests.

Characterization of the complete mitochondrial genome of Drosophila busckii (Diptera: Drosophilidae).

Project description:In this study, we sequenced and analyzed the complete mitochondrial genome of Drosophila busckii (Diptera: Drosophilidae). The mitogenome was 15,214 bp in length, containing 13 protein-coding genes, 22 tRNA genes, 2 rRNA genes. The gene organization of D. busckii is identical to the ancestral gene arrangement found in most insects. All protein-coding genes started with ATN, except for cox2 and nad5, which used noncanonical codon TTG and GTG, respectively.

Starvation Alters Gut Microbiome in Black Soldier Fly (Diptera: Stratiomyidae) Larvae.

Project description:Unlike for vertebrates, the impact of starvation on the gut microbiome of invertebrates is poorly studied. Deciphering shifts in metabolically active associated bacterial communities in vertebrates has led to determining the role of the associated microbiome in the sensation of hunger and discoveries of associated regulatory mechanisms. From an invertebrate perspective, such as the black soldier fly, such information could lead to enhanced processes for optimized biomass production and waste conversion. Bacteria associated with food substrates of black soldier fly are known to impact corresponding larval life-history traits (e.g., larval development); however, whether black soldier fly larval host state (i.e., starved) impacts the gut microbiome is not known. In this study, we measured microbial community structural and functional shifts due to black soldier fly larvae starvation. Data generated demonstrate such a physiological state (i.e., starvation) does in fact impact both aspects of the microbiome. At the phylum level, community diversity decreased significantly during black soldier fly larval starvation (p = 0.0025). Genus level DESeq2 analysis identified five genera with significantly different relative abundance (q < 0.05) across the 24 and 48 H post initiation of starvation: Actinomyces, Microbacterium, Enterococcus, Sphingobacterium, and Leucobacter. Finally, we inferred potential gene function and significantly predicted functional KEGG Orthology (KO) abundance. We demonstrated the metabolically active microbial community structure and function could be influenced by host-feeding status. Such perturbations, even when short in duration (e.g., 24 H) could stunt larval growth and waste conversion due to lacking a full complement of bacteria and associated functions.
