Characterization of the complete mitochondrial genome of the Qiaoke sheep (Ovis aries).

ABSTRACT: Qiaoke sheep (Ovis aries) is a local sheep breed in Gansu province, China. It is a kind of Tibetan sheep that used for both meat and wool after long-term breeding. In this paper, the complete mitochondrial genome of Qiaoke sheep was sequenced. The total length of the mitochondrial genome is 16616 bp, and the base composition is 33.65% A, 13.14% G, 25.88% C and 27.33% T. The genome has a total of 37 genes, including 13 protein-coding genes, 22 tRNA genes, two ribosomal RNA genes and a control region (D-loop region). This complete sequence would enlarge useful genomic information for further studies.

Project description:Jialuo sheep is one of the important production materials of herdsmen in the pastoral areas of northwestern Sichuan in China. We reported the complete mitogenome of Jialuo sheep for the first time. It is 16,617 bp in length, containing 13 protein-coding genes, 2 ribosomal RNA genes, 22 transfer RNA genes, and a control region (D-loop). In the evolutionary relationship, the mitogenome of Jialuo sheep is closer to Texel sheep. This report will help the further studies of sheep species classification and resource protection.

The complete mitochondrial genome sequence and phylogenetic analysis of Alpine Merino sheep (Ovis aries).

Project description:Alpine Merino sheep is one of the most important fine-wool sheep breeds in China. In this study, we present the complete mitogenome of Alpine Merino sheep for the first time. The genome has a length of 16,619bp, containing 13 protein-coding genes, 2 ribosomal RNA genes, 22 transfer RNA genes, and a control region (D-loop). Phylogenetically, the Alpine Merino sheep is closer to Oula Tibetan sheep and Tashkurgan sheep. This report provided new data for the phylogeny of Alpine Merino sheep.

Phylogenetic position of a copper age sheep (Ovis aries) mitochondrial DNA.

Project description:BackgroundSheep (Ovis aries) were domesticated in the Fertile Crescent region about 9,000-8,000 years ago. Currently, few mitochondrial (mt) DNA studies are available on archaeological sheep. In particular, no data on archaeological European sheep are available.Methodology/principal findingsHere we describe the first portion of mtDNA sequence of a Copper Age European sheep. DNA was extracted from hair shafts which were part of the clothes of the so-called Tyrolean Iceman or Ötzi (5,350-5,100 years before present). Mitochondrial DNA (a total of 2,429 base pairs, encompassing a portion of the control region, tRNA(Phe), a portion of the 12S rRNA gene, and the whole cytochrome B gene) was sequenced using a mixed sequencing procedure based on PCR amplification and 454 sequencing of pooled amplification products. We have compared the sequence with the corresponding sequence of 334 extant lineages.Conclusions/significanceA phylogenetic network based on a new cladistic notation for the mitochondrial diversity of domestic sheep shows that the Ötzi's sheep falls within haplogroup B, thus demonstrating that sheep belonging to this haplogroup were already present in the Alps more than 5,000 years ago. On the other hand, the lineage of the Ötzi's sheep is defined by two transitions (16147, and 16440) which, assembled together, define a motif that has not yet been identified in modern sheep populations.

Phylogenetic and characterization of the complete mitochondrial genome relationship of Argali sheep (Ovis ammon).

Project description:In this study, we sequenced the complete mitochondrial genome of Argali sheep (Ovis ammon). The total length was 16,612 bp, which contained 13 protein-coding genes (PCGs), 22 tRNA genes, 2 rRNA genes, and a control region (D-loop). Eight tRNA genes with one PCG (ND6) encoded on the L-strand, others were encoded on the H-strand. The neighbor-joining analysis shows that Argali sheep has a close relationship with the same genus species of Ovis aries and Ovis orientalis. This study has provided new data for the phylogeny of Argali sheep.

Do sheep (Ovis aries) discriminate human emotional odors?

Project description:While sheep can detect and discriminate human emotions through visual and vocal cues, their reaction to human body odors remains unknown. The present study aimed to determine whether sheep (Ovis aries) can detect human odors, olfactorily discriminate stressed from non-stressed individuals, and behave accordingly based on the emotional valence of the odors. Axillary secretions from 34 students were collected following an oral examination (stress odor) or a regular class (non-stress odor). Fourteen female and 15 male lambs were then exposed to these odors through a habituation-dishabituation procedure. The habituation stimulus was presented four times for one minute, followed by the dishabituation stimulus presented once for one minute. Behavioral variables included spatiality relative to target odors, approach/withdrawal, ear positioning, sniffing, ingestion, and vocalization. Both female and male lambs more often positioned their ears backwards/forwards, and asymmetrically when exposed to the dishabituation stimulus, but regardless of their stress or non-stress value. They also changed their approach behavior when exposed to the dishabituation stimuli. Lambs displayed some behavioral signs of discrimination between the habituation and dishabituation odors, but regardless of their relation to stress or non-stress of human donors. In sum, this exploratory study suggests that young sheep respond negatively to the odor of unfamiliar humans, without showing any specific emotional contagion related to the stress odor. This exploratory study suggests young ovines can detect human body odor, a further step toward understanding the human-sheep relationship.

Identification and characterization of microRNAs in the intestinal tissues of sheep (Ovis aries).

Project description:Sheep are small ruminants, and their long intestines exhibit high digestive and absorptive capacity in many different rearing conditions; however, the genetic bases of this characteristic remains unclear. MicroRNAs (miRNAs) play a major role in maintaining both intestinal morphological structure as well as in regulating the physiological functions of this organ. However, no study has reported on the miRNA expression profile in the intestinal tissues of sheep. Here, we analyzed and identified the miRNA expression profile of three different intestinal tissues (i.e., duodenum, cecum, and colon) of sheep (Ovis aries) using high-throughput sequencing and bioinformatic methods. In total, 106 known miRNAs were identified, 458 conserved miRNAs were detected, 192 unannotated novel miRNAs were predicted, and 195 differentially expressed miRNAs were found between the different tissues. Additionally, 3,437 candidate target genes were predicted, and 17 non-redundant significantly enriched GO terms were identified using enrichment analysis. A total of 99 candidate target genes were found to significantly enriched in 4 KEGG biological pathways. A combined regulatory network was constructed based on 92 metabolism-related candidate target genes and 65 differentially expressed miRNAs, among which 7 miRNAs were identified as hub miRNAs. Via these mechanisms, miRNAs may play a role in maintaining intestinal homeostasis and metabolism. This study helps to further explain the mechanisms that underlie differences in tissue morphology and function in three intestinal segments of sheep.

Project description:Here, we analyzed and identified the miRNA expression profile of three different intestinal tissues (i.e., duodenum, cecum, and colon) of sheep (Ovis aries) using high-throughput sequencing and bioinformatic methods. In total, 128 known miRNAs were identified, 526 novel miRNAs were predicted, and 202 differentially expressed miRNAs were found between the different tissues. Additionally, 4,422 candidate target genes were predicted, and 185 non-redundant GO annotation terms were identified using enrichment analysis. A total of 529 target genes were found to participate in 37 KEGG biological pathways, and 270 of these genes were significantly enriched in the metabolism category.

Proteomic analyses of sheep (ovis aries) embryonic skeletal muscle.

Project description:The growth and development of embryonic skeletal muscle plays a crucial role in sheep muscle mass. But proteomic analyses for embryonic skeletal development in sheep had been little involved in the past research. In this study, we explored differential abundance proteins during embryonic skeletal muscle development by the tandem mass tags (TMT) and performed a protein profile analyses in the longissimus dorsi of Chinese merino sheep at embryonic ages Day85 (D85N), Day105 (D105N) and Day135 (D135N). 5,520 proteins in sheep embryonic skeletal muscle were identified, and 1,316 of them were differential abundance (fold change ≥1.5 and p-value < 0.05). After the KEGG enrichment analyses, these differential abundance proteins were significant enriched in the protein binding, muscle contraction and energy metabolism pathways. After validation of the protein quantification with the parallel reaction monitoring (PRM), 41% (16/39) significant abundance proteins were validated, which was similar to the results of protein quantification with TMT. All results indicated that D85N to D105N was the stage of embryonic muscle fibers proliferation, while D105N to D135N was the stage of their hypertrophy. These findings provided a deeper understanding of the function and rules of proteins in different phases of sheep embryonic skeletal muscle growth and development.

The whole genome's base composition is 33.7% A, 25.8% C, 13.1% G and 27.4% T, and the percentage of AT-rich regions is 61.1%.

Sarcocystis species in wild and domestic sheep (Ovis ammon and Ovis aries) from China.

Project description:BackgroundSarcocystis species are intracellular protozoan parasites that can pose a threat to animal health and food safety. The aim of this study was to investigate the prevalence of infection with Sarcocystis infection in sheep from China.ResultsIn total, 52.51% (335/638) of tissue samples from domestic sheep contained sarcocysts through examination by light microscopy. The organisms were identified as S. tenella and S. arieticanis by molecular assays. Macroscopic S. gigantea and S. medusiformis were not found. The average sarcocysts loading was 18.07 ± 29.87 per square centimeter in the myocardium of domestic sheep. Furthermore, two specimens of argali (Ovis ammon) were examined and sarcocysts were found in the myocardium of one animal. According to the sequence of the cox1 gene of sarcocysts from argali, it was speculated as S. tenella.ConclusionsWe found a high prevalence and parasite load of Sarcocystis in sheep from both central and northwest China. This report is the first to indicate that argali may be a natural intermediate host for S. tenella.
