Spatial phylogenetics of butterflies in relation to environmental drivers and angiosperm diversity across North America.

ABSTRACT: Broad-scale, quantitative assessments of insect biodiversity and the factors shaping it remain particularly poorly explored. Here we undertook a spatial phylogenetic analysis of North American butterflies to test whether climate stability and temperature gradients have shaped their diversity and endemism. We also performed the first quantitative comparisons of spatial phylogenetic patterns between butterflies and flowering plants. We expected concordance between the two groups based on shared historical environmental drivers and presumed strong butterfly-host plant specializations. We instead found that biodiversity patterns in butterflies are strikingly different from flowering plants, especially warm deserts. In particular, butterflies show different patterns of phylogenetic clustering compared with flowering plants, suggesting differences in habitat conservation between the two groups. These results suggest that shared biogeographic histories and trophic associations do not necessarily assure similar diversity outcomes. The work has applied value in conservation planning, documenting warm deserts as a North American butterfly biodiversity hotspot.

Project description:The edge of the North West European Shelf (NWES) is characterised by a steep continental slope and a northward flowing slope current. These topographic/hydrographic features separate oceanic water and shelf water masses hence potentially separate phytoplankton communities. The slope current may facilitate the advective transport of phytoplankton, with mixing at the shelf edge supporting nutrient supply and therefore phytoplankton production. On the west Scottish shelf in particular, little is known about the phytoplankton communities in and around the shelf break and adjacent waters. Hence, to improve our understanding of environmental drivers of phytoplankton communities, biological and environmental data were collected on seven cross-shelf transects across the Malin and Hebridean Shelves during autumn 2014. Density profiles indicated that shelf break and oceanic stations had a 100 m deep mixed surface layer while stations on the shelf were generally well mixed. Analysis of similarity and multidimensional scaling of phytoplankton counts revealed that phytoplankton communities on the shelf were significantly different to those found at the shelf break and at oceanic stations. Shelf stations were dominated by dinoflagellates, with diatoms contributing a maximum of 37% of cells. Shelf break and oceanic stations were also dinoflagellate dominated but displayed a lower species diversity. Significant difference between shelf and shelf break stations suggested that the continental slope limited cross shelf phytoplankton exchange. Northern and southern phytoplankton communities on the shelf were approximately 15% dissimilar while there was no latitudinal gradient for stations along the slope current, suggesting this current provided south to north connectivity. Fitting environmental data to phytoplankton ordination showed a significant relationship between phytoplankton community dissimilarities and nutrient concentrations and light availability on the shelf compared to shelf break and oceanic stations in the study area.

Project description:In the search for novel drug candidates, diverse environmental microbiomes have been surveyed for their secondary metabolite biosynthesis potential, yet little is known about the biosynthetic diversity encoded by divergent microbiomes from different ecosystems, and the environmental parameters driving this diversity. Here, we used targeted amplicon sequencing of adenylation (AD) and ketosynthase (KS) domains along with 16S sequencing to delineate the unique biosynthetic potential of microbiomes from three separate habitats (soil, water, and sediments) exhibiting unique small spatial scale physicochemical gradients. The estimated richness of AD domains was highest in marine sediments with 656 ± 58 operational biosynthetic units (OBUs), while the KS domain richness was highest in soil microbiomes with 388 ± 67 OBUs. Microbiomes with rich and diverse bacterial communities displayed the highest PK potential across all ecosystems, and on a small spatial scale, pH and salinity were significantly, positively correlated to KS domain richness in soil and aquatic systems, respectively. Integrating our findings, we were able to predict the KS domain richness with a RMSE of 31 OBUs and a R2 of 0.91, and by the use of publicly available information on bacterial richness and diversity, we identified grassland biomes as being particularly promising sites for the discovery of novel polyketides. Furthermore, a focus on acidobacterial taxa is likely to be fruitful, as these were responsible for most of the variation in biosynthetic diversity. Overall, our results highlight the importance of sampling diverse environments with high taxonomic diversity in the pursuit for novel secondary metabolites. IMPORTANCE To counteract the antibiotic resistance crisis, novel anti-infective agents need to be discovered and brought to market. Microbial secondary metabolites have been important sources of inspiration for small-molecule therapeutics. However, the isolation of novel antibiotics is difficult, and the risk of rediscovery is high. With the overarching purpose of identifying promising microbiomes for discovery of novel bioactivity, we mapped out the most significant drivers of biosynthetic diversity across divergent microbiomes. We found the biosynthetic potential to be unique to individual ecosystems, and to depend on bacterial taxonomic diversity. Within systems, and on small spatial scales, pH and salinity correlated positively to the biosynthetic richness of the microbiomes, Acidobacteria representing the taxa most highly associated with biosynthetic diversity. Ultimately, understanding the key drivers of the biosynthesis potential of environmental microbiomes will allow us to focus bioprospecting efforts and facilitate the discovery of novel therapeutics.
