Using <i>in vivo</i> muscle stem cell (satellite cell)-specific extracellular vesicle (EV) tracking, satellite cell depletion, <i>in vitro</i> cell culture, and single-cell RNA sequencing, we show satellite cells communicate with other cells in skeletal muscle during mechanical overload.
