The method of colloid titration with poly(diallyldimethylammonium) chloride has been improved to detect the endpoint with an off-vessel light reflectance sensor. The digital color sensor used measures light reflectance by means of light guides, with no immersion into the reaction solution. In such a method, the optical signal is free of disturbances caused by sticky flocs in the solution. Project description:Nanoplastics with small particle size and high surface area/volume ratio make them easy to absorb environmental pollutants and affect their bioavailability. In this study, polystyrene nanoplastic beads (PS-NPBs) with a particle size of 100nm and sunscreen butyl methoxydibenzoylmethane (BMDBM) contained in personal care products were chosen as target pollutants to study their developmental toxicity and interactive effects on zebrafish embryo. The exposure period was 2-12 hours post fertilization (hpf). The BMDBM and PS-NPBs significantly upregulated the genes related to antioxidant enzymes and downregulated the gene expression of aromatase and DNA methyltransferases, but the influenced genes were not exactly the same, and the combined exposure reduced the adverse effects on the expression of all genes. With the help of single-cell RNA sequencing technology, the neural mid cells were identified as the target cells of both pollutants, and brain development, head development and notch signaling pathway were the functions they commonly altered. The key genes and functions that are specifically affected by BMDBM and/or PS-NPBs were identified. The BMDBM mainly affect the differentiation and fate of neurons in central nervous system through the regulation of her5, her6, her11, lfng, pax2a and fgfr4. The PS-NPBs regulated the expression of olig2, foxg1a, fzd8b, six3a, rx1, lhx2b, nkx2.1a and sfrp5 to alter nervous system development, retinal development, and stem cell differentiation. The phenotypic responses of zebrafish larvae at 120 hpf were tested, and significant inhibition of locomotor activity was found, indicating that early disrupting effect on central nervous system would have a sustained impact on the behavior of zebrafish.

Project description:BackgroundLight exposure regulates the human circadian system and more widely affects health, well-being, and performance. With the rise in field studies on light exposure's effects, the amount of data collected through wearable loggers and dosimeters has also grown. These data are more complex than stationary laboratory measurements. Determining sample sizes in field studies is challenging, as the literature shows a wide range of sample sizes (between 2 and 1,887 from a recent review of the field and approaching 105 participants in first studies using large-scale 'biobank' databases). Current decisions on sample size for light exposure data collection lack a specific basis rooted in power analysis. Therefore, there is a need for clear guidance on selecting sample sizes.MethodsHere, we introduce a novel procedure based on hierarchical bootstrapping for calculating statistical power and required sample size for wearable light and optical radiation logging data and derived summary metrics, taking into account the hierarchical data structure (mixed-effects model) through stepwise resampling. Alongside this method, we publish a dataset that serves as one possible basis to perform these calculations: one week of continuous data in winter and summer, respectively, for 13 early-day shift-work participants (collected in Dortmund, Germany; lat. 51.514° N, lon. 7.468° E).ResultsApplying our method on the dataset for twelve different summary metrics (luminous exposure, geometric mean, and standard deviation, timing/time above/below threshold, mean/midpoint of darkest/brightest hours, intradaily variability) with a target comparison across winter and summer, reveals required sample sizes ranging from as few as 3 to more than 50. About half of the metrics-those that focus on the bright time of day-showed sufficient power already with the smallest sample. In contrast, metrics centered around the dark time of the day and daily patterns required higher sample sizes: mean timing of light below mel EDI of 10 lux (5), intradaily variability (17), mean of darkest 5 hours (24), and mean timing of light above mel EDI of 250 lux (45). The geometric standard deviation and the midpoint of the darkest 5 hours lacked sufficient power within the tested sample size.ConclusionsOur novel method provides an effective technique for estimating sample size in light exposure studies. It is specific to the used light exposure or dosimetry metric and the effect size inherent in the light exposure data at the basis of the bootstrap. Notably, the method goes beyond typical implementations of bootstrapping to appropriately address the structure of the data. It can be applied to other datasets, enabling comparisons across scenarios beyond seasonal differences and activity patterns. With an ever-growing pool of data from the emerging literature, the utility of this method will increase and provide a solid statistical basis for the selection of sample sizes.

Project description:Titanium dioxide is a common additive in many food, personal care, and other consumer products used by people, which after use can enter the sewage system and, subsequently, enter the environment as treated effluent discharged to surface waters or biosolids applied to agricultural land, incinerated wastes, or landfill solids. This study quantifies the amount of titanium in common food products, derives estimates of human exposure to dietary (nano-) TiO(2), and discusses the impact of the nanoscale fraction of TiO(2) entering the environment. The foods with the highest content of TiO(2) included candies, sweets, and chewing gums. Among personal care products, toothpastes and select sunscreens contained 1% to >10% titanium by weight. While some other crèmes contained titanium, despite being colored white, most shampoos, deodorants, and shaving creams contained the lowest levels of titanium (<0.01 ?g/mg). For several high-consumption pharmaceuticals, the titanium content ranged from below the instrument detection limit (0.0001 ?g Ti/mg) to a high of 0.014 ?g Ti/mg. Electron microscopy and stability testing of food-grade TiO(2) (E171) suggests that approximately 36% of the particles are less than 100 nm in at least one dimension and that it readily disperses in water as fairly stable colloids. However, filtration of water solubilized consumer products and personal care products indicated that less than 5% of the titanium was able to pass through 0.45 or 0.7 ?m pores. Two white paints contained 110 ?g Ti/mg while three sealants (i.e., prime coat paint) contained less titanium (25 to 40 ?g Ti/mg). This research showed that, while many white-colored products contained titanium, it was not a prerequisite. Although several of these product classes contained low amounts of titanium, their widespread use and disposal down the drain and eventually to wastewater treatment plants (WWTPs) deserves attention. A Monte Carlo human exposure analysis to TiO(2) through foods identified children as having the highest exposures because TiO(2) content of sweets is higher than other food products and that a typical exposure for a US adult may be on the order of 1 mg Ti per kilogram body weight per day. Thus, because of the millions of tons of titanium-based white pigment used annually, testing should focus on food-grade TiO(2) (E171) rather than that adopted in many environmental health and safety tests (i.e., P25), which is used in much lower amounts in products less likely to enter the environment (e.g., catalyst supports, photocatalytic coatings).

Project description:Personal care products are increasingly being marketed to protect skin from the potentially harmful effects of air pollution. Here, we experimentally measure ozone deposition rates to skin and the generation rates and yields of oxidized products from bare skin and skin coated with various lotion formulations. Lotions reduced the ozone flux to the skin surface by 12% to 25%; this may be due to dilution of reactive skin lipids with inert lotion compounds or by reducing ozone diffusivity within the resulting mixture. The yields of volatile squalene oxidation products were 25% to 70% lower for a commercial sunscreen and for a base lotion with an added polymer or with antioxidants. Lower yields are likely due to competitive reactions of ozone with lotion ingredients including some ingredients that are not intended to be ozone sinks. The dynamics of the emissions of squalene ozonation product 6 methyl-2-heptenone (6MHO) suggest that lotions can dramatically reduce the solubility of products in the skin film. While some lotions appear to reduce the rate of oxidation of squalene by ozone, this evidence does not yet demonstrate that the lotions reduce the impact of air pollution on skin health.

Project description:The impact of pharmaceutical and personal care products (PPCPs) on the performance of biological wastewater treatment plants (WWTPs) has been widely studied using whole-community approaches. These contaminants affect the capacity of microbial communities to transform nutrients; however, most have neither honed their examination on the nitrifying communities directly nor considered the impact on individual populations. In this study, six PPCPs commonly found in WWTPs, including a stimulant (caffeine), an antimicrobial agent (triclosan), an insect repellent ingredient (N,N-diethyl-m-toluamide (DEET)) and antibiotics (ampicillin, colistin and ofloxacin), were selected to assess their short-term toxic effect on enriched nitrifying cultures: Nitrosomonas sp. and Nitrobacter sp. The results showed that triclosan exhibited the greatest inhibition on nitrification with EC50 of 89.1 μg L-1. From the selected antibiotics, colistin significantly affected the overall nitrification with the lowest EC50 of 1 mg L-1, and a more pronounced inhibitory effect on ammonia-oxidizing bacteria (AOB) compared to nitrite-oxidizing bacteria (NOB). The EC50 of ampicillin and ofloxacin was 23.7 and 12.7 mg L-1, respectively. Additionally, experimental data suggested that nitrifying bacteria were insensitive to the presence of caffeine. In the case of DEET, moderate inhibition of nitrification (<40%) was observed at 10 mg L-1. These findings contribute to the understanding of the response of nitrifying communities in presence of PPCPs, which play an essential role in biological nitrification in WWTPs. Knowing specific community responses helps develop mitigation measures to improve system resilience.

Project description:Background and objectivesColloid osmotic pressure (COP) is a principal determinant of intravascular fluid homeostasis and a pillar of fluid therapy and transfusion. Transfusion-associated circulatory overload (TACO) is a leading complication of transfusion, and COP could be responsible for recruiting additional fluid. Study objective was to measure COP of blood products as well as investigate the effects of product concentration and storage lesion on COP.Materials and methodsThree units of each product were sampled longitudinally. COP was measured directly as well as the determinants thereof albumin and total protein. Conventional blood products, that is red blood cell (RBC), fresh-frozen plasma (FFP) and platelet concentrates (PLTs), were compared with their concentrated counterparts: volume-reduced RBCs, hyperconcentrated PLTs, and fully and partially reconstituted lyophilized plasma (prLP). Fresh and maximally stored products were measured to determine changes in protein and COP. We calculated potential volume load (PVL) to estimate volume recruited using albumin's water binding per product.ResultsColloid osmotic pressure varies widely between conventional products (RBCs, 1·9; PLTs, 7·5; and FFP, 20·1 mmHg); however, all are hypooncotic compared with human plasma COP (25·4 mmHg). Storage lesion did not increase COP. Concentrating RBCs and PLTs did not increase COP; only prLP showed a supraphysiological COP of 47·3 mm Hg. The PVL of concentrated products was lower than conventional products.ConclusionColloid osmotic pressure of conventional products was low. Therefore, third-space fluid recruitment is an unlikely mechanism in TACO. Concentrated products had a lower calculated fluid load and may prevent TACO. Finally, storage did not significantly increase oncotic pressure of blood products.

Project description:The dataset provides information on Pharmaceutical and Personal Care Products (PPCPs) detected in the Albufera Natural Park (Valencia, Spain), a typical Mediterranean coastal wetland. These PPCPs constitute an important group of organic pollutants highly representative of the human impact. The concentrations values measured in soil, sediment and water and the statistical relationship of contaminants between them and with the environmental parameters could help to understand their fate in different compartments. The data also reported the occurrence and removal efficiency (%) for each contaminant in ten wastewater treatment plants (WWTPs), located in the surrounding area. This dataset could provide an idea on the effectiveness of WWTP treatments and the capacity of released PPCPs to affect the ecosystem. The extraction of analytes was based on solid-phase extraction (SPE) for water and solvent extraction followed by the previous SPE as clean-up for soil and sediment. Determination was carried out by high performance liquid chromatography tandem mass spectrometry (HPLC-MS/MS) with a triple-quadrupole. The present dataset was analyzed within the article entitled: "Pharmaceuticals and personal care products in a Mediterranean coastal wetland: Impact of anthropogenic and spatial factors and environmental risk assessment"[1].

Project description:In the last few decades, there has been a growing interest in understanding the behavior of personal care products (PCPs) in the aquatic environment. In this regard, the aim of this study is to estimate the accumulation and effects of four PCPs within the clam Ruditapes philippinarum. The PCPs selected were triclosan, OTNE, benzophenone-3, and octocrylene. A progressive uptake was observed and maximum concentrations in tissues were reached at the end of the exposure phase, up to levels of 0.68 µg g-1, 24 µg g-1, 0.81 µg g-1, and 1.52 µg g-1 for OTNE, BP-3, OC, and TCS, respectively. After the PCP post-exposure period, the removal percentages were higher than 65%. The estimated logarithm bioconcentration factor ranged from 3.34 to 2.93, in concordance with the lipophobicity of each substance. No lethal effects were found although significant changes were observed for ethoxyresorufin O-demethylase activity, glutathione S-transferase activity, lipid peroxidation, and DNA damage.

Project description:Titanium dioxide (TiO2) is a common component in personal care products (PCP), which through use enters the wastewater treatment plant (WWTP) and ultimately the environment. A citizen science approach is utilized here to inform the prevalence and usage of TiO2 containing PCP on a household scale, which generates information as to the quantity of TiO2 entering the WWTP, and the portion ultimately discharged to the environment. Meanwhile, citizen science sourced inventories were generated to estimate the quantity of TiO2, and potentially nanoscale TiO2 entering the WWTP from consumer products and to determine which products had the greatest contribution. The estimated values were compared with water samples from the WWTP which quantified the amount of total titanium present using ICP-AES. These values were at a similar level with other top-down estimation approaches and suggest that a citizen science approach is valid to estimate the loading of TiO2, and potentially other emerging contaminants, while at the same time engaging with community stakeholders.
